Huid heeft betrekking op de buitenste oppervlakte van het lichaam en beschermt de inwendige organen. De huid heeft verschillende functies, met inbegrip van bescherming tegen ziekteverwekkers, opvangen en opslaan van water, regulering van de lichaamstemperatuur, en lichaam afscheiden afval2. Huid protheses kunnen worden ingedeeld, afhankelijk van de bron van de huid; transplantaties met behulp van de huid van een andere donor allografts worden genoemd, en enten met de huid van de patiënt zijn autografts. Hoewel het een autograft is de aangewezen behandeling vanwege zijn lage afwijzing risico, zijn huid biopsieën moeilijk uit te voeren op patiënten met ernstige letsels of een onvoldoende aantal huidcellen. Bij patiënten met ernstige brandwonden zijn drie keer het aantal huidcellen nodig om grote gebieden bestrijken. De beperkte beschikbaarheid van huidcellen van het lichaam van een patiënt leidt tot situaties waarin allogenous transplantatie noodzakelijk is. Een allograft wordt tijdelijk gebruikt totdat autologe transplantatie kan worden uitgevoerd omdat het meestal wordt afgewezen door het immuunsysteem van de gastheer na ongeveer 1 week3. Om te overwinnen afwijzing door het immuunsysteem van de patiënt, moeten protheses komen uit een bron met dezelfde immuun identiteit als de patiënt4.
Menselijke iPSCs zijn een opkomende bron van cellen voor stamcel therapie5. Menselijke iPSCs worden gegenereerd uit lichaamscellen, met behulp van herprogrammering factoren zoals OCT4, SOX2, Klf4 en c-Myc6. Met behulp van menselijke iPSCs overwint de ethische en immunologische kwesties van embryonale stamcellen (SER's)7,8. Menselijke iPSCs pluripotent hebben en kunt onderscheiden in drie lagen van de kiem9. De aanwezigheid van HLA, een kritische factor in de regeneratieve geneeskunde, bepaalt de immuunrespons en de mogelijkheid van verwerping10. Het gebruik van patiënt-afgeleide iPSCs verhelpt de problemen van cel-source beperking en immuunsysteem afwijzing. CBMCs hebben ook naar voren gekomen als een bron van alternatieve cel voor regeneratieve geneeskunde11. Verdere, homozygoot HLA-type iPSCs kunt grote schaal toepassen van verschillende patiënten12. Een bank CBMC-iPSC is een roman en efficiënte strategie voor celtherapie en allogene regeneratieve geneeskunde12,13,14. Resultaten van deze studie suggereren dat een 3D huid CBMC-iPSC-afgeleide organoid een nieuwe tool voor in vitro en in vivo dermatologische onderzoek is.

De huid is het grootste orgaan van het lichaam en heeft vele functies. De huid fungeert als een fysieke barrière en de beschermer van het lichaam en regelt lichaamsfuncties. Biomimetics is de nabootsing van de modellen, systemen en elementen van de natuur met het oog op het oplossen van complexe menselijke problemen1. Huid biomimetics is een nuttig instrument voor onderzoek van de ziekte in vitro en in vivo regeneratieve geneeskunde. Menselijke geïnduceerde pluripotente stamcellen (iPSCs) hebben het kenmerk van onbeperkte proliferatie en de mogelijkheid van differentiatie op drie lagen van de kiem. Menselijke iPSCs worden gegenereerd uit verschillende primaire cellen, zoals cellen van het bloed, keratinocyten, fibroblasten en meer. Onder hen, hebben koord bloed mononucleaire cellen (CBMCs) naar voren gekomen als een bron van alternatieve cel vanuit het perspectief van allogene regeneratieve geneeskunde. CBMCs zijn nuttig in de regeneratieve geneeskunde omdat menselijke leukocyten antigeen (HLA) te typen naar de cel bankwezen essentieel. Wij bieden een methode voor de differentiatie van CBMC-iPSCs in keratinocyten en fibroblasten en generatie van een 3D huid organoid. Keratinocyten CBMC-iPSC-afgeleide en fibroblasten hebben kenmerken vergelijkbaar met een oplaadbare batterij-lijn. De 3D huid organoids worden gegenereerd door een epidermale laag op een dermale laag bedekken. Door het transplanteren van deze 3D huid organoid, wordt een gehumaniseerd muizen model gegenereerd. Deze studie toont aan dat een 3D menselijke iPSC afkomstige huid organoid een nieuwe, alternatieve tool voor dermatologische onderzoek in vitro en in vivo wellicht.

