Het endotheel wordt blootgesteld aan talrijke proatherogene factoren, waaronder lipiden, ontstekingsmediatoren en vloeibare shear stress1,11,12. Directe observatie van endotheelcellen in situ biedt de speciale voordelen voor het analyseren van veranderingen in celmorfologie, intercellulaire kruispunten en molecuul expressie patronen in reactie op de letsel stimuli.
Eerdere studies hebben twee verschil...

De vroege veranderingen in de vasculatuur beginnen voornamelijk in het endotheel, dat fungeert als een selectieve barrière tussen het bloed en de wand van het vat met zijn intercellulaire strakke junctieve complexen1. Substantieel bewijs wijst op een cruciale rol voor de mechanische effecten van de bloedstroom in modulerende endotheliale functie2. Vloeibare shear stress, een wrijvingskracht gegenereerd door de doorbloeding, differentieel vormen endotheliale celmorfologie en functie, afhankelijk van de specifieke stroom paradigma's op verschillende vasculaire locaties2,3. Atherosclerotische laesies komen bij voorkeur op plaatsen van verstoorde bloedstroom (d-flow), zoals krommingen van het vat, stroom afscheiders en vertakkingspunten, in vergelijking met regio's van steady flow (s-flow), zoals het rechte segment van de slagader. Daarom moet directe observatie van endotheliale morfologie en molecuul expressie patronen belangrijke inzichten geven in de structurele en functionele fenotypes van endotheliale cellen in verschillende stromings paradigma's.
Gekweekte endotheelcellen mogen het eigenlijke fenotype niet uitdrukken zoals in vivo, deels als gevolg van het verlies van de impact van vloeibare shear stress, omringende cytokines en celcelcellen of celextracellulaire matrix interacties. Om dit te helpen, kan de intacte endotheliale celmonolayer worden bestudeerd op dwarsdoorsneden met behulp van klassieke immunohistochemie. Echter, de endotheliale monolaag is zo dun en kwetsbaar dat het meestal niet duidelijk kan worden waargenomen. En face immunohistochemie is gebruikt om het inwendige oppervlak van het endotheel te observeren, maar is gecompliceerd of onregelmatig in de resultaten omdat het endotheel gemakkelijk uit het onderliggende weefsel kan worden ontdaan, of slechts een deel van de arteriële wand van ratten of konijnen, waarvan de wanden dik zijn, is4,5gemonteerd.
Muismodellen hebben in veel opzichten aanzienlijke voordelen ten opzichte van andere dieren. Hier gebruiken we een gemodificeerde en gezicht immunofluorescentie techniek om endotheliale cellen van de aortaboog en thoracale aortin C57BL/6 muis te analyseren. Een dergelijke techniek is op grote schaal gebruikt voor het bestuderen van de endotheliale pathofysiologie in verschillende stromingspatronen en in de ontwikkeling van atherosclerose6,7,8,9,10. Deze methode stelt wetenschappers in staat om het hele oppervlak van het endotheel duidelijk te observeren en om de expressie patronen van een gegeven eiwit te vergelijken in gebieden onder verschillende vocht afschuiving stress.

Afwijkende veranderingen in endotheliale fenotype en morfologie worden beschouwd als initiële gebeurtenissen in de pathogenese van atherosclerose. Directe observatie van het intacte endotheel zal waardevolle informatie bieden voor het begrijpen van de cellulaire en moleculaire gebeurtenissen in de dysfunctionele endotheel cellen. Hier beschrijven we een gemodificeerde en gezicht immunofluorescentie kleuringstechniek die wetenschappers in staat stelt om duidelijke beelden van het intact endotheliale oppervlak te verkrijgen en de molecuul expressie patronen in situ te analyseren. De methode is eenvoudig en betrouwbaar voor het observeren van de gehele endotheliale monolaag op verschillende locaties van de aorta. Deze techniek kan een veelbelovend instrument zijn voor het begrijpen van de pathofysiologie van atherosclerose, vooral in een vroeg stadium.

Een 12-weekse C57BL/6-muis werd geëerd en geperfoniseerd met een normale zoutoplossing die 40 eenheden/mL heparine en vervolgens voorgekoelde 4% Paraformaldehyde bevat. Using En Face Immunofluorescence Staining to Observe Vascular Endothelial Cells Directly

Hier presenteren we een protocol voor immunofluorescentie kleuring om de endotheliale cellen van de muis aorta direct te observeren. Deze techniek is nuttig bij het bestuderen van het cellulaire en moleculaire fenotype van endotheel cellen in verschillende stromingspatronen en in de ontwikkeling van atherosclerose.

