ヒト白血球抗原タイピングは細胞バンキングシステムに不可欠であるため、臍帯血単核細胞(CBMC)は再生医療の潜在的な細胞源として出現している。CBMC誘導多能性幹細胞、またはCMBC-iPSCは、ケラチノサイトおよび線維芽細胞に分化することができる。3D皮膚オルガノイドを生成するために、我々は皮膚科の研究を使用しています。CBMC-iPSCをビトロネクチンコーティングされた100ミリリットルプレートに摂氏37度と10%の二酸化炭素で培養することから始めます。細胞が80%の合流度に達したら、PBSで培養物を洗浄し、1皿当たり1ミリリットルの1ミリリットルで摂氏37度と5%の二酸化炭素で2分間処理します。細胞が剥離したら、1皿当たり3ミリリットルのE8培地で反応を停止し、遠心分離によって細胞を採取する。ペレットを計数用のE8培地5ミリリットルに再懸濁し、1回10〜6番目の細胞を15ミリリットルの円錐形チューブに移して遠心分離する。2.5ミリリットルの胚体形成培地で移された細胞を再中断し、10−マイクロモルロー関連キナーゼ、またはROCK阻害剤を添加する。ピペットを使用して、125マイクロリットルの細胞液滴をコーティングされていない培養プレートの蓋に移します。すべての液滴が置かれたら、皿をひっくり返して、液滴が37°Cと5%の二酸化炭素で24時間蓋からぶら下がるようにします。翌日、新鮮なE8培地で蓋を洗浄し、胚体溶液を50ミリリットルの円錐チューブに移します。胚体が室温で1分間沈着することを許可し、上清を吸引し、培養用の新鮮なE8培地で胚体を摂氏37度で90ミリメートルのペトリ皿に、分化するまで5%の二酸化炭素を再懸濁させる。CBMC-iPSCケラチノサイト分化の場合、胚体培養中のE8培地を新鮮なE8培地に置き換え、1ミリリットル当たり1ナノグラムの骨形態形成タンパク質4を加え、摂氏37度および5%の二酸化炭素で24時間インキュベーションする。翌日、実証されているように胚体を50ミリリットルの円錐形チューブに収穫し、体が室温で1分間沈着することを可能にする。次に、上清をケラチノサイト分化培地1、またはKDM1の6ミリリットルに置き換え、10ミクロモルROCK阻害剤を添加し、胚体をIV型コラーゲンコーティング100ミリメートル皿に移す。培養のゼロから8日目に、1日おきに培地を新鮮なKDM1に置き換え、骨形態形成タンパク質4および表皮成長因子のそれぞれを1ミリリットル当たり3マイクロモルモルノイン酸および25ナノグラムで補う。9日目から12日目の間に、1日おきに培地をKDM2に置き換え、3マイクロモルモルモルレチノイン酸、骨形態形成タンパク質4のミリリットル当たり25ナノグラム、および表皮成長因子のミリリットル当たり20ナノグラムを補う。日13〜30の間に、1日おきに培地をKDM3に変え、骨形態形成タンパク質4の1ミリリットル当たり10ナノグラム、および表皮成長因子の1ミリリットル当たり20ナノグラムを補う。CBMC-iPSC線維芽細胞分化の場合、胚体培養を線維芽細胞分化培地1の6ミリリットルで再中断し、10マイクロモルロック阻害剤を添加し、胚体懸濁液を基底膜マトリックスコーティング100ミリメートル皿に移す。3日間の培養後、0.5ナノモル骨形態形成タンパク質4で培養を4日間、その前に培地を線維芽細胞分化培地2に変更する。7日目の培養で、培地を1日おきに1週間ずつFDM2に変更します。培養の14日目に、実証したように1ミリモルEDTAで細胞を取り外し、遠心分離のために3ミリリットルの線維芽細胞分化培地2の細胞を回収する。細胞をカウント用の線維芽細胞分化培地1の5ミリリットルに再懸濁し、2回10〜6番目の細胞を非コーティングされた皿に移す。培養21日目に、培養物から2回10〜6番目の細胞を、新鮮な線維芽細胞分化培地1でI型コラーゲンコーティング100ミリメートル皿に移す。培養28日目に、iPSC由来の線維芽細胞の2倍の10〜6個を新しい非コーティング皿に移す。3次元皮膚オルガノイド生成のために、培養物からiPSC由来の線維芽細胞を収穫し、遠心分離のために15ミリリットル円錐形のチューブに2回10〜5番目の細胞を移す。線維芽細胞ペレットを1.5ミリリットルの線維芽細胞分化培地1、および1.5ミリリットルの中和型I型コラーゲン溶液中和で再懸濁する。6ウェルマイクロプレートのインサート上の細胞を室温で30分間インキュベートします。溶液が固まったら、インサートの上部に2ミリリットルの培地を加え、ウェルの底に3ミリリットルを加えます。次に、ゲル化が完了し、マトリックスが収縮しなくなるまで、5〜7日間、線維芽細胞マトリックスをインキュベーターに入れる。インキュベーションの終わりに、iPSC由来の角化細胞を収穫し、遠心分離のために15ミリリットルの円錐形チューブに1回10〜6番目の細胞を移す。ペレットを低カルシウム上皮培地1の50~100マイクロリットルに再懸濁し、繊維芽細胞マトリックス上の培地をケラチノサイト細胞溶液に置き換えた。摂氏37度で15分後、挿入物の上部にある培地を上皮培地1の2ミリリットルに、底面の培地を同じ3ミリリットルで交換し、プレートをインキュベーターに戻します。2日後、挿入物中の培地とウェルを、さらに2日間培養するために通常のカルシウム上皮培地2に交換する。培養4日目に、培地をすべて取り除き、3ミリリットルのコンニフィケーション培地を底ウェルにのみ添加し、空気液体界面を生成する。その後、下流分析のために3D皮膚オルガノイドを収穫する前に、最大14日間、プレートをインキュベーターに戻します。CBMC-iPSC由来の角化細胞は、一次角化細胞と同様の形態を有し、これらの細胞においてケラチノサイトマーカーの発現が増加する。CBMC-iPSC由来の線維芽細胞は原発性線維芽細胞に類似した形態を有し、多能性幹細胞マーカーの発現は10-4でダウンレギュレートされ、線維芽細胞マーカーはアップレギュレートされている。3D培養中に3D皮膚オルガノイドの厚さが増加し、iPSC由来のケラチノサイトおよび線維芽細胞から3D皮膚オルガノイドが生成されることを確認する。2週間後、移植された3D皮膚オルガノイド移植片は、HNE分析によって確認されるように、レシピエントマウス皮膚に効率的に組み込まれる。また、角化細胞成熟および表皮分化マーカーは、CBMC-iPSC由来の3D皮膚オルガノイドで発現され、機能的分化、効率的な移植、およびマウス皮膚欠損の効果的な治癒を実証する。私たちは、皮膚の近くを模倣するために3D皮膚オルガノイドの層状層を誘導する高カルシウム培地で過去に使用します。分析は、皮膚が階層化されていることを示しています。CBMC-iPSCは皮膚移植片の潜在的な細胞源であり、CBMC-iPSC由来の3D皮膚オルガノイドは、皮膚科学、美容スクリーニング、再生医療に関する研究に使用することができます。
