Hier bieden we een gedetailleerd protocol voor de bereiding van de muis hoofd en undecalcified bot weefsels, en cryosnijden voor immunokleuring van celproliferatie, celdood, en BMP-signalering markers. We geven ook een gedetailleerd beeld van de strategie voor het verkrijgen van kwantitatieve gegevens van immunofluorescentie beelden. Het gezicht is een belangrijk onderdeel van de menselijke identiteit, en bestaat uit verschillende soorten weefsels, zoals epitheel, spier, bot, kraakbeen, tand. Deze weefsels zijn afgeleid van alle drie de kiem lagen: Ectoderm, endoderm en mesoderm1,2. Voor een goede patronen en ontwikkeling van craniofaciale weefsels moeten celproliferatie, overlijden en differentiatie sterk worden gecoördineerd en gereguleerd door specifieke signalerings trajecten, zoals WNT-, FGF-, hh-en BMP-trajecten3,4 ,5. Defecten in proliferatie, overleving of differentiatie van cellen zullen leiden tot craniofaciale misvormingen, die behoren tot de vaakst voorkomende aangeboren geboorteafwijkingen. Transgene muizen zijn nuttige hulpmiddelen om mechanismen te bestuderen van craniofaciale morfogenese en pathogenese1,2,3,4,5. Het begrijpen van de veranderingen in craniofaciale structuren tijdens de ontwikkeling en pathogenese zal helpen om de belangrijkste ontwikkelingsprincipes te verduidelijken, evenals de mechanismen van craniofaciale misvormingen1,2,3 ,4,5.
De kleuring van hele Mount of gesectioneerde weefsels met specifieke antilichamen is een onschatbare techniek voor het bepalen van de ruimtelijke verdeling van eiwitten van belang 6. Formeel kan weefsel immunokleuring afhangen van immunohistochemie (IHC) of immunofluorescentie (IF). Vergeleken met het ondoorzichtige reactieproduct dat wordt gegenereerd met een chromogene substraat zoals 3, 3 '-Diaminobenzidine (DAB) door IHC, indien het gaat om het gebruik van fluorescerende conjugaten die zichtbaar zijn door fluorescentiemicroscopie. Daarom, als kan duidelijk onderscheid maken tussen positieve cellen van achtergrondruis, en laat afbeeldingen kwantitatief worden geanalyseerd en verbeterd op een eenvoudige manier door software zoals imagej en Adobe Photoshop7,8. De hele Mount kleuring aanpak werkt op kleine blokken weefsel (minder dan 5 mm dik), die driedimensionale informatie over de locatie van eiwitten/antigenen kan bieden zonder de noodzaak voor reconstructie uit de rubrieken9,10 . Echter, in vergelijking met weefsel secties, hele Mount immunokleuring is tijdrovend en vereist grote hoeveelheden antilichaam oplossingen. Niet alle antilichamen zijn compatibel met de basisbenadering voor hele montage. Bovendien zal de onvolledige penetratie van antilichamen resulteren in ongelijke kleuring of valse negatieve kleuring. Hier zullen we ons concentreren op de immunofluorescentie detectie van eiwitten/antigenen op verdeelde weefsels. Voor harde weefsels (bv, hoofd, tand, lang bot), calcium depositie tijdens ontwikkeling/pathogenese maakt het monster moeilijk om sectie en gemakkelijk afgespoeld tijdens immunokleurings behandeling11,12. De meeste van de momenteel beschikbare protocollen decalcify harde weefsels vóór inbedding te maken snijden gemakkelijker, dat is tijdrovend en kan de morfologie en antigenen van monsters vernietigen als onjuist behandeld11,12. Om de problemen te overwinnen, hebben we een aanpak geoptimaliseerd voor het cryosnijden van harde weefsels zonder decalcificatie, wat leidt tot een verbeterde visualisatie van hun morfologie en verdeling van signalering van eiwitten.
Het hier beschreven protocol wordt gebruikt om de Morfometrische en histologische veranderingen in de craniofaciale weefsels van BMP-transgene muizen te bepalen. In het bijzonder omvat het Protocol (1) het oogsten en ontleden van het hoofd weefsel, (2) sectie en immunokleuring van experimentele markers (Ki67, pSmad1/5/9) samen met TUNEL kleuring, (3) beeldvorming van de secties met behulp van fluorescentiemicroscoop, en ten slotte (4) analyseren en kwantificeren van de resultaten. Het protocol voor de voorbereiding en cryosectie harde weefsels zonder decalcificatie wordt ook beschreven13. Deze methoden zijn geoptimaliseerd voor craniofaciale weefsels. Ze zijn ook toepasbaar op andere weefsels van verschillende leeftijden van monsters met passende aanpassingen.

