Hoewel recente studies calcium beeldvorming van DRG-neuronen in vivo16hebben bereikt, blijft het uitvoeren van in vivo Patch-clamp-opnames van individuele DRG-nociceptoren uiterst uitdagend. Daarom is een in vivo single-Fiber aanpak voor het pijn veld van blijvende betekenis. Single-Fiber opname in dit protocol laat objectieve observatie van geleidings falen verschijnselen toe, en de combinatie van deze techniek met het ex vivo preparaat dat in de huidige studie is ontwikkeld, maakt het onderzoek ...

De normale overdracht van informatie langs zenuwvezels garandeert de normale werking van het zenuwstelsel. Abnormale werking van het zenuwstelsel wordt ook weerspiegeld in de elektrische signaaloverdracht van zenuwvezels. Zo kan de mate van demyelinisatie bij centrale demyelineaflaesies worden geclassificeerd via vergelijking van veranderingen in zenuw geleidings snelheid voor en na interventie toepassing1. Het is moeilijk om intracellulair record zenuwvezels, behalve in speciale preparaten zoals de inktvis reus axon2. Daarom is elektrofysiologische activiteit alleen beschrijfbare via de extracellulaire opname van enkelvoudige vezels. Als een van de klassieke elektrofysiologische methoden, heeft single-Fiber opname een langere geschiedenis dan andere technieken. Echter, minder elektrofysiologen grijpen deze methode ondanks de uitgebreide toepassing. Daarom is een gedetailleerde introductie van het standaardprotocol voor de opname van één vezel nodig voor de juiste toepassing.
Hoewel verschillende Patch-clamp technieken moderne elektrofysiologische studie hebben gedomineerd, speelt single-Fiber opname nog steeds een onvervangbaar rol bij het opnemen van de activiteiten van zenuwvezels, vooral vezels die perifeer gevoel overbrengen met hun sensorische cel lichaam gelegen in dorsale wortel ganglion (DRG). Het voordeel van het gebruik van single-Fiber opname hier is dat in vivo Fiber Recording een lange observatietijd biedt met de capaciteit om reacties op natuurlijke stimuli op te nemen in preklinische modellen zonder verstoring van de intracellulaire omgeving3 , 4.
Een toenemend aantal studies in de afgelopen twee decennia heeft onderzocht complexe functies langs zenuwvezels5, en geleiding falen, die wordt gedefinieerd als een staat van mislukte zenuw impuls transmissie langs de axon, was aanwezig in veel verschillende perifere zenuwen6,7. De aanwezigheid van geleidings falen in ons onderzoek diende als een intrinsiek Zelfremmend mechanisme voor de modulatie van aanhoudende Nociceptieve input langs C-vezels8. Dit geleidings falen was significant verzwakt onder de voorwaarden van hyperalgesia4,9. Daarom, gericht op de factoren die betrokken zijn bij conductie falen kan een nieuwe behandeling voor neuropathische pijn vertegenwoordigen. Om het geleidings falen te observeren, moet het afvuren patroon worden geregistreerd en geanalyseerd op basis van opeenvolgend geloosde spikes op basis van single-Fiber opname.
Om het mechanisme van geleidings falen grondig te begrijpen, is het noodzakelijk om de transmissie eigenschappen van de axon te identificeren, of preciezer, de membraan eigenschappen van DRG-neuronen, gebaseerd op hun pseudo-unipolaire anatomische eigenschappen. Veel eerdere studies op dit gebied zijn uitgevoerd op gezien DRG neuronen10,11, die mogelijk niet haalbaar voor het onderzoek van de geleiding falen als gevolg van twee obstakels. Eerste, verschillende mechanische en chemische methoden worden gebruikt in het dissociatie proces te vrije DRG neuronen, die kan resulteren in ongezonde cellen of verander het fenotype/eigenschappen van de neuronen en Verwar de bevindingen. Ten tweede, de bijgevoegde perifere zenuwen zijn in principe verwijderd, en geleiding falen verschijnselen zijn niet waarneembaar in deze preparaten. Daarom is een preparaat van intact DRG neuronen met een bijgevoegde zenuw verbeterd om te voorkomen dat de bovengenoemde obstakels.

