De methode van het isoleren van pure exosomes is essentieel voor de studie van de functie van exosomes. Duchenne muscular dystrophy (DMD) is een X-gebonden recessieve, progressieve neuromusculaire ziekte veroorzaakt door een mutatie in gen DMD en het verlies van functionele Dystrofine1. Dystrofine voornamelijk in skeletspieren en myocard wordt uitgedrukt, en minder wordt uitgedrukt in gladde spieren, endocriene klieren en neuronen2,3. DMD is de meest voorkomende vorm van spierdystrofie met een incidentie van één per 3.500 tot 5.000 pasgeboren jongens wereldwijd4,5. Individuen ontwikkelen meestal progressieve spier necrose, verlies van onafhankelijke wandelen door vroege adolescentie, en dood in de decennia van de tweede naar de derde van hun leven als gevolg van hartfalen en respiratoir falen6.
Verwijde cardiomyopathie, aritmieën en congestief hartfalen zijn gemeenschappelijke cardiovasculaire uitingen van DMD7,8. De ziekte kan niet worden genezen, ondersteunende behandeling kan symptomen verbeteren of vertragen van de progressie van hartfalen, maar het is heel moeilijk om het hart functie9,10.
Soortgelijk aan DMD patiënten, spierdystrofie X-gebonden (MDX) muizen hebben een tekort in het Dystrofine-eiwit en huidige symptomen van cardiomyopathie11, en worden daarom veel gebruikt bij DMD verbonden cardiomyopathie onderzoek. Om te herstellen van Dystrofine in betrokken spieren, heeft allogene stamcel therapie bewezen een effectieve behandeling voor DMD12,13,14. Exosomes, 30-150 nm membraan blaasjes uitgescheiden door verschillende celtypes, spelen een sleutelrol in de cel-naar-cel communicatie via genetische materiaal vervoer, zoals messenger-RNA (mRNA) en niet-coderende RNAs15,16,17 ,18,19,20,21.
Onze vorige studies hebben aangetoond dat exosomes afgeleid van myogenic voorlopercellen (MPC), zoals C2C12 cellijn, Dystrofine kunt overbrengen mRNA naar host cardiomyocytes na directe cardiale injectie22, waarmee wordt aangegeven dat allogene levering van MPC-afgeleide exosomes (MPC-Exo) kunt Transient DMD genexpressie in MDX-muizen herstellen. Dit artikel richt zich op de zuivering en -transplantatie technieken MPC-Exo.

purification and transplantation of myogenic progenitor cell derived exosomes to improve cardiac function in duchenne muscular dystrophic mice

Duchene Muscular Dystrophy (DMD) is een X-gebonden recessieve genetische ziekte die wordt veroorzaakt door een gebrek aan functionele Dystrofine-eiwit. De ziekte kan niet worden genezen, en naarmate de ziekte vordert, ontwikkelt de patiënt symptomen van verwijde cardiomyopathie, aritmie en congestief hartfalen. De DMDMDX mutant muizen doen niet express Dystrofine en worden vaak gebruikt als een muismodel van DMD. In onze recente studie, die we hebben vastgesteld dat intramyocardial injectie van brede type (WT)-myogenic voorlopercellen cellen afkomstige exosomes (MPC-Exo) Transient hersteld de uitdrukking van Dystrofine in het myocardium van DMDMDX mutant muizen, die werd geassocieerd met een tijdelijke verbetering van de hartfunctie suggereren dat kan WT-MPC-Exo bieden een optie voor het verlichten van de cardiale symptomen van DMD. Dit protocol presenteert de methode om de hartfunctie bij duchenne's spierdystrofie muizen tijdelijk te verbeteren door exon afgeleid van normale myogene voorlopercellen te transplanteren. De exosomale transplantatie kan de hartfunctie bij DMD-muizen verbeteren in combinatie met een verhoogde expressie van dystrofie in de harten van de ontvanger. De juiste methode voor exonisolatie, zuivering en intramyocardiale transplantatie dragen tot het succes van de exontransplantatie bij om hartfunctie voor muizen met de spierdystrofie van Duchenne te verbeteren.MPC-afgeleide exosoomtherapie kan de skeletspierfunctie verbeteren door exosoomgemedieerde normale dystrofische mRNA-overdracht. Deze video toont de kritische technieken voor exosoom zuivering, intramyocardiale exosoom levering, en echocardiografie post-transplantatie. De demonstratie van echocardiografie zal worden gedaan door Yan Shen, technicus van ons lab.Om te beginnen met deze procedure, zaad vijf miljoen C2C12 cellen in een 15 centimeter cultuur schotel met 20 milliliter van volledige DMEM met 10%FBS en antibiotica. Incubeer bij 37 graden Celsius met 5%kooldioxide. Gebruik vervolgens een swingende emmerrotor om FBS op 100,000 keer G en 18 uur lang bij vier graden Celsius te bereiden om exosoomarm medium voor te bereiden.Gooi de pellet weg en verzamel de supernatant. Wanneer de monolaagcellen 80% samenvloeiing in het kweekschotel bereiken, vervangt u de volledige DMEM door exosoomarm medium. Gebruik een transfer pipet om de supernatant te verzamelen van de cel cultuur schotel om de 48 uur.Centrifugeren de buizen op 150 keer G gedurende tien minuten om de resterende cellen te verwijderen. Filtreer de supernatant door