Weefsel immunokleuring biedt zeer specifieke en betrouwbare detectie van eiwitten van belang binnen een bepaald weefsel. Hier beschrijven we een volledig en eenvoudig protocol om eiwit expressie te detecteren tijdens craniofaciale morfogenese/pathogenese met behulp van muis craniofaciale weefsels als voorbeeld. Het protocol bestaat uit het voorbereiden en cryosnijden van weefsels, indirecte immunofluorescentie, beeld verwerving en kwantificering. Daarnaast wordt een methode beschreven voor de bereiding en het cryosnijden van onverdecalcificeerde harde weefsels voor immunokleuring, waarbij gebruik wordt gemaakt van craniofaciale weefsels en lange botten als voorbeeld. Deze methoden zijn essentieel om de eiwit expressie en morfologische/anatomische veranderingen in verschillende weefsels te bepalen tijdens craniofaciale morfogenese/pathogenese. Ze zijn ook toepasbaar op andere weefsels met passende modificaties. Kennis van de histologie en hoge kwaliteit van secties zijn van cruciaal belang om wetenschappelijke conclusies uit experimentele uitkomsten te trekken. Mogelijke beperkingen van deze methodologie omvatten maar zijn niet beperkt tot specificiteit van antilichamen en kwantificerings problemen, die hier ook worden besproken.

Hier presenteren we ons eenvoudige protocol voor immunofluorescentiedetectie op eiwitten op gedesnede materialen. Deze methode vereist geen antigeen ophalen. We zullen muizenkoppen gebruiken, inclusief onverkalkte harde weefsels.Het belangrijkste voordeel van deze techniek is het overslaan van antigeen ophalen en ontkalking van harde weefsels. Die leiden tot een goede kwaliteit van immunostaining resultaten. Deze methode bespaart ook zowel tijd als arbeid.Deze methode is ook van toepassing op andere weefsels met passende wijzigingen. Om mooie secties van onverkalkte weefsels te maken, ontleed je je doelweefsel verder om omliggende weefsels te trimmen en behandel indien nodig de monsters met 10%EDTA bij vier graden Celsius gedurende twee dagen voor cryoprotectie. Om embryo's van zwangere muizen te ontleden, pak de huid onder het midden van de buik van een geëuthanaseerde muis met tangen, snijd alleen door de huid en trek dan zachtjes naar de huid om het te scheiden van de onderliggende buikspierwand. Na dezelfde lijn van de huid incisie, gesneden in de buikholte.Verwijder de baarmoeder met een reeks embryo's. Verwijder vervolgens de embryo's door de baarmoederwand voorzichtig weg te snijden, waardoor de extraembryonische weefsels zoals de dooierzak en amnion worden verwijderd. Snijd en isoleer het hoofd van elk embryo, en met tangen breng elk hoofd in een aparte put van een 12-put plaat met 4%PFA.Incubeer op vier graden Celsius voor vier uur op te lossen. Spoel de hoofden in PBS op vier graden Celsius met zachte schudden voor 12 uur. Om de hoofden te cryoprotecteren, breng ze met behulp van tangen in een nieuwe 12-well plaat met twee milliliter van 30% sacharose in PBS.Incubeer met zachte agitatie bij vier graden Celsius tot de hoofden zinken naar de bodem van de schotel. Om de cryoprotected hoofden in te sluiten, breng ze over in een mal met optimale snijtemperatuur compound, en evenwicht gedurende enkele minuten. Met behulp van tangen, pas de locatie en richting van de monsters, zodat de bijgesneden kant van de monsters naar de onderkant van de inbeddingsvorm.Plaats vervolgens de mal op droog ijs te bevriezen, en op te slaan in een plastic zak op min 80 graden Celsius tot klaar voor cryosectioning. Voor postnataal weefsel, na het verwijderen van de huid en vetweefsel van een geëuthanaseerde, drie weken tot drie maanden oude muis, snijd en isolaat het hoofd en verwijder de onderkaak. Dan, fix en cryoprotect het hoofd zoals gedaan met embryo hoofden.Insluiten in 8%gelatine op een vergelijkbare manier als embryo hoofden in OCT, en houd de Cryomolds in een plastic zak op min 80 graden Celsius tot cryosectioning. Stel de juiste cryostattemperatuur in. Plaats de monsters in de cryostatkamer gedurende ongeveer 30 minuten om te equilibrate aan de cryostat temperatuur.Verwijder het blok