use of in vivo single-fiber recording and intact dorsal root ganglion with attached sciatic nerve to examine the mechanism of conduction failure

Single-Fiber opname is de laatste decennia een klassieke en effectieve elektrofysiologische techniek geweest vanwege de specifieke toepassing voor zenuwvezels in de centrale en perifere zenuwstelsels. Deze methode is met name van toepassing op dorsale wortel ganglia (DRG), die primaire sensorische neuronen zijn die een pseudo-unipolaire structuur van zenuw processen vertonen. De patronen en kenmerken van de actie potentialen doorgegeven langs axonen zijn beschrijfbare in deze neuronen. De huidige studie gebruikt in vivo single-Fiber opnames om de geleiding falen van de sciatische zenuwen in volledige Freund van adjuvans (CFA) behandelde ratten te observeren. Omdat het onderliggende mechanisme niet kan worden bestudeerd met behulp van in vivo single-Fiber opnames, worden Patch-clamp-opnames van DRG neuronen uitgevoerd op preparaten van intact DRG met de bijgevoegde sciatische zenuw. Deze opnames onthullen een positieve correlatie tussen geleidings falen en de stijgende helling van de nahyperpolarisatie potentiaal (AHP) van DRG-neuronen bij met CFA behandelde dieren. Het protocol voor in vivo enkelvoudige glasvezel opnames maakt de classificatie van zenuwvezels mogelijk via de meting van de geleidings snelheid en het monitoren van abnormale omstandigheden in zenuwvezels bij bepaalde ziekten. Intact DRG met bijgevoegde perifere zenuw maakt observatie van de activiteit van DRG neuronen in de meeste fysiologische omstandigheden. Overtuigend, single-Fiber opname gecombineerd met elektrofysiologische opname van intacte Drg's is een effectieve methode om de rol van geleidings falen tijdens het analgetische proces te onderzoeken.

De uitkomst van het single-Fiber opname protocol is afhankelijk van de kwaliteit van de vezel dissectie. Het dier voor in vivo experimenten moet in een goede situatie verkeren om de zenuw stam gezond te houden voor gemakkelijk dissectie (zie advies in de sectie discussie). Een drug toepassing bad is nodig in veel gevallen voor de levering van geneesmiddelen op vezels. Single-fiber Recording speelt een belangrijke rol bij het registreren van de activiteiten van zenuwvezels. Vooral activiteiten die perifeer gevoel overbrengen van ontvankelijke velden naar neuronen in dorsale wortel ganglion. In golf van vezel opname, biedt de lengte van de duur tijd met de capaciteit om reacties op de natuur stimuli record, en was later verstoring van de intercellulaire omgeving.In een enkele vezel opname vereist neutron automatisch goed voor de situatie. De therapeut roept de vereiste voor de voorbereiding van geïntegreerde TESTS DRG de heupzenuw geïntegreerd is. Het resultaat van demo situatie toont bijna alle details van zenuwen in goede, fysieke sticker stadium.En ten derde, hele DRG voorbereiding zoals zijn geïntegreerd. Om deze procedure te beginnen, bereiden en desinfecteren alle chirurgische instrumenten voorafgaand aan de operatie. Bereid vervolgens een tot twee liter van de extracellulaire oplossing van De Ringer en bewaar op vier graden Celsius tot gebruik.Om de heupzenuw romp bloot voor het opnemen, eerst, opengesneden de verdoofde rat huid en spieren op het dorsale deel van de dij. Voer vervolgens een stompe dissectie uit langs de dijbeen biceps. Isoleer de heupzenuwstam zorgvuldig met oogheelkundige schaar en een glasscheidingsnaald en houd het weefsel vochtig met behulp van de oplossing van Ringer.Bevestig het dier vervolgens op een zelfgemaakte metalen hoepel door de huid in de sleuf eromheen te naaien. Trek de huid iets omhoog om een vloeibaar bad vast te stellen. Stel een centimeter heupzenuwstam aan de proximale kant bloot.Plaats een klein, bruin platform onder de zenuwslam om het contrast te verbeteren om de fijne zenuwstam duidelijk te observeren. Breng vervolgens wat warme vloeibare paraffine aan op de bovenkant van de zenuwstam om het drogen van het vezeloppervlak te voorkomen. Verwijder de vloeistof als er een exsudatie rond de zenuwslamst.Als u de opname wilt opnemen, selecteert u de platinafilament als opnameelektrode. Eet en maak een kleine haak aan het einde. Bevestig daarna de elektrode aan een micromanipulator.Plaats in het bad een referentieelektrode naast het onderhuidse weefsel. Splits de spinale dura en de Pia mater en verkrijg de heupzenuw. Onder een stereomicroscoop bij 25% vergroting, pak een fijne fascicle en schort het ongeveereximale uiteinde van de axon op de haak van de opname-elektrode.Identificeer het ontvankelijke veld van een enkele, niet-conceptieve c-vezel met behulp van een mechanische simulis en thermische stimulus. Als het afvuren van