Navelstrengbloed mononucleaire cellen, of CBMC, zijn naar voren gekomen als een potentiële cel bron voor regeneratieve geneeskunde, omdat menselijke leukocyten antigeen typen is essentieel voor de cel banksysteem. Door CBMC geïnduceerde pluripotente stamcellen, of CMBC-iPSCs, kunnen worden onderscheiden in keratinocyten en fibroblasten. Om 3D-huidorganoïde te genereren, gebruiken we dermatologisch onderzoek.Begin met het kweken van CBMC-iPSC op vitronectine-gecoate 100-milliliter platen bij 37 graden Celsius en 10% kooldioxide. Wanneer de cellen hebben bereikt 80%samenvloeiing, was de culturen met PBS, en behandel ze met een milliliter van een-millimolar EDTA per plaat gedurende twee minuten op 37 graden Celsius en 5%kooldioxide. Wanneer de cellen zijn losgemaakt, stop de reactie met drie milliliter E8 medium per plaat, en verzamel de cellen door centrifugatie.Schors de pellets opnieuw in vijf milliliter E8-medium voor het tellen en breng één keer-10-tot-de-zesde cellen over naar een conische buis van 15 milliliter voor centrifugatie. Schort de overgedragen cellen opnieuw op met 2,5-milliliter embryonale lichaamsvormingsmedium, aangevuld met 10-micromolar rho-associated kinase, of ROCK-remmer. Gebruik een pipet om 125 microliter druppeltjes cellen over te brengen op een ongecoat kweekplaatdeksel.Wanneer alle druppels zijn geplaatst, draai de schotel om de druppels te hangen aan het deksel voor 24 uur op 37 graden Celsius en 5%kooldioxide. Was de volgende dag het deksel met een vers E8-medium en breng de embryonale lichaamsoplossing over op een conische buis van 50 milliliter. Laat de embryonale lichamen genoegen nemen met een minuut bij kamertemperatuur, voordat het de supernatant aanspireert en de embryonale lichamen opnieuw opschont met vers E8-medium voor hun cultuur in een petrischaaltje van 90 millimeter bij 37 graden Celsius en 5% kooldioxide tot hun differentiatie.Voor CBMC-iPSC keratinocyten differentiatie, vervang het E8 medium uit de embryonale lichaamscultuur door verse E8 medium, aangevuld met een nanogram per milliliter bot morfogenetische eiwit 4 voor een 24-uurs incubatie bij 37 graden Celsius en 5% kooldioxide. De volgende dag, oogst de embryonale lichamen in een 50-milliliter conische buis zoals aangetoond, en laat de lichamen om zich te vestigen voor een minuut op kamertemperatuur. Vervang vervolgens de supernatant door zes milliliter keratinocyten differentiatie medium 1, of KDM1, aangevuld met 10 micromolar ROCK remmer, en breng de embryonale lichamen naar een Type IV Collageen-gecoate 100-millimeter schotel.Op dagen nul tot en met acht van de cultuur, vervang het medium om de andere dag door verse KDM1, aangevuld met drie-micromolar retinoïnezuur en 25 nanogram per milliliter elk van bot morfogenetische eiwit 4 en epidermale groeifactor. Tussen dagen negen tot en met 12, vervang het medium om de andere dag door KDM2, aangevuld met drie-micromolar retinoïnezuur, 25 nanogram per milliliter bot morfogenetische eiwit 4, en 20 nanogram per milliliter van epidermale groeifactor. Tussen dagen 13 tot en met 30, verander het medium om de andere dag naar KDM3, aangevuld met 10 nanogram per milliliter bot morfogenetische eiwit 4, en 20 nanogram per milliliter van epidermale groeifactor.Voor CBMC-iPSC fibroblast differentiatie, opnieuw schorsen van de embryonale lichaamscultuur in zes milliliter van fibroblast differentiatie medium 1, aangevuld met 10-micromolar ROCK remmer, en breng de embryonale lichaamssuspensie naar een kelder membraan matrix-gecoate 100-millimeter schotel. Behandel de cultuur na drie dagen incubatie gedurende vier dagen met 0,5 nanomolar botmorfogenetisch eiwit 4, voordat u het medium tot fibroblast differentiatie medium 2 verandert. Op dag zeven van de cultuur, verander het medium naar FDM2 om de