Dit protocol helpt mensen bij het analyseren van endotheelcelmorfologie, en de expressie van moleculen in regio's, onder verschillende vochtschuifstress. Het belangrijkste voordeel van deze techniek is dat het een directe beoordeling van vasculaire endotheelcellen onder verschillende vochtschaar stress biedt. Bij het uitvoeren van deze procedure zijn de belangrijkste stappen het gebruik van vers bereide paraformaldehyde en het uitvoeren van een goede perfusie.Begin deze procedure met verdovendering van een 12 weken oude C57BL/6 muis, zoals beschreven in het tekstprotocol. Bevestig de juiste verdovendering door zachtjes knijpen de staart. Plak de poten van de muis vast aan een stapel papieren handdoeken, met plakband.Houd de huid van de muis met tangen omhoog en snijd de huid met een schaar van de buik naar de bovenkant van de thorax. Open de buikholte, onder de ribbenkast, met een scherpe schaar. Til het borstbeen op met tangen en snijd het middenrif door.Snijd vervolgens de ribbenkast weg om de borstholte bloot te leggen. Snijd de vena cava net boven de lever af met een schaar. Druk doorsneed de arteriële boom gedurende vijf minuten, met voorgekoelde normale zoutoplossing met 40 eenheden per milliliter heparine, door de linker ventrikel, totdat de longen en lever bleek worden.Vervolgens doornemen met voorgekoelde 4%paraformaldehyde in PBS gedurende vier minuten. Verwijder alle spieren, organen en vet, totdat de aorta is blootgesteld. Plaats de muis onder een ontledenmicroscoop.Gebruik delicate tangen en een delicate schaar om de aorta duidelijk bloot te leggen onder een ontledenmicroscoop en verwijder het bindweefsel langs de aorta, zo schoon mogelijk. Ontleden de thoracale aorta van het hart naar de celeliakie met een delicate schaar. Plaats de aorta in een zes centimeter cel cultuur schotel, met PBS.Snijd de aorta langs de lange weg open, eerst langs de mindere curve, en gebruik vervolgens de microscissors om de drie takken van de aortaboog open te snijden, inclusief de innominaat, linker gemeenschappelijke halsslagader, en de linker subclavian slagader langs de grotere curve totdat het rechte segment is voldaan. Permeabiliseren van de aorta met 1%polyoxyethyleen octyl fenyl ether, in PBS gedurende 10 minuten. Blokkeer vervolgens de aorta met 10% normaal geitenserum in tris-gebufferde zoutoplossing met 2,5% polysorbaat 20 gedurende een uur bij kamertemperatuur, in een 12 putcelkweekplaat.Volgende incubeer de aorta in de blokkerende buffer met vijf gram per milliliter konijn anti-VCAM1, en vijf gram per milliliter rat anti-VE-Cadherin, 's nachts op vier graden Celsius. Na het spoelen van het monster drie keer met wasoplossing, breng de fluorescentie geconjugeerde secundaire antilichamen gedurende een uur bij kamertemperatuur, houden op een donkere plaats. Spoel nog eens drie keer in de wasoplossing.Verdort de aorta met DAPI gedurende 10 minuten, op een donkere plaats. Spoel vervolgens de bevlekte aorta drie keer in wasoplossing. Plaats de aorta op een afdeksel met het lumenoppervlak naar beneden, en beweeg het langzaam naar antifade montage-oplossing die eerder is gedaald op de cover slip.Strek de aorta voorzichtig uit om het exemplaar plat te houden. Keer de afdekstrook om en leg deze op de glazen schuif. Zorg ervoor dat er geen luchtbellen tussen het monster en het glas.Bestudeer de aorta met een laserscanning confocale microscoop. Analyseer kleurintensiteiten van verschillende kanalen uit de gewenste regio in de en face-afbeeldingen, met Image-Pro Plus-software. En gezicht immunofluorescentie vlekken beelden van vasculaire endotheelcellen uit de muis aorta worden getoond.En gezicht immunofluorescentie van de vasculaire cel hechting eiwit een, en expressie met VE-Cadherin als een endotheel marker, worden getoond onder verschillende stroompatronen uit verschillende regio's van de muis aorta. DAPI werd ook tegengewerkt om de celkernen voor betere visualisatie te tonen. Wanneer u door de Z-stapels onder de microscoop kijkt, kunnen de endotheel en gladde spiercellen gemakkelijk worden onderscheiden door hun celkernenmorfologie.De endotheelcelkernen zijn ovaal gevormd en groter dan de spindelvormige, gladde spiercelkernen. Expressie van VCAM-1 was overvloediger in gebieden onder verstoorde stroom, bij mindere kromming van de aortaboog, vergeleken met gebieden onder gestage stroom, bij een grotere kromming van de aortaboog, en bij de thoracale aorta. Hier worden gebieden met een kleinere kromming van een muisaorta aangeduid als d-flow gebieden, en de grotere kromming en thoracale.Met de juiste modificatie kan deze techniek ook worden toegepast op de analyse van vasculaire doorlaatbaarheid en de positie van macromoleculen in de endotheelcel intima.
