このプロトコルは、特別な装置を使用せずに3D細胞培養モデルの信頼性を高める簡単な方法を使用します。この技術の主な利点は、立方体装置内の初期培養条件を制御することによって、再現可能な3D培養実験を行うことができる点である。5ミリメートルポリカーボネート立方体フレームを準備した後、事前に冷却されたスライドとアイスボックスにフレームを置き、ピペットを使用して、立方体フレームの上面から下面に予熱された1.5%アガロースの12マイクロリットルを追加します。アガロースを広げて平らな表面を得て、ポリマーを治癒させます。ピンセットを使用してフレームをガラスの端までスライドさせ、開いた側がスライドに戻す前にフレームを下向きにするようにフレームを回転させます。フレームの次の2つの表面をより多くのアガロースで満たした後、ちょうど示したように、アガロースを開いた面から容器に落とし、第4および第5の側面にアガロース壁を形成する。初期細胞クラスター形状を設定するには、目的とする細胞培養用の適切な細胞外マトリックスをハイブリッドゲルキューブ培養空間に注入し、作製したマイクロモールドをキューブにセットする。その後、立方体を摂氏37度で25分間二酸化炭素インキュベーターに入れます。細胞外マトリックスが硬化したら、マトリックスが劣化しないように慎重にマイクロモールドを持ち上げます。所望の型状のポケットは、細胞外マトリックスで製造される。細胞を濃縮するには、適切な実験細胞培養培地で遠心分離後の実験細胞懸濁液を濃縮し、細胞外マトリックス内のポケットに細胞を注入する。立方体を37°Cで20分間二酸化炭素インキュベーターに入れ、細胞が細胞外マトリックスポケットに落ちるようにし、マイクロモールドによって作成されたスペースを満たします。インキュベーションの終わりに、24ウェルプレートの1つのウェルに立方体を置き、適切な細胞培養培地の100マイクロリットルをウェルに加えます。追加の細胞外マトリックスを注入してポケットを閉じ、キューブをインキュベーターに25分間戻します。細胞外マトリックスが硬化すると、20°Cのアガロースの約10マイクロリットルを立方体に上面に落とし、表面を閉じ、37°Cで10〜20分間滴を硬化させた。その後、キューブ内の細胞への栄養の移動を促進するための浸透圧を促進するために、新鮮な媒体で立方体全体をカバーします。多方向観察による非侵襲的な3D形状認識のために、プレートを顕微鏡ステージに配置し、明視野または位相コントラスト顕微鏡で立方体の各辺の画像を取得し、ピンセットを使用して、キャプチャ間で画像化されるキューブサービスを回転させます。多方向観察による免疫蛍光イメージングの場合、まずキューブを含むウェルから上澄み物を吸引し、室温で20分間4%パラホルムアルデヒドで立方体を固定する。固定の終わりに、1回の洗浄につき10分間PBSでキューブを2回洗います。0.5%トリトンx-100で立方体を非浸透させ、摂氏4度で10分間PBSを行います。次に、室温で60分間、ヤギ血清および免疫蛍光バッファーとの非特異的結合を遮断する前に、1回の洗浄につき10分間、PBSで立方体を3回洗浄します。次に、標準的な抗体染色プロトコルに従って目的の適切な一次抗体で細胞を染色します。次に、ちょうど実証したように、レーザーまたは蛍光顕微鏡上の立方体の6つの側面すべてを画像化します。この代表的な実験では、ハイブリッドゲルキューブの多方向イメージングにおいて、通常のヒト気管支上皮細胞で培養し、培養物の位相コントラストおよび免疫蛍光イメージングによる初期円筒形またはプリズム形細胞培養の生成を示す。ここで、ハイブリッドゲルキューブにおいて、まずは通常のヒト上皮細胞の免疫染色が円筒形に制御され、気管支樹の生成後に示される。多次元イメージングによって明らかにされるように、枝は円筒軸に対して垂直を示す。細胞の密度が低いとインキュベーション後にその細胞の品質が低下する可能性があるため、細胞外マトリックスポケットに高密度の細胞を注入することが重要です。この方法パターンは、多方向イメージングによる定量的測定による3D細胞パターンの再現性形成を行い、皿の発達のメカニズムを研究する。
