私たちの技術の主な利点は、ベッドサイドに深度電極を埋め込んだ患者のケース位置、瞳孔サイズ、および単一ニューロンの同時記録を可能にすることです。これにより、人間の脳に、その活動が目の動きに敏感で、現在固定されている刺激のアイデンティティに敏感なニューロンがあるかどうかを判断することができます。この技術は特定の疾患を研究するようには設計されていませんが、異常な眼球運動やその他の眼運動異常をもたらす神経疾患の研究に適しています。この手順を実証するには、私の研究室の研究員であるナンド・チャンドラバディアとジェームズ・リーです。そして、シダーズ・シナイの神経診断技術者エリカ・クアン。まず、電極を記録システムに接続し、患者を直立して座ります。次に、刺激コンピュータを電気生理システムとアイトラッカーに接続します。リモート非侵襲赤外線アイトラッキングシステムを堅牢なモバイルカートに設置します。次に、LCDディスプレイを保持するフレキシブルアームをカートに取り付けます。アイトラッキングカートに完全に充電された無停電電源装置を設置し、アイトラッキングに関連するすべてのデバイスを外部電源ではなく電源に接続します。患者に接続されたIVデバイスがバッテリで動作しており、壁に差し込まれていないことを確認します。アイトラッキングソフトウェアを起動します。患者と液晶画面の間の距離を60〜70センチメートルに調整します。また、画面の表面が患者の顔とほぼ平行になるように、LCDスクリーンの角度を調整します。眼トラッカーが頭の動きに合わせて調整できるように、患者の額にステッカーを貼ります。今度は、眼トラッカーのカメラが患者の鼻の高さ程度になるように、患者の頭部に対して画面の高さを調節する。患者にボタンボックスまたはキーボードを提供します。実験を開始する前に、トリガとボタンの押下が正しく記録されていることを確認します。取得ソフトウェアを起動します。まず、ブロードバンドローカルフィールドの電位を視覚的に調べ、回線ノイズによって汚染されていないか確認します。それ以外の場合は、標準の手順に従ってノイズを除去してください。単一ニューロンを識別するために、バンドパスは300ヘルツから8キロヘルツまでの信号をフィルタリングします。各マイクロワイヤバンドルのリファレンスとして、8 本のマイクロワイヤのいずれかを選択します。記録する前に、データを nrd ファイルとして保存できます。ターゲットマーカー、ヘッド距離、瞳孔、角膜反射が準備完了としてマークされるように、アイトラッカーと患者の間の距離と角度を調整します。これは、アイトラッキングソフトウェアで緑色で示されています。記録する目をクリックし、サンプリングレートを500ヘルツに設定します。瞳孔と角膜反射のしきい値の自動調整を使用します。各ブロックの先頭に組み込みのグリッド方式でアイトラッカーを調整します。目の位置がグリッドとしてうまく登録されたことを確認します。それ以外の場合は、キャリブレーションをやり直します。キャリブレーションを受け入れ、検証を行います。最大検証誤差が 2 度未満で、平均検証誤差が 1 度未満の場合は、検証を受け入れます。それ以外の場合は、検証をやり直します。次に、ドリフト補正を行い、実際の実験に進みます。このビジュアル検索タスクでは、このグループで行われた以前のスタディの刺激を使用し、前に説明したタスク手順に従います。参加者にタスクの指示を提供します。検索配列内のターゲット項目を見つけ、できるだけ早く応答するように参加者に指示します。ターゲットが見つからない場合は、応答ボックスの左ボタンを押し、ターゲットが存在しないと思う場合は右ボタンを押します。参加者に対して、ターゲット存在試験とターゲット不在試験があることを明示的に指示します。刺激プレゼンテーションソフトウェアを起動し、タスクを実行します。1秒間の目標キューを提示し、刺激表示ソフトウェアを使用して検索配列を提示します。ボタンを押して記録し、試用版のフィードバックを参加者に提供します。この方法の使用法を説明するために、患者が視覚探索タスクを実行している間にヒト内側側頭葉から228個の単一ニューロンが記録された。このタスクの間に、ニューロンの活性が標的と散気器の固定を区別するかどうかを調査した。応答がボタンプレスで整列すると、ターゲット-存在試験とターゲット不在試験の間に差異活性を示すニューロンが見つかりました。黒い線は検索キューの発症とオフセットを表します。このニューロンは、ターゲット存在試験の発射速度を増加させるが、標的不在試験では増加しなかった。逆に、このニューロンは標的存在試験の発射率を低下させるが、標的不在試験では減少しなかった。内側側頭葉ニューロンのサブセットは、標的の固定と散気者の間で有意に異なる活動を示した。さらに、そのような標的ニューロンの1つのタイプは、気晴らしに対するターゲットに対するより大きな応答を有し、他方は標的に対する気晴らしに対するより大きな応答を有した。この結果は、内側側頭葉ニューロンのサブセットが、現在の固定が標的に着陸したかどうかをコード化することを示している。アイトラッカーが録音にノイズを入れないようにすることが重要です。実験を行う前に、ノイズを除去する必要があります。ノイズが続く場合は、被験者に刺激を表示するために使用される画面に外部電源があることを確認します。ハイエンドのオーディオアプリケーションに通常使用される高品質の電源用の交換電源。この手順に従って、スパイクと眼球運動の分析が行われ、研究者が眼球運動に対する神経応答を調査することを可能にする情報をデータから抽出する。この方法は、ニューロンがシーン内の興味深いオブジェクトに対して固定をまとめて駆動する方法など、固定の分析を必要とする多くの未回答の質問を探索する方法を開きます。