andere dag voor een week.Op dag 14 van de cultuur, los de cellen met een-millimolar EDTA zoals aangetoond, en het verzamelen van de cellen in drie milliliter fibroblast differentiatie medium 2 voor centrifugatie. Schorsen de cellen in vijf milliliter fibroblast differentiatie medium 1 voor het tellen, en breng twee keer-10-tot-de-zesde cellen naar een niet-gecoate schotel. Op dag 21 van de cultuur, breng twee keer-10-naar-de-zesde cellen van de cultuur naar een Type I Collagen-gecoate 100-millimeter schotel in verse fibroblast differentiatie medium 1.Op dag 28 van de cultuur, overdracht twee-keer-10-to-the-six van de iPSC-afgeleide fibroblasten naar een nieuwe niet-gecoate schotel. Voor driedimensionale huid organoïde generatie, oogst de iPSC-afgeleide fibroblasten uit de cultuur, en breng twee keer-10-tot-de-vijfde cellen naar een 15-milliliter conische buis voor centrifugatie. Schort de fibroblastpellet opnieuw op in 1,5 milliliter fibroblastdifferentiatie medium 1 en 1,5 milliliter geneutraliseerde Type I Collagen-oplossing.Incubeer de cellen op een insert in een zes-well microplaat gedurende 30 minuten bij kamertemperatuur. Wanneer de oplossing is gestold, voeg twee milliliter medium aan de bovenkant van de insert, en drie milliliter aan de onderkant van de put. Plaats vervolgens de fibroblastmatrix vijf tot zeven dagen in de couveuse, totdat de gelatie is voltooid en de matrix niet meer samentrekt.Aan het einde van de incubatie, oogst de iPSC-afgeleide keratinocyten, en breng een keer-10-tot-de-zesde cellen naar een 15-milliliter conische buis voor centrifugatie. Stel de pellet opnieuw op in 50 tot 100 microliter met een laag calciumepitheelmedium 1 en vervang het medium over de fibroblastmatrix door de keratinocytenceloplossing. Vervang na 15 minuten bij 37 graden Celsius het medium aan de bovenkant van de wisselplaat door twee milliliter epitheelmedium 1 en het medium in de onderste put met drie milliliter van hetzelfde en breng de plaat terug naar de couveuse.Na twee dagen, vervang het medium in de insert en de put met normale calcium epitheel medium 2 voor twee extra dagen van de cultuur. Op dag vier van de cultuur, verwijder alle van het medium, en voeg drie milliliter van cornificatie medium aan de bodem goed alleen, om een lucht-vloeibare interface te genereren. Breng de plaat vervolgens tot 14 dagen terug naar de couveuse voordat de 3D-huidorganoïde wordt geoogst voor downstream-analyse.CBMC-iPSC-afgeleide keratinocyten hebben morfologie vergelijkbaar met primaire keratinocyten, en de expressie van keratinocyten markers wordt verhoogd in deze cellen. CBMC-iPSC-afgeleide fibroblasten hebben morfologie vergelijkbaar met primaire fibroblasten, en de expressie van pluripotente stamcelmarker oct-4 is down-gereguleerd, terwijl fibroblast markers zijn up-gereguleerd. De dikte van de 3D-huidorganoïde neemt toe tijdens de 3D-cultuur, wat bevestigt dat de 3D-huidorganoïde wordt gegenereerd uit keratinocyten en fibroblasten van iPSC.Na twee weken, een getransplanteerde 3D huid organoïde graft efficiënt opneemt in een ontvanger muishuid, zoals bevestigd door HNE analyse. Verder worden keratinocytenrijpting en epidermale differentiatiemarkers uitgedrukt in de CBMC-iPSC-afgeleide 3D-huidorganoïden, die een functionele differentiatie, efficiënte enting en effectieve genezing van de huidafwijkingen van de muis aantonen. We gebruiken in het verleden met het hoge calciummedium dat gestratificeerde lagen van 3D-huidorganoïde induceert om de huid in de buurt na te bootsen.analyse toont aan dat de huid is gestratificeerd. CBMC-iPSCs zijn een potentiële celbron voor huidtransplantaten, en CBMC-iPSC-afgeleide 3D-huidorganoïden kunnen worden gebruikt in studies met betrekking tot dermatologie, cosmetische screening en regeneratieve geneeskunde.