de zenuwvezel reageert op de mechanische stimuli en warm water, dan beschouwen als een polymoto, niet-conceptieve c vezel. Steek vervolgens twee naaldprikkelelektroden met een interval van twee millimeter in de huid van het identificerende veld voor de levering van de elektrische stimuli.Geef de golfvorm van een actiepotentieel weer op de oscilloscoop en gebruik computer een d-board met een signaalbemonsteringssnelheid van 20 kilohertz. Neem vervolgens de pieken op met behulp van software voor het verkrijgen van gegevens en analyseer deze later met professionele software. Om geleidingsfalen te meten, leveren de repetitieve elektrode stimuli in verschillende frequenties aan een c vezel gedurende 60 seconden.Laat een interval van 10 minuten voor de vezel om te ontspannen tussen stimuli. Bereken vervolgens de verhouding tussen het aantal storingen en het aantal geleverde repetitieve stimuluspulsen en vermenigvuldig met 100%om de mate van geleidingsfalen te verkrijgen. Om de dorsale wortel ganglion bloot te leggen, eerst snijd de huid van de middellijn van de rug op de l4 naar l5 segment.Verwijder vervolgens de spieren wervelkolom proces wervel bor en dwars proces met behulp van een bot rongeur, en bloot het ruggenmerg NDRG lichaam. Bedek het blootgestelde ruggenmerg, NDRG, met katoen gedrenkt met de extracellulaire oplossing van Ringer om neurale activiteit te behouden. Stop het bloeden en verwijder het bloed indien nodig.Verwijder vervolgens l4 met behulp van oogheelkundige scharen tot s1-botstructuur boven de wervelschaamen om de DRG en de aangesloten ruggenmerg aan te stellen. Maak een snee op de huid om de heupzenuw bloot te leggen aan de middelste dij. Scheid en los de heupzenuw van het distale uiteinde van de zenuw waar het gaat in de spier.En ligate de zenuwstam met chirurgische lijn aan het einde van de zenuw voorafgaand aan het snijden. Scheid vervolgens de heupzenuw van het onderliggende bindweefsel door het zenuwligatiepunt op te heffen. Verwijder voor dura uit het ruggenmerg, en scheid de DRG van het onderliggende bindweefsel totdat het het aangrenzende deel van de heupzenuw bereikt.Zo isoleren de hele voorbereiding van DRG met een bijgevoegde heupzenuw. Verwijder bij 4x vergroting het oppervlak van de DRG voorzichtig met behulp van een pincet op het oppervlak van l4 tot l6 DRG. Plaats de DRG met bijgevoegde heupzenuw in een glazen buis met een milliliter gemengde enzymen.Verteren in een 37 graden Celsius waterbad gedurende 15 minuten. Til na 15 minuten het einde van de chirurgische lijn op en verplaats het preparaat naar een schotel gevuld met de extracellulaire oplossing van De Ringer om het enzym uit te wassen. Breng vervolgens de verteerde DRG over naar een container gevuld met de extracellulaire oplossing van OxygenEur voor het opnemen.Als u de opname wilt opnemen, bereidt u intracellulaire oplossing voor en slaat u deze op bij nul graden Celsius tot gebruik. Met behulp van een slice anker, stabiliseren van de ganglia en sluit de zenuw einde aan een zuigkracht stimulerende elektrode. Bij 40x vergroting, visualiseren en selecteer een DRG neuron met een water emersion doelstelling.Vouw een elektrode en vul deze met intracellulaire oplossing. Bevestig de elektrode op de houder en breng positieve druk in de pipet met een uiteindelijke weerstand van vier tot zeven megaohms. Breng vervolgens de elektrode naar het celoppervlak.Breng vervolgens negatieve druk uit op de pipet om een afdichting te vormen. Zodra een gigaohm-afdichting is bereikt, stelt u het membraanpotentieel in op ongeveer min 60 millivolt en stelt u alle celopnamemodus vast. Vervolgens, leveren repetitieve stimuli van 5 tot 50 hertz aan de heupzenuw de door de zuigelektrode te screenen op geleiding falen.Meet de amplitude van AHP van baseline tot piek en de 80%AHP duur. Dit cijfer toont de originele opeenvolgende opnames van enkele c5 re-firings van ratten in reactie op 10 hertz elektrische stimulatie. Elke twintigste sweep wordt getoond en weergegeven van boven naar beneden.De insert toont een representatief actiepotentieel. Hier zijn de opnames van enkele c vezels van CFA geïnjecteerd ratten in reactie op dezelfde stimulatie als het vorige paneel. Dit cijfer toont de continue opnames van serie afvuren reacties op vijf hertz stimulatie onder controleomstandigheden, of toediening van verschillende concentratie van ZD7288 in een kleine diameter DRG neuron van CFA beoordeelde ratten.De insets tonen uitgebreide sporen voor de opgegeven opnameperioden. Donkere vlekken vertegenwoordigen spike mislukkingen. De AHP toonde een grotere stijgende helling in de controle.Terwijl een kleinere stijgende helling na 125 micromolars bij toepassing ZD7288 werd waargenomen.
