In onze assay gebruikten we pNPP als de ALP substraat. Dit is een werkende oplossing ontworpen voor ELISA en niet te worden verdund. Na hydrolyse door ALP, een gele product ontwikkelt en kan worden gemeten op 405 nm. Alkalische fosfatase (ALP) is overal uitgedrukt in zowel prokaryote en eukaryote cellen1. Het kan het katalyseren van de hydrolyse van monofosfaat uit verschillende moleculen zoals nucleotiden, eiwitten, alkaloïden, fosfaatesters en afgeleide anhydriden van fosforzuur. Bij de mens is eukaryote ALP aanwezig in vele weefsels, waaronder bot, lever, darm en placenta2. Het speelt een belangrijke rol in proteïne fosforylatie, celgroei, apoptosis, stamcel processen evenals normale skelet mineralisatie. Eukaryotische ALP is ook een belangrijke serum-indicator voor de aanwezigheid van ziekten in bot, lever en andere weefsels/organen wanneer verhoogde3,4.
Prokaryote ALP is geconstateerd in een verscheidenheid van bacteriële cellen, met inbegrip van E. coli5, S. aureus6,7 en sommige gemeenschappelijke pens bacteriën in de bodem-8. Bacteriële ALP activiteit is gebruikt als een biosensor in de detectie van bacteriële besmetting10, pesticiden en zware metalen9 . De constitutieve uitdrukking van ALP is gebruikt om te identificeren van de staphylococcen11 en Serratia onderscheiden van Enterobacter12. Verder wordt voorgesteld dat de constitutieve ALP productie is gecorreleerd met pathogeniteit in staphylokokken13. Hoewel ALP is onderzocht in verschillende instellingen,3,4, is nog weinig gerapporteerd voor de activiteit en de functie in biofilms culturen.
Een biofilm heeft gedocumenteerd te hebben een verschillend functionerende bacteriële leven ten opzichte van haar free-living bacteriële cel tegenhanger14. In S. aureus, is de vorming van een biofilm geconstateerd in een verscheidenheid van klinische omstandigheden en rekeningen voor de resistentie tegen antibiotica en chronische ontsteking15,16. Veel moleculen te vinden in een biofilm matrix zoals polysacchariden, eiwitten, nucleïnezuren en lipiden had gemeld, maar het mechanisme van biofilm vorming is niet volledig begrepen14. Om te begrijpen van de rol van ALP in biofilm vorming, we gekweekt van S. aureus biofilms in 96 goed weefselkweek platen en de activiteit van de ALP met behulp van para-nitrophenylphosphate (pNPP) gemeten.
De pNPP van het molecule is een kant-en-klare substraat voor ALP en is wijd verbeid gebruikt voor het meten van de ALP activiteit6,17,18. Deze colorimetrische bepaling is gebaseerd op de omzetting van para-nitrofenyl fosfaat (pNPP) te para-nitrofenol, resulterend in een gekleurd product op 405 nm. Vergeleken met andere conventionele ALP assay, zoals agarose gel elektroforese19, tarwekiemen agglutinin (WGA) neerslag, en WGA-HPLC20, deze test zeer specifiek is, zorgt gevoelig, gemakkelijk te reproduceren, en de meeste bovenal voor hoge doorvoer.

Alkalische fosfatase (ALP) is een gemeenschappelijk enzym uitgedrukt in zowel prokaryote en eukaryote cellen. Het katalyseert de hydrolyse van fosfaat monoesters uit veel moleculen met een pH van fundamentele en speelt een onmisbare rol in het metabolisme van fosfaat. Bij de mens is eukaryote ALP een van de meest gebruikte enzymatische signalen in de diagnose van verschillende ziektes zoals cholestase en rachitis. In S. aureus, ALP wordt gedetecteerd uitsluitend op de celmembraan; het wordt ook uitgedrukt als een secretoire vorm ook. Nog, er is weinig bekend over de functie bij de vorming van biofilms.
