特定の抗体を用いたマウント全体または断面組織の染色は、目的のタンパク質6の空間分布を決定するための非常に貴重な技術である。正式には、組織免疫染色は、免疫組織化学(IHC)または免疫蛍光(IF)のいずれかに依存することができます。IHCによる3,3'-Diaminobenziジン(DAB)などの発色基板で生成された不透明反応生成物と比較して、IFは蛍光顕微鏡で見える蛍光コンジュゲートの使用を含む。したがって、IFは明らかにポジティブな細胞をバックグラウンドノイズから区別し、ImageJやAdobe Photoshop 7、8などのソフトウェアによって画像を定量的に分析し、強化することができる。全体のマウント染色アプローチは、セクション9、10からの再建を必要とせずにタンパク質/抗原の位置に関する3次元情報を提供することができ、組織の小さなブロック(厚さ5ミリメートル未満)で動作します。.しかし、組織切片と比較して、マウント免疫染色全体は時間がかかり、大量の抗体溶液を必要とします。すべての抗体が基本的なマウントアプローチと互換性があるわけではありません。さらに、抗体の不完全な浸透は、不均一な染色または偽陰性染色をもたらす。ここでは、切除組織上のタンパク質/抗原の免疫蛍光検出に焦点を当てます。硬い組織(例えば、頭部、歯、長骨)の場合、発症/病因の間のカルシウム沈着は、サンプルを切除しにくくし、免疫染色治療中に容易にすすいでくるぐる。現在利用可能なプロトコルのほとんどは、セクション化を容易にするために埋め込む前に硬い組織をデカール化し、これは時間がかかり、不適切に扱われた場合、サンプルの形態および抗原を破壊する可能性があります11,12.この問題を克服するために、脱石せずに硬質組織を凍結切除するアプローチを最適化し、その形態の可視化とシグナル伝達タンパク質の分布を改善しました。

ここでは、切片切り材料上のタンパク質に対する蛍光免疫検出のための簡単なプロトコルを紹介します。この方法は、抗原の検索を必要としません。難組織を含むマウスヘッドを使用します。この技術の主な利点は、硬い組織の抗原の検索および脱灰をスキップすることです。それらは免疫染色結果の良質につながる。この方法はまた時間および労働両方を節約する。この方法は、適切な修正を伴う他の組織にも適用可能である。解剖されていない組織から素敵な切片を作るために、ターゲット組織をさらに解剖して周囲の組織をトリミングし、必要に応じて凍結保護の前に2日間摂氏4度で10%EDTAでサンプルを処理します。この方法の手順について、視覚的なデモンストレーションで詳細かつ明確な説明を提供します。まず、PBSを含む10センチメートルといくつかの3.5センチメートルの皿と、各妊娠中のマウスの各井戸にPBSに4%PFAの2ミリリットルを含む1つの12ウェル培養プレートを準備し、それらをすべて氷の上に置きます。妊娠中のマウスから胚を解剖するには、安楽死させたマウスの腹の中心下の皮膚を鉗子でつかみ、皮膚のみを切断し、皮膚を穏やかに引っ張って下の腹筋壁から分離する。皮膚切開の同じラインに続いて、腹腔内に切り込む。胚の文字列を含む子宮を取り除きます。次に、子宮壁を優しく切断して胚を取り除き、黄身嚢や羊膜などの胚外組織を除去する。各胚から頭部を切断して分離し、鉗子で各頭部を4%PFAを含む12ウェルプレートの別々のウェルに移す。摂氏4度で4時間インキュベートして固定します。12時間穏やかな揺れで4°CでPBSで頭をすすいでください。ヘッドを凍結保護するには、鉗子を使用して、PBSで30%スクロースの2ミリリットルを含む新しい12ウェルプレートに移します。頭が皿の底に沈むまで摂氏4度で穏やかな攪拌でインキュベートします。凍結保護ヘッドを埋め込むには、最適な切断温度化合物を含む金型に移し、数分間平衡化します。鉗子を使用して、サンプルの位置と方向を調整して、サンプルのトリム側が埋め込み金型の底面に向かるようにします。その後、乾燥氷の上にカビを置いて凍結し、凍結切断の準備ができるまでマイナス80度のビニール袋に入れて保管します。