een filter van 0,22 micrometer om celpuin te elimineren en het gefilterde medium over te brengen naar ultrahelde buizen. Gebruik vervolgens een swingemmerrotor om de buizen op 100, 00 keer G en 4 graden Celsius gedurende 120 minuten te ultracentrifugeren om de exosomen neer te slaan.Resuspend de exosoom-bevattende pellet in PBS en vul vervolgens de gehele ultra-clear buis met PBS. Ultracentrifuge deze suspensie op 100, 00 keer G en bij 4 graden Celsius gedurende 120 minuten om vervuilende eiwitten te elimineren. Gooi de supernatant weg en verleg de exosoompel in 100 microliter pbs.Bewaar bij min 80 graden Celsius voor toekomstig gebruik. Ten eerste, fix elke verdoofde muis in de supine positie op een chirurgisch platform door het beveiligen van elke ledemaat met tape. Plaats een 3-0 hechting horizontaal onder de boventanden om de bovenkaak op zijn plaats te houden.Breng vervolgens ontharingscrème aan op de linkerkant van de borst van de muis om de spar van de huid te verwijderen. Met behulp van een 24-meter katheter, uit te voeren endotracheale intubatie via de mondholte. En gebruik een knaagdier ventilator om de muis te ventileren met kamerlucht met een snelheid van 195 ademhalingen per minuut.Desinfecteer de huid met 70% alcohol en 10%povidone jodium. Maak vervolgens een schuine incisie van 10 tot 15 millimeter van de linker achtersteven rand naar de linker oksel. Gebruik schaar om de pectoralis major en pectoralis minor te snijden.En maak een linkse thoracotamie door de vierde intercoastal ruimte. Steek voorzichtig oprolbanden in om de borstholte te verspreiden tot een breedte van 10 millimeter, waarbij u ervoor zorgt dat de linkerlong niet wordt beschadigd. Gebruik hierna twee rechte pincet om het pericardium te verwijderen.Trek ze uit elkaar en plaats ze achter de oprolbare tips om het hart bloot te leggen. Gebruik een 31-meter insulinenaald om MPC-Exo of PBS intramyocardiaal te injecteren in de voorste wand van de linkerventrikel op één locatie. Gebruik vervolgens 6-0 nylon hechtingen om de borstholte, de pectoralisspieren en de huid achter elkaar te sluiten.Twee dagen na de PBS exosoomtransplantatie verdoven de muizen zoals beschreven in het tekstprotocol. Gebruik tape om een muis in de supine positie vast te stellen en voorverwarmde akoestische gel toe te passen op de linker borst. Gebruik vervolgens echocardiografie om de linker ventriculaire functie te beoordelen.Verkrijg het zicht op de linkerventrikel in twee dimensies en draai de ultrasone sonde 90 graden om een korte asweergave aan de linker ventrikel op het papillaire spierniveau te verkrijgen. Neem hierna echocardiografische afbeeldingen in de M-modus op en meet de linker ventriculaire einddiastolische diameter, het linker ventriculaire eindsystolische volume, het linker ventriculaire einddiastolvolume en het linker ventriculaire eindsystolische volume. De hartslag van de muizen moet ten minste 400 bpm worden gecontroleerd voor de hartmetingsfunctie.Nadat exosomen zijn geïsoleerd en gezuiverd uit C2C12-cellen, wordt hun aanwezigheid bevestigd met behulp van transmissieelektronenmicroscopieanalyse. Het transmissieelektronenmicroscopiebeeld toont de morfologie van de heldere en ronde vesikels van C2C12-afgeleide exosomen. Westerse blot analyse bevestigt de aanwezigheid van exosoom markers waaronder CD63 en Tsg101.Een doorschijnend oedeemgebied wordt waargenomen na de intramyocardiale injectie in de voorste wand van de linkerventrikel van mdx-muizen, wat aangeeft dat de injectie in het myocardium succesvol is. Immunofluorescente kleuring voor dystrofine wordt vervolgens uitgevoerd om te bepalen of cardiale MPC-Exo levering herstelt dystrofisch-eiwit expressie in mdx harten. Gedeeltelijke restauratie van dystrofie expressie wordt waargenomen met membraan lokalisatie in sommige van cardiomyocyten.De hartfunctie wordt gemeten door echocardiografie twee dagen na intramyocardiale levering om te bepalen of transplatie van MPC-Exo de hartfunctie bij mdx-muizen verbetert. De MPC-Exo behandeling wordt gezien ter verbetering van de binnenwand beweging in vergelijking met PBS, wat suggereert dat MPC-Exo transplantatie verbeterde cardiale functie in mdx muizen. Met behulp van een 31 meter insulinenaald met de tip op ongeveer 20 graden is van cruciaal belang voor het succes voor de levering van de meeste exosomen in het myocardium en maximaliseert de blootstelling van geïnjecteerde exosomen om cardiomyocyten host.Na de procedure kan dystrofineexpressie worden gedetecteerd na MPC-afgeleide exosoominjectie door immunofluorescente vlekken die erop wijzen dat cardiale MPC-afgeleide exosoomlevering dystrofisch-eiwitexpressie herstelt. Deze techniek zal de weg vrijmaken voor cardiomyopathie behandeling met exosoom-dragende therapeutische genetische materialen, waaronder DNA, mRNA, lncRNA, of microRNAs.
