De LC-MS/MS-methode die voor de studies werd ontwikkeld, gebruikte de selectiviteit van meervoudige reactie monitoring om me-IPA en et-IPA in het Mongoose serum nauwkeurig te kwantificeren. Rabies virus (RABV) is een negatief gevoel enkel strandde Lyssavirus, en circuleert tussen diverse wild reservoir soorten binnen de bestellingen Carnivora en Chiroptera. Meerdere soorten mangoesten zijn reservoirs van RABV, en de kleine Indische mangoest (Herpestes auropunctatus) is het enige stuwmeer in Puerto Rico en andere Caribische eilanden in het westelijk halfrond1,2,3 . De controle van RABV circulatie in wild reservoirs wordt meestal bewerkstelligd door een strategie van orale rabiësvaccinatie (ORV). In de Verenigde Staten (VS) wordt deze beheeractiviteit gecoördineerd door het USDA/APHIS-en Wildlife Services National rabies Management Program (NRMP)4. Momenteel bestaat er geen Wildlife ORV programma in Puerto Rico. Onderzoek naar rabiës vaccins en verschillende Bait types voor mangoest is uitgevoerd met veelbelovende resultaten die suggereren dat een orv-programma voor mangoest mogelijk is5,6,7,8.
Het monitoren van de impact van ORV berust op het inschatten van de opname van aas door DOELDIERSOORTEN, wat meestal gepaard gaat met het evalueren van een verandering in RV antilichamen seroprevalentie. Echter, deze strategie kan uitdagend zijn in gebieden met een actieve Wildlife orv Programma's of in gebieden waar RV is enzoötische boviene en achtergrondniveaus van rabv neutraliserende antilichamen (rvna) aanwezig zijn in het reservoir soorten. In dergelijke situaties kan een biomarker die is opgenomen in het aas of de externe Bait matrix nuttig zijn.
Verschillende biologische markers zijn gebruikt voor het monitoren van de opname van aas bij tal van soorten, waaronder wasberen (Procyon lotor)9,10, stoats (Mustela Hermelijn)11,12, Europese Badgers ( Meles meles) 13, wilde zwijnen (Sus scrofa)14, kleine Indische mangoest15 en prairie honden (cynomysludovicianus)16,17, onder anderen. In de VS bevatten operationele orv-lokaas vaak een 1% tetracycline biomarker in de Bait matrix om aas-opname18,19te bewaken. Nadelen van het gebruik van tetracycline omvatten echter een toenemende bezorgdheid over de verspreiding van antibiotica in het milieu en dat de detectie van tetracycline typisch invasief is, waarbij tandextractie of vernietiging van het dier nodig is om bot te verkrijgen voorbeelden20. Rhodamine B is geëvalueerd als een marker in een verscheidenheid van weefsels en kan worden gedetecteerd met behulp van ultraviolet (UV) licht en fluorescentie in haar en snorharen10,21.
Iophenoxinezuur (IPA) is een wit kristallijn poeder dat is gebruikt om de consumptie van aas in coyotes te evalueren(Canis latrans)22, Arctic Fox (Vulpes lagopus)23, Red Fox (Vulpes vulpes)24, wasberen 9 , 25, everzwijn14, edelhert (Cervus elaphus scoticus)26, Europese Badgers12 en fretten (M. furo)27, onder verschillende andere zoogdiersoorten. Retentietijden van IPA varieert per soort van minder dan twee weken in sommige buideldieren28,29, tot ten minste 26 weken in hoefdieren26 en meer dan 52 weken in huis honden (Canis lupus familiaris)30. Retentietijden kunnen ook dosis-afhankelijke31zijn. Iophenoxinezuur bindt sterk aan serumalbumine en werd historisch ontdekt door het meten van bloed jodium niveaus32. Deze indirecte aanpak werd vervangen door krachtige vloeistofchromatografie (HPLC)-methoden om de iophenoxinezuur concentraties direct te meten met UV-detectie33, en uiteindelijk met vloeibare chromatografie en massaspectrometrie (lcm's) 34,35. Voor deze studie werd een zeer gevoelige en selectieve vloeistofchromatografie met tandem massaspectrometrie (LC-MS/MS)-methode ontwikkeld die meerdere reactie monitoring (MRM) gebruikt om twee analogen van iopfenoxinezuur te kwantificeren. Ons doel was om deze LC-MS/MS-methode te gebruiken om het markerings vermogen van 2-(3-hydroxy-2, 4, 6-triiodobenzyl) propanoïne zuur (methyl-IPA of me-IPA) en 2-(3-hydroxy-2, 4, 6-triiodobenzyl) butaanzuur (ethyl-IPA of et-IPA) en bij levering in een ORV aas naar Captive mangoest.
