3d analysis of multi-cellular responses to chemoattractant gradients

3D Analysis of Multi-cellular Responses to Chemoattractant Gradients

単純な2Dインビトロ培養と3D組織のような環境の違いは、生きている組織の空間的および化学的複雑さを表す3Dシステムへの関心を高めています。製造プロセスは、施設やフォトリソグラフィ技術を必要としません。しかし、3D PDMS装置は、3D生理学的環境用途に必要なベクトルを含む。中流体デバイスの準備のために、適切な三次元コンピュータ支援設計ソフトウェアプログラムを使用して、ポリジメチルシロキサンまたはPDMSデバイス用の金型のマスクを設計し、耐熱性樹脂を備えたステレオリソグラフィー装置を使用して金型を印刷します。金型の準備ができたら、10対1の比率で硬化剤を用いて金型あたり3ミリリットルのPDMSモノマー溶液を約混合し、真空乾燥剤で混合物を1時間脱気します。乾燥の終わりには、粘着テープを使用して金型表面からほこりを取り除き、鋳型を脱気PDMS溶液で丁寧に充填します。次に、PDMSを摂氏80度で2時間硬化させてから、金型を室温で冷却します。金型をタッチに冷却する場合は、ブレードを使用して金型とPDMSの境界を切断し、PDMSを金型から丁寧に剥離します。PDMSを22 x 22ミリのカバースリップに収めるためにトリミングし、入口に穴を開けます。低い糸くず組織と70%エタノールを使用して、デバイスを拭き取り、カバースリップし、大きなほこり粒子を除去するために新しい粘着テープでデバイスをダブ。その後、PDMSデバイスをオートクレーブし、8分間濡れ、15分間乾燥して121°Cでカバースリップし、PDMS部分の底面を治療し、コロナ放電ガンで覆い物を組織培養フードで5分間処理してから、ピースを一緒に接着します。デバイスをロードするには、まず1%ペニシリンとストレプトマイシンと1%インスリントランスフェリンセレン-xを補充増殖培地の適切な量で目的の細胞を再中断します。次に、乳腺細胞懸濁液の50マイクロリットルを425マイクロリットルの作製した中和コラーゲン溶液と混合し、200マイクロリットルのピペットにコラーゲン細胞懸濁液を充填する。チャンバー全体が満杯になるまでPDMSデバイスに懸濁液を注入し、デバイスを摂氏37度で5%の二酸化炭素で1時間インキュベーターに入れ、両側のリザーバーに成長培地を充填し、共焦点イメージングまでインキュベーターに戻します。3Dイメージングと定量化の場合は、共焦点顕微鏡に生きた細胞イメージング培養チャンバーを設置し、温度と二酸化炭素をそれぞれ摂氏37度と5%に事前に設定します。イメージングチャンバーに水を加え、チャンバーに湿った拭き取りを加え、必要に応じてチャンバーに入れ、PDMSデバイスをチャンバーに入れます。傾斜形成の要因の源として、一方の貯水池に表皮成長因子を加え、他の貯水池を残して、空間的に等級表皮成長因子分布の開発のためのシンクとして機能する。次に、目的のオルガノイドの体積をカバーするZスタックの範囲を設定し、イメージングを開始します。実験の最後に、適切な画像解析プログラムを使用して、オルガノイドから伸びる枝の長さと角度、または個々の細胞の移動を測定します。インシリコとインビトロの両方のテストでは、ソースまたはシンクリザーバの補充なしに約2日間持続する細胞培養領域全体に安定した線形上皮成長因子勾配の形成が明らかになっており、表皮成長因子である6.4キロダルトンタンパク質は、比較的短期間に実証されたコラーゲンゲル内に安定な拡散ベースの勾配を形成できることを示唆した。乳腺オルガノイドは、1ミリリットル当たり0.5ナノモルを有する線形勾配に表皮成長因子を添加した場合、3日間にわたって方向性バイアスのない空間的に均一な2.5ナノモル表皮成長因子の存在下で複数の枝を形成する。ただし、分岐形成は有意な方向バイアスを表示します。注意すべきは、ギャップ接合抑制剤の添加は、オルガノイドが勾配に応答する能力を抑制する。コラーゲン溶液のpHは、ゲル化および細胞生存率にとって重要である。混合する前にコラーゲンを中和しないと、細胞の生存率が低くなります。この方法は、より現実的な組織モデリングに対応するために拡張することができ、ゲル内の個々の細胞からの血管ネットワーク形成に対応することができます。