met het monster uit de Cryomold, en monteer het met een OCT-druppel op de monster chuck en bevriezen, zorg ervoor dat de bijgesneden kant van het monster het verst van de chuck en tegenover de exploitant te houden. Laad de op het blok gemonteerde klem op de cryostat-objecthouder. Stel de bladhouder zo in dat de bladhoek drie tot vijf graden ten opzichte van het monster is.Verzamel 10-micrometer secties op gecoate microscoop dia's. Laat de secties op kamertemperatuur tot het helemaal droog is, en bewaar ze vervolgens op min 80 graden Celsius. Om te beginnen met de kleuring, neem dia's van min 80 graden Celsius, en houd ze op kamertemperatuur gedurende een uur aan de lucht drogen van de secties.Spoel de glijbanen in 0,1%PBST drie keer gedurende vijf minuten elk uit te wassen OCT en permeabiliseren de secties. Voeg 200 microliter blokkeeroplossing toe aan elke dia en broed gedurende 30 minuten bij kamertemperatuur uit. Verwijder vervolgens de blokkeringsoplossing zonder de dia te spoelen.Voeg 100 microliters primaire antilichamen toe die in blokkerende oplossing aan elke dia worden verdund, en broeden 's nachts bij vier graden Celsius uit. Spoel de glijbanen drie keer gedurende 10 minuten af met PBS bij kamertemperatuur. Voeg 100 microliter secundaire antilichamen verdund in blokkerende oplossing, en incubeer gedurende een uur bij kamertemperatuur, beschermd tegen licht.Spoel in PBS drie keer gedurende 10 minuten elk op kamertemperatuur, nog steeds beschermd tegen licht. Als u de dia's wilt monteren, voegt u twee druppels anti-fade-medium toe met DAPI op elke dia, dekt u af met een coverlip en slaat u op vier graden Celsius op tot het beeldklaar is. Frontale botten van controle embryo's waren positief voor pSmad1/5/9, downstream BMP signalering factoren, en Ki67, een cel proliferatie marker.In neurale kuif-specifieke, constitutief geactiveerde BMPR1A mutant embryo's, echter, pSmad1/5/9 niveaus werden verhoogd, terwijl Ki67 niveaus werden verlaagd. Bovendien werden meer apoptotische cellen waargenomen in de frontale botten van gemuteerde embryo's dan die van controleembryo's. Coronale cryosections van gelatine-ingebedde hoofden tonen duidelijk GFP signaal en RFP signaal van een tdTomato cassette in de neusbot en neusweefsels, waaruit blijkt dat gelatine niet interfereert met fluorescerende signalen.Toen deze secties werden immunostained voor SOX9 of Osterix en E11/Podoplanin, werden secties van goede kwaliteit verkregen uit de meeste van de drie weken harde weefsels. Aan de andere kant, met drie maanden oude monsters, werden secties van goede kwaliteit alleen verkregen in sommige van de harde weefsels, waaronder de trabeculaire compartimenten van het dijbeen, neusweefsel en de schedel, met inbegrip van de nasale-premaxilla hechting en de omliggende botten van het hoofd. SOX9-positieve cellen werden specifiek gedetecteerd in de chondrocyten van de groeiplaat en het gewricht van het dijbeen en het neusseptum.In de drie weken oude snijtand werd SOX9 ontdekt in de mesenchymale cellen. Osterix en E11 dubbele kleuring resultaten tonen aan dat Osterix werd gedetecteerd in osteoblasten, terwijl E11 werd ontdekt in osteocyten van botten van het dijbeen en het hoofd. In de drie weken durende snijtand was Osterix positief in odontoblasten, terwijl E11 positief was in follikel mesenchymale cellen.Gelieve geen monsters met 4% PFA te overfixen. Voor het doorsnee van onverkalkte harde weefsels in gelatine is de beste temperatuur min 25 graden Celsius en lager. Na deze procedure kunnen fluorescentieniveaus worden gekwantificeerd zoals beschreven in ons protocol.Deze metingen zullen informatieve gegevens opleveren om het verschil van het relatieve eiwitgehalte tussen verschillende groepen aan te tonen. Na de ontwikkeling maakte deze techniek de weg vrij voor dubbele of drievoudige kleuring om co-expressiepatronen van verschillende eiwitten te krijgen. 4%PFA is gevaarlijk.Het kan leiden tot huidallergieën, ernstige oogschade, orgaanschade, of kanker. Gebruik persoonlijke beschermingsmiddelen en was de huid grondig na behandeling.
