Bruk av eksisterende hel plante herbivory metoder er unødvendig å oppnå målet med denne spesielle studien (dvs. skjermen et sett av kandidat gener for deres reaksjon på infestation). Herbivory setter i gang en rekke molekylære hendelser der en plante kan både identifisere angrep og montere en passende respons for sin overlevelse. En plante får to grunnleggende signaler fra tygge insekter; en fra den fysiske skaden på vevet og den andre fra insekt-spesifikke stoffer. Skade-Associated molekylære mønstre (demper) er utgitt som svar på skader opprettet av larvestadiet elektiske og utløse en veldefinert såret respons som resulterer i en økning i hormonet jasmonic syre og transkripsjon av Forsvaret gener1. En av de mest kjente demper er anlegg, en polypeptid som er dannet av kløften av større prosystemin protein etter et blad er såret2,3. Den jasmonic syre såret responsen er ytterligere modulert av herbivore-assosiert molekylær mønstre (HAMPs), som kan utledes fra Caterpillar spytt, gut innhold (regurgitant) og avføring (frass)4. Insekter bruker disse stoffene til enten å øke eller unngå forsvars responsen5. Transkripsjon faktorer deretter relé meldingen fra hormon signaler i forsvaret responsen via regulering av nedstrøms forsvaret gener6,7,8.
Noen plante-insekt interaksjon studier brukes i laboratoriet innstillingene er av simulert type, med et mål om tilnærme den naturlige metoden for fôring av insekt. Simulert herbivory oppnås vanligvis ved å skape kunstige skader å plante vev med ulike verktøy som etterligner den spesifikke mekanismen for insekt elektiske tilstrekkelig til å forårsake frigjøring av demper og utløse produksjon av forsvars gener. Solanum tuberosum var. Kennebec spres ved Nodal overføring i et enkelt vev kultur medium og overføres til jord for videre vekst hvis ønskelig.
