Rabies virus (RABV) er en negativ følelse enkelt strandede lyssavirus, og cirkulerer blandt forskellige dyreliv reservoir arter inden for ordrerne Carnivora og Chiroptera. Flere arter af Mongoose er reservoirer af RABV, og den lille indiske Mongoose (Herpestes auropunktatus) er det eneste reservoir i Puerto Rico og andre caribiske øer i den vestlige halvkugle1,2,3 . Kontrollen af RABV cirkulation i dyreliv reservoirer er typisk opnås gennem en strategi for oral rabiesvaccination (ORV). I USA (USA) koordineres denne ledelses aktivitet af USDA/APHIS/Wildlife Services national rabies Management program (NRMP)4. I øjeblikket ingen Wildlife ORV program findes i Puerto Rico. Rhodamine B er blevet evalueret som en markør i en række forskellige væv og kan påvises ved hjælp af ultraviolet (UV) lys og fluorescens i hår og whiskers10,21.
Iophenoxic Acid (IPA) er et hvidt, krystallinsk pulver, der er blevet brugt til at evaluere indtagelse af agn i coyoter (Canis latrans)22, Arctic Fox (Vulpes lagopus)23, rød ræv (Vulpes vulpes)24, vaskebjørn 9 , 25, vildsvin14, kronhjorte (Cervus elaphus scoticus)26, European badgers12 og fritter (M. Furo)27, blandt flere andre pattedyrarter. Opbevaringstider for IPA varierer efter art fra mindre end to uger i nogle pungdyr28,29, til mindst 26 uger i hovdyr26 og over 52 uger i tamhunde (Canis lupus familiaris)30. Opbevaringstider kan også være dosisafhængige31. Iophenoxinsyre binder kraftigt til serumalbumin og blev historisk påvist ved måling af blodjod-niveauer32. Denne indirekte tilgang blev fortrængt af højtydende væskekromatografi (HPLC)-metoder til direkte måling af iophenoxinsyre koncentrationer med UV-detektion33, og til sidst med væskekromatografi og massespektrometri (LCMS) 34af35. Til denne undersøgelse, en meget følsom og selektiv væskekromatografi med tandem massespektrometri (LC-MS/MS) metode blev udviklet, der udnytter multiple reaktion overvågning (MRM) at kvantificere to analoger af iophenoxic syre. Vores mål var at bruge denne LC-MS/MS metode til at evaluere mærkning evne 2-(3-hydroxy-2, 4, 6-triiodobenzyl) propanoinsyre (methyl-IPA eller Me-IPA) og 2-(3-hydroxy-2, 4, 6-triiodobenzyl) butanoinsyre (ethyl-IPA eller et-IPA) og når de leveres i en ORV agn til fangenskab mongooses.
Mongooses blev levende fanget i bur fælder loget med kommercielt tilgængelige røget pølser og fiskeolie. Mongooses blev anbragt i individuelle 60 cm x 60 cm x 40 cm rustfrit stål bure og fodret en daglig ration af ~ 50 g kommerciel tør kattefoder, suppleret to gange om ugen med en kommercielt tilgængelig kylling lår. Vand var til rådighed ad libitum. Vi leverede to derivater af IPA, ethyl-IPA og methyl-IPA, til bundne mongooses i placebo ORV lokoer. Alle lokkemad var sammensat af en 28 mm x 20 mm x 9 mm folie blisterpakning med en ekstern belægning (herefter "Bait matrix"), der indeholder pulveriseret hønseæg og gelatine (tabel over materialer). Lokkemad indeholdt 0,7 mL vand eller IPA-derivat og vejede ca. 3 g, hvoraf ~ 2 g var den udvendige agn matrix.
Vi tilbød 16 captive mongooses et-IPA i tre koncentrationer: 0,14% (2,8 mg et-IPA i ~ 2 g agn matrix; 3 hanner [m], 3 hunner [f]), 0,4% (2,8 mg et-IPA i 0,7 mL blisterpakning volumen; 3m, 3f), og 1,0% (7,0 mg ethyl-IPA i 0,7 mL blisterpakning volumen; 2m , 2f). Den samlede dosis på 2,8 mg svarer til en dosishastighed på 5 mg/kg27,36 og er baseret på en gennemsnitlig Mongoose vægt på 560 g i Puerto Rico. Vi valgte 1% som den højeste koncentration som forskning tyder smag aversion til nogle biomarkører kan forekomme i koncentrationer > 1% i nogle arter37. Vi tilbød kun 1% dosis i blisterpakningen, da flokkulering forhindrede den opløftning i at opløses i opløsningsmidlet tilstrækkeligt til at blive jævnt indarbejdet i agn matrixen. En kontrolgruppe (2m, 1f) modtog lokkemad fyldt med sterilt vand og ingen IPA. Vi tilbød lokoer til mongooses om morgenen (~ 8 a.m.) under eller før fodring af deres daglige vedligeholdelse ration. Agn rester blev fjernet efter ca. 24 timer. Vi indsamlede blodprøver før behandling, en dag efter behandling og derefter ugentligt op til 8 uger efter behandling. Vi bedøvet monokser ved indånding af isoflurangas og opsamlet op til 1,0 mL fuldblod ved venepunktur af kraniel Vena cava som beskrevet for fritter38. Vi centrifugerede hele blodprøver, overførte sera til cryovials og opbevarede dem ved-80 °C indtil analyse. Ikke alle dyr blev udtaget i alle perioder for at minimere virkningerne af gentagen blodprøvetagning på dyrenes sundhed. Kontroldyrene blev udtaget på dag 0 og derefter ugentligt i op til 8 uger efter behandlingen.
Vi leverede mig-IPA i tre koncentrationer: 0,035% (0,7 mg), 0,07% (1,4 mg) og 0,14% (2,8 mg), alle indarbejdet i agn matrix, med 2 hanner og 2 kvinder pr. behandlingsgruppe. To mænd og to hunner fik loits fyldt med sterilt vand og ingen IPA. Agn tilbyder tider og Mongoose anæstesi er beskrevet ovenfor. Vi indsamlede blodprøver før behandling på dag 1, og derefter ugentligt op til 4 uger efter behandling.
Vi testede serum koncentrationsdata for normalitet og anslåede midler for serum IPA koncentrationer af forskellige behandlingsgrupper. Vi brugte en lineær blandet model til at sammenligne gennemsnitlige serum et-IPA koncentrationer samlet på tværs af individer. Agn type (matrix/blisterpakning) var en fast effekt i tillæg til eksperimentel dag, hvorimod Animal ID var en tilfældig effekt. Alle procedurer blev kørt ved hjælp af fælles statistisk software (tabel over materialer) og signifikans blev evalueret ved α = 0,05.