Het doel van dit manuscript is om een snelle en betrouwbare test voor het meten van de ALP activiteit in S. aureus biofilm die geen eiwit isolatie vereist. Met behulp van p-nitrofenyl fosfaat (pNPP) als substraat, we gemeten ALP activiteit in S. aureus biofilm gevormd in de weefselkweek 96-wells-platen. Activiteit is gebaseerd op de vorming van de oplosbare reactieproduct gemeten door absorptie van 405 nm. Figuur 1 toont een representatief resultaat van ALP activiteit van biofilm culturen van S. aureus in 96 goed weefselkweek platen. 75 μL van verkrijgbare pNPP oplossing was toegevoegd aan elk goed en bebroede bij kamertemperatuur. Na incubatie gedurende verschillende tijden (0 min, 15 min, 30 min, 45 min, 60 min en 75 min, respectievelijk) 75 μL van 5 M NaOH toevoegde aan zet de reactie stop. Dit protocol presenteerde een eenvoudige en betrouwbare methode om gealkde fosfataseactiviteit in de biofilmcultuur van S.aureus te meten. Het zal ons helpen om de functie van ALP in biofilmvorming te begrijpen. Het grote voordeel van deze techniek is een hoge doorvoer.Het is zeer gevoelig en gemakkelijk uit te voeren. Demonstreren van de procedure zal Kevin Danikowski, een voormalige student van Harper College. Om TSB voor te bereiden, voeg alvleeskliervertavel van caseïne, papaïsche vertering van sojameel, natriumchloride, dextrose en dipotassiumfosfaat toe aan gedestilleerd water.Bedek de kolf met aluminiumfolie en doe het in een autoclave om te steriliseren voor gebruik. Om een TSA voor te bereiden, voeg agar toe aan TSB en zet het in de autoclave voor sterilisatie. Laat het dan afkoelen tot kamertemperatuur.Giet het vervolgens in de petrischaal met een verhouding van 20 milliliter per gerecht. Om een biofilm kweekmedium voor te bereiden, voeg glucose toe aan autoclaved TSB om de concentratie van 10 gram per liter te bereiken. Met behulp van een vacuümfiltratiekit filtrate de oplossing met een filter van 0,2 micrometer.Om te inenten, gebruik een gesteriliseerde inentingslus om een individuele kolonie Staph aureus uit de petrischaal te plukken en deze in de kweekbuis te plaatsen gevuld met 10 milliliter TSB. Zet de buis in de couveuse om 's nachts te groeien op 37 graden Celsius. Met een micropipet, teken 100 microliters van 's nachts cultuur en breng het naar een cultuur buis gevuld met 10 milliliter TSB met glucose voor een 1:100 volume verhouding verdunning.Vortex voorzichtig te mengen voor een consistente concentratie. Gebruik een micropipet om 200 microliter van verdunde cultuur over te brengen in een totaal van 18 putten van een 96 put plaat. Incubeer de 96 put plaat op 37 graden Celsius gedurende 24 uur voor biofilm vorming.Voor de beste biofilmvorming van S.aureus is het beter om een platte of ronde bodem 96 putplaat te kiezen. Daarna kantel je de plaat iets om de supernatant te decanteren door aspiratie vanaf de rand van elke put. Was elk goed met 1X PBS.Doe de plaat in de centrifuge om vijf minuten te draaien bij 8.000 omwentelingen per minuut en decanteer de supernatant door aspiratie. Voeg 75 microliters gebufferd ALP-substraat toe bestaande uit commercieel verkrijgbaar PNPP aan elke put en gebruik een timer om elk keer punt in drievoud op te nemen. Voeg na elk incubatietijdpunt 75 microliters van vijf molaire natriumhydroxide toe aan elk van de drie putten om de reactie te stoppen.Centrifugeer de plaat gedurende vijf minuten bij 800 omwentelingen per minuut. Gebruik een pipet om 100 microliter supernatant over te brengen in een nieuwe 96 putplaat voor calorimetrische meting. Gebruik een 96 goed plaatlezer om de absorptie van elke put te meten op 405 nanometer.Gebruik de tijdputten van 30 minuten om achtergrondgeluid te regelen. Herhaal het hele experiment in drie 96 putplaten. Dit protocol ontwikkelt een snelle en betrouwbare test om alp-activiteit in S.aureus biofilm te meten die geen eiwitisolatie vereist.Het representatieve resultaat van de ALP-activiteit laat een verhoogde trend zien gedurende maximaal 75 minuten. Na 75 minuten werd geen toename van de ALP-activiteit waargenomen. Na de ontwikkeling u potentiële ALP-remmers screenen die de vorming van biofilms kunnen verstoren. Het resultaat zal belangrijke informatie in de richting van biofilm management in medisch veld.Vijf molaire natriumhydroxide kan gevaarlijk zijn in geval van contact.