出生後組織の場合、安楽死させた3週間から3ヶ月のマウスの皮膚および脂肪組織を除去した後、頭部を切断して分離し、下顎を取り除く。その後、胚の頭部で行われたように頭を固定し、凍結します。OCTの胚頭部と同様の方法で8%ゼラチンに埋め込み、クライオムドルを下切りまでマイナス80度のビニール袋に入れます。適切なクライオスタット温度を設定します。クライオスタットチャンバーにサンプルを約30分間置き、クライオスタット温度に平衡させます。クリオマールからサンプルを取り外し、OCTドロップで試料チャックに取り付けて凍結し、サンプルのトリミングされた側をチャックから最も遠く離れ、オペレータに向かうことを確認します。ブロックマウントチャックをクライオスタットオブジェクトホルダーに積み込みます。ブレードの角度がサンプルに対して 3 ~ 5 度になるように、ブレード ホルダーを調整します。コーティングされた顕微鏡のスライドに10マイクロメートルのセクションを集める。完全に乾燥するまで室温でセクションを残し、マイナス80度で保存します。染色から始めるには、マイナス80度からスライドを取り出し、室温で1時間保ち、セクションを空気乾燥させます。0.1%PBSTでスライドを3回5分間洗い流し、OCTを洗い流し、セクションを透過させます。各スライドに200マイクロリットルのブロッキング溶液を加え、室温で30分間インキュベートします。次に、スライドをすすかずにブロッキング溶液を取り除きます。各スライドにブロッキング溶液で希釈した一次抗体の100マイクロリットルを加え、摂氏4度で一晩インキュベートする。スライドを室温で10分間3回、PBSで洗い上がってください。ブロッキング溶液に希釈した2次抗体の100マイクロリットルを加え、光から保護された室温で1時間インキュベートする。PBSで室温でそれぞれ10分間3回リンスし、依然として光から保護する。スライドを取り付けるには、各スライドにDAPIを使用して2滴のアンチフェードメディアを追加し、カバースリップで覆い、イメージを作成できるまで摂氏4度で保存します。コントロール胚の前頭骨は、pSmad1/5/9、下流BMPシグナル伝達因子、および細胞増殖マーカーであるKi67に陽性であった。神経堤特異的な、構成的に活性化されたBMPR1A変異胚では、しかしながら、ki67レベルが減少している間にpSmad1/5/9レベルが増加した。さらに、対照性胚の細胞よりも変異胚の前頭骨においてより多くのアポトーシス細胞が観察された。ゼラチン埋め込まれた頭部の冠状動脈は、鼻骨および鼻組織のtdTomatoカセットからのGFP信号およびRFP信号を明確に示し、ゼラチンが蛍光シグナルを妨げないことを示す。これらの切片がSOX9またはオステルクスおよびE11/ポドプラニンに対して免疫染色された場合、3週間の硬い組織のほとんどから良質の切片が得られた。一方、3ヶ月前のサンプルでは、大腿骨、鼻組織、頭蓋骨の骨膜部分(鼻前顎縫合糸、頭部の周囲の骨を含む)を含む硬い組織の一部でのみ良質の切片が得られた。SOX9陽性細胞は、大腿骨および鼻中隔からの成長板および関節の軟骨細胞において特異的に検出された。生後3週間の切歯では、間葉細胞でSOX9が検出された。オステリクスとE11の二重染色結果は、オステリクスが骨芽細胞で検出され、E11は大腿骨および頭部からの骨の骨細胞で検出されたことを示す。3週間の切歯では、オステリクスは歯根芽細胞で陽性であり、E11は卵胞間葉細胞で陽性であった。4%PFAでサンプルをオーバーフィックスしないでください。ゼラチン中の固硬組織の除菌では、最高温度はマイナス25°C以下です。この手順に従って、蛍光レベルは、我々のプロトコルに記載されているように定量化することができる。これらの測定値は、異なるグループ間のタンパク質の相対的なレベルの違いを示す有益なデータを提供します。その開発後、この技術は、異なるタンパク質の共発現パターンを得るための二重または三重染色のための道を開いた。4%PFA は危険です。皮膚アレルギー、重篤な眼の損傷、臓器の損傷、または癌を引き起こす可能性があります。取扱い後は、個人用保護具を使用し、肌を十分に洗ってください。
