Alkalischer Phosphatase (ALP) drückt sich ubiquitär in beide prokaryotischen und eukaryotischen Zellen1. Es kann katalysieren die Hydrolyse von Monophosphate aus unterschiedlichen Molekülen wie Nukleotide, Proteine, Alkaloide, Phosphat Ester und Anhydride der Phosphorsäure. Beim Menschen ist die eukaryotische ALP in vielen Geweben einschließlich Leber, Knochen, Darm und Plazenta2. Es spielt eine wichtige Rolle in Protein-Phosphorylierung, Zellwachstum, Apoptose, Stammzell-Prozesse sowie normale skelettartigen Mineralisierung. Eukaryotische ALP ist auch eine wichtige Serum-Indikator für das Vorliegen von Krankheiten in Knochen, Leber und andere Gewebe/Organe, wenn erhöht3,4.
Prokaryotische ALP wurde in einer Vielzahl von bakteriellen Zellen, einschließlich der E. Coli-5, S. Aureus6,7 und einige gemeinsame Pansen-Bakterien in die Böden8erkannt. Bakterienaktivität ALP wurde als Biosensor zur Erkennung von Pestiziden, Schwermetallen9 und bakterielle Kontamination10verwendet. Alkalischer Phosphatase (ALP) ist eine gemeinsame Enzym in prokaryotischen und eukaryotischen Zellen exprimiert. Es katalysiert die Hydrolyse von Phosphat Monoesters aus vielen Molekülen bei grundlegenden pH und Phosphat-Stoffwechsel spielt eine unverzichtbare Rolle. Beim Menschen ist eukaryotische ALP eines der am häufigsten verwendete enzymatische Signale bei der Diagnose verschiedener Krankheiten, wie z. B. Cholestase und Rachitis. In S. Aureus, ALP wird ausschließlich auf der Zellmembran erkannt; Es wird auch als sekretorischen Form sowie ausgedrückt. Allerdings ist wenig über seine Funktion im Biofilmbildung bekannt.
Dieses Manuskript soll einen schnellen und zuverlässigen Assay zur ALP Aktivität in S. Aureus Biofilm zu messen, die kein Protein Isolierung erfordert zu entwickeln. Mit p-Nitrophenyl-Phosphat (pNPP) als Substrat, messen wir ALP Aktivität in S. Aureus Biofilm in 96-Well-Zellkultur-Platten gebildet.
