血管脂肪组织环绕血管，调节心血管生理学。该协议用于回答与人类脂肪祖祖在血管微环境中的功能有关的关键问题。脂肪祖细胞根据解剖位置而变化。我们表明，多能祖细胞的种群可以从心血管疾病患者的血管周围脂肪组织中成功获得。证明这个程序将是斯宾塞斯科特，一个医科学生从我的实验室。获得500毫克的人类血管脂肪组织或PVAT从手术室，如文本协议中所述。将新鲜的人类PVAT从DMEM转移到含有25毫升抗生素溶液的50毫升锥形管中。在4摄氏度下用摇摆孵育20分钟。当PVAT在抗生素溶液中时，在37摄氏度下解冻一个等分液缓冲液。将 50 微升 100 倍抗生素/抗神秘药溶液加入 5 毫升分离缓冲液中，并使用 0.22 微米注射器过滤器进行消毒。将一毫升明胶溶液加入24井板的一个井中。在层流罩中，使用无菌钳子和剪刀将PVAT从抗生素溶液转移到无菌培养皿。向组织中加入一毫升预热分离缓冲液。使用无菌钳子和解剖剪刀将整个组织细细地切碎成泥浆。将一毫升浆料转移到四毫升分离缓冲液中。在200转/时预热37摄氏度的轨道摇床中孵育其侧面的管子一小时。一小时后，不应存在可见的组织片段，溶液将显示为多云细胞悬浮液。通过 70 微米电池过滤器在 50 毫升锥形管上过滤溶液。用额外的10毫升抗生素溶液冲洗过滤器，以捕获尽可能多的细胞。不要挤压过滤器。现在，在摆动桶离心机中，以 300 倍 g 的分粒 12 分钟。离心后，管子将被分离成脂肪顶层的脂肪脂肪细胞，相间和颗粒。颗粒是含有内皮细胞、免疫细胞、血细胞和祖细胞的频闪血管部分。将颗粒在 10 毫升 HBS 中重新浓缩，以 300 倍 g 的离心机进行 5 分钟。重复此步骤，在 HBSS 中总共洗两次。现在从24井板中吸出明胶。用HSSS轻轻清洗一次井，以去除未绑定的明胶。将具有完整红血球的血管分馏粒重新在生长介质的一毫升中，并播种到明胶涂层的井上。然后将人类FGF2加入培养培养剂中每毫升25毫微克的最终浓度。在37摄氏度下孵育24小时，二氧化碳为5%。生长细胞24小时后，去除生长介质，用HSS洗涤细胞五次。这个洗涤步骤去除红血球和死细胞。然后添加一毫升的新鲜生长介质，每毫升FGF2补充25毫微克到每井。每 48 小时更换一次介质，确保每次补充 25 毫微克每毫升新鲜 FGF2。通过细胞一旦达到100%汇合7至10天后，移植。为此，吸气生长介质，用一毫升 HBSS 洗涤单层两次。从井中吸出所有HBSS，并加入几滴细胞分离溶液。点击并旋转板几次，并在37摄氏度下孵育5%CO2，5至7分钟，以提升细胞。然后向分离的细胞添加大约一毫升的新鲜培养培养。将分离细胞的500微升分配到两个24孔板的井中，每个孔包含500微升的生长介质和25纳米的FGF2。此外，培养人类骨髓中质干细胞或MSC菌落，如文本协议所述。将12井板的井中适当数量的骨髓和PVAT衍生细胞进行板。对于致生和成骨条件，每井约有20万至22.5万个细胞。而对于软体条件，每井有15万到17.5万个细胞。然后通过添加细胞分离溶液，将细胞与人类PVAT祖细胞种群和人类骨髓MSC群体分离。在37摄氏度和5%CO2下孵育分离溶液中的细胞5分钟。将种群拉入单独的 15 毫升圆锥瓶中。将小瓶以500倍g旋转7分钟，使细胞产生颗粒。然后将细胞重新在一毫升PBS中，并使用血细胞计估计细胞数。和以前一样，在 12 个井的盘子中镀上细胞。为诱导和非诱导性致畸和骨质条件提供单独的菜肴。并在诱导条件的每个井中加入1.5毫升的致生和骨质传导介质。然后添加1.5毫升的致生和骨质非诱导介质到每个井的非诱导条件。在37摄氏度和5%CO2下开始孵育致性、骨质诱导细胞和非诱导细胞群。以500倍g将剩余的人类PVAT祖细胞和人类骨髓MSCs旋转7分钟。确定重新捐献剩余的骨髓和PVAT衍生细胞颗粒所需的体积，以达到每10微升10万细胞的密度。然后重新暂停在MSC生长介质的计算体积中的颗粒，用于软骨基因血统诱导。使用移液器轻轻移动细胞的体积，以确保均匀分布。现在移液10微升的浓缩细胞溶液进入每井的中心，形成10万个细胞的微质量。将一毫升无菌水放在相邻的井中，以防止蒸发。在37摄氏度和5%CO2下孵育微质量培养两小时，使微质量聚集。两小时后，仔细添加每毫升10毫微克的夹心性分化介质，每个诱导条件井中加入10毫微克。现在，在非诱导条件井中小心地添加 1.5 毫升的非感应介质。将诱导成软杂条件中的微质量刮入盒式磁带，以便像文本协议中描述的那样使样品脱水、嵌入和染色。致生分化研究与人类骨髓衍生的MSC和PVAT衍生的祖细胞同时进行。在非诱导条件下，无脂积明显。这与油红O中中性脂质染色后所表现出的诱导条件形成鲜明对比，虽然人类大脂PVET衍生细胞的分化程度更为强健，但两个人类细胞源都显示出分化致发性血统的能力。骨源分化协议用于人类骨髓衍生的MSCs和PVET衍生细胞。非诱导细胞没有染色与阿里扎林红。在骨质分化协议之后，人类MSC开发出钙化结节，与阿里扎林红染色，而人类大动脉PVET细胞则没有。来自人类骨髓MSC和人类PVAT的细胞表现出软骨分化的特点，在微质量中大量积累胶原蛋白。微质量是由人类骨髓MSC和主要PVET衍生细胞形成的，也表现出丰富的糖醇的积累，如阿尔西安蓝染色所示。从形态学上看，与乳牙相似的结构被检测出，细胞被胶原蛋白沉积包围。在酶分离之前对血管周围脂肪组织进行精细的切碎，并确保每个血统分化测定都镀上适当的细胞密度，这一点至关重要。遵循这个程序，定量PCR结合西方印迹和流细胞学应使用特征的血统特异性标记的分化祖体从近血管脂肪和骨髓来源。通过这项技术，我们可以开始了解肥胖期间调节血管脂肪组织扩张和功能障碍的机制，以及祖细胞对血管功能和心血管疾病的影响。