Den lille indiske Mongoose (Herpestes auropunktatus) er et reservoir af rabiesvirus (rabv) i Puerto Rico og omfatter over 70% af dyr rabiestilfælde rapporteret årligt. Kontrollen af RABV cirkulation i dyreliv reservoirer er typisk opnås ved en strategi for oral rabiesvaccination (ORV). I øjeblikket ingen Wildlife ORV program findes i Puerto Rico. Forskning i orale rabiesvacciner og forskellige agn typer for mongooses er blevet gennemført med lovende resultater. Vi tilbød også 12 bundne mongooses ORV lokkemad indeholdende methyl-iophenoxic Acid (Me-IPA) i koncentrationer på 0,035%, 0,07% og 0,14% i den eksterne agn matrix. Vi indsamlede en serum prøve før agn tilbud og derefter ugentligt i op til otte uger post tilbud. Vi ekstraherede Iophenoxic syrer fra sera til acetonitril og kvantificerede ved hjælp af væskekromatografi/massespektrometri. Vi analyserede sera for et-IPA eller Me-IPA ved væskekromatografi-massespektrometri. Vi fandt passende mærkning evne til mindst otte og fire uger for et-og mig-IPA, hhv. Begge IPA-derivater kunne være egnede til felt evaluering af ORV agn optagelse i mongooses. På grund af den lang levetid af mærket i Mongoose sera skal man være forsigtig med ikke at tolerere resultater ved at bruge det samme IPA-derivat i de efterfølgende evalueringer.

Analysis of Iophenoxic Acid Analogues in Small Indian Mongoose Herpestes Auropunctatus Sera for Use as an Oral Rabies Vaccination Biological Marker

Vi skitserede en procedure til påvisning og kvantificering af iophenoxinsyre i desmerdyr serum. Resultaterne tyder på, at iophenoxinsyre kan anvendes som en biologisk markør for at kontrollere betaoptagelse i arten. Den største fordel ved denne metode er, at det giver mulighed for kvantificering af iophenoxic syre analoger på dele per milliard niveauer, ved hjælp af en enkel at udføre protein nedbør teknik.Til mobil fase A kombineres en milliliter myresyre med en liter ultra rent vand. For mobil fase B, kombinere en milliliter myresyre med en liter acetonitril. For fortyndingsmiddel, kombinere en milliliter trifluoretsyre med 200 milliliter acetonitrile.Dernæst vejer ca. 10 milligram methyl-IPA på microbalance, og registrere massen til plus eller minus 0001 milligram. Methyl-IPA overføres kvantitativt til en målekolbe i 10 milliliter A ved hjælp af fire til fem milliliter acetonitril. Sonikere i et minut for at opløse alle faste stoffer og derefter bringe til volumen med acetonitril.Der overføres ca. otte milliliter af hver bestand til otte milliliter ravglasglas med PTFE-forede hætter, og prøverne opbevares ved stuetemperatur. Derefter overføres lageret til et 8 milliliter ravfarvet glashætte med en PTFE-foret hætte og opbevares ved stuetemperatur. Den resterende opløsning overføres i rør A til farligt affald, og det tomme rør smides ud i en beholder til biofarligt affald. Fjern nu acetononitrile og trifluoroacetic fra rør B med en blid strøm af nitrogengas i et 45 grader Celsius vandbad.Tilsæt 0,25 milliliter acetonitril til rør B ved hjælp af en gentagelse pipettor og vortex mix i fire til fem sekunder. Derefter centrifuge ved 1200 gange G i to til fire sekunder for at indsamle væsken i bunden af røret.
