在这里,我们描述了一个简单的方法,我们开发解剖神经元细胞结构的体内基因控制。基于神经元前体的体外工程,将其移植到新生儿大脑中,并配对对"测试"和"控制"细胞的形态评价,从而在快速且经济实惠的方式。为了研究神经元结构的体内基因控制,必须解决三个关键技术问题:(1)实现适当的模式性兴趣基因(GOI)表达和准确的定量控制;(2) 获得不同神经元轮廓的恰当分段可视化;(3) 在雇用有限数量的动物的同时,得出结果的统计意义。
如果可用,携带四环素(tt)控制的转基因的小鼠突变线可能是解决第一个问题1的最佳工具。或者,可以采用体细胞转因。在这种情况下,转基因通过电穿孔2或病毒转导3传递。接下来,它被保留为表皮体(例如,在标准电穿孔4)或它集成到基因组(随机,通过逆转录病毒整合5;或在一个定义的位置,通过CRISPR促进的同源重组(SLENDR)6).
其次,神经元轮廓可视化可以通过 (a) 稀疏均匀标记或 (b) 密集差分标记来实现。至于稀疏标记,先进的高尔基样方法可采用7种,选定的神经元小组可以由生物细胞蛋白8填充,而盐和胡椒标签可以通过稀疏表达的转基因获得。这种转基因可能显示变异转录(Thy-EGFP)9,或者可以通过随机重组(MORF)10激活。 至于(b),最先进的策略包括多絮荧光蛋白转基因阵列(Brainbow)11中的Cre介导随机重组,以及以前由猪Bac-转位酶驱动的荧蛋白基因基因组整合通过体细胞转发生(CLoNE)12.
至于第三个问题,形态学结果往往受到来自动物间差异和细胞注射意外性的巨大随机变异的影响。因此,通常使用大量的动物来达到评估GOI形态活动所需的统计能力。
前面提到的方法往往依靠先进的技术技能,需要明显的财政资源,这可能限制它们在科学界的传播。为了规避这些问题,我们设想了一个简单明了的管道,以快速、经济的方式解剖体内神经结构的基因控制。其灵感来自于以前为快速体内评估抗冲击转基因活性13而开发的类似联合移植设计。
具体来说,人们认为,将体外工程"绿色"神经前体("测试"和"控制"细胞)共同移植到"黑色"受体新生儿大脑中可以同时解决上述三个关键问题。事实上,前体的体外慢病毒工程,在控制良好的条件下,允许保持神经元转基因表达的可变性至少,远远低于通常与体内体细胞操作(以前执行14,15和我们未发布的结果)。由此产生的基因表达的准确控制与tt控制的转基因模型相比。然而,这个程序的成本远远低于那些源于转基因小鼠生产线的维护。其次,手部细胞注射很容易,需要最少的训练。此外,可以很方便地调整注入每个大脑的标记前体的数量,以达到足够多分布的前体累积数量,同时将移植动物的总数保持在最低限度。最后但并非最不重要的,共同注射不同含氟标记的"测试"和"控制"前体,以及随后对结果的成对统计分析,抵消了动物间实验变异性的影响,使统计意义的结果,即使分析人数有限的个人13。
应该强调的是,这种方法虽然快速而便宜,但主要有两个局限性。首先,它旨在研究神经元结构的细胞自主基因控制,并不适合解决环境控制问题。其次,当移植的神经元前体通过异形时间安排到达其最终位置时,这种方法比模型神经架构控制更可取。