Mangoest waren levend gevangen in kooi vallen met in de handel verkrijgbare gerookte worstjes en visolie. Mangoest werden ondergebracht in individuele 60 cm x 60 cm x 40 cm roestvrijstalen kooien en gevoed met een dagelijks rantsoen van ~ 50 g commercieel droog kattenvoer, tweemaal per week aangevuld met een in de handel verkrijgbare kippen dij. Water was beschikbaar ad libitum. We leverden twee derivaten van IPA, ethyl-IPA en methyl-IPA, aan gevangen mangoest in placebo ORV Baits. Alle aas bestond uit een 28 mm x 20 mm x 9 mm folie blisterverpakking met een uitwendige coating (hierna "Bait matrix") met poedervormige kip ei en gelatine (tabel van materialen). Baits bevatte 0,7 mL water of IPA-derivaat en woog ongeveer 3 g, waarvan ~ 2 g de externe Bait matrix was.
We boden 16 Captive mangoest et-IPA in drie concentraties aan: 0,14% (2,8 mg et-IPA in ~ 2 g Bait matrix; 3 mannetjes [m], 3 vrouwtjes [f]), 0,4% (2,8 mg et-IPA in 0,7 mL blisterverpakking volume; 3m, 3F), en 1,0% (7,0 mg ethyl-IPA in 0,7 mL blisterverpakking volume; 2m , 2F). De totale dosis van 2,8 mg komt overeen met een doserings percentage van 5 mg/kg27,36 en is gebaseerd op een gemiddeld mongans gewicht van 560 g in Puerto Rico. We selecteerden 1% als de hoogste concentratie als onderzoek suggereert dat de smaak afaversie van sommige biomarkers kan voorkomen bij concentraties > 1% bij sommige soorten37. We boden de dosis van 1% alleen aan in de blisterverpakking, omdat flocculatie verhinderde dat de opgeloste stof in het oplosmiddel voldoende werd opgenomen om gelijkmatig in de Bait matrix te worden verwerkt. Eén controlegroep (2m, 1f) kreeg aas gevuld met steriel water en geen IPA. We boden loten aan mangoest in de ochtend (~ 8 a.m.) tijdens of voorafgaand aan het voeden van hun dagelijkse onderhoud rantsoen. Aas resten werden verwijderd na ongeveer 24 uur. We verzamelde bloedmonsters voorafgaand aan de behandeling, een dag na de behandeling en vervolgens wekelijks tot 8 weken na de behandeling. We verdoofd mangoest door inademing van Isofluraan gas en verzameld tot 1,0 ml volbloed door venipunctie van de craniale Vena Cava zoals beschreven voor fretten38. We hebben volbloed monsters gecentrifugeerd, sera overgebracht naar cryoflacons en ze opgeslagen bij-80 °C tot de analyse. Niet alle dieren werden bemonsterd gedurende alle perioden om de effecten van herhaalde bloed trekkingen op de gezondheid van de dieren te minimaliseren. Controledieren werden bemonsterd op dag 0, daarna wekelijks gedurende maximaal 8 weken na de behandeling.
We leverden me-IPA in drie concentraties: 0,035% (0,7 mg), 0,07% (1,4 mg) en 0,14% (2,8 mg), allemaal opgenomen in de Bait matrix, met 2 mannetjes en 2 vrouwtjes per behandelgroep. Twee mannetjes en twee vrouwtjes kregen aas gevuld met steriel water en geen IPA. Aas bieden tijden en Mongoose anesthesie worden hierboven beschreven. We verzamelde bloedmonsters voorafgaand aan de behandeling op dag 1, en vervolgens wekelijks tot 4 weken na de behandeling.
We testten de serumconcentratie gegevens voor normaliteit en geschatte gemiddelden voor serum IPA-concentraties van verschillende behandelingsgroepen. We gebruikten een lineair gemengd model om de gemiddelde serum-et-IPA-concentraties te vergelijken die over individuen werden samengevoegd. Bait type (matrix/blisterverpakking) was een vast effect naast de experimentele dag, terwijl dierlijke ID een willekeurig effect was. Alle procedures werden uitgevoerd met behulp van gemeenschappelijke statistische software (tabel met materialen) en significantie werd geëvalueerd bij α = 0,05.

De kleine Indische mangoest (Herpestes auropunctatus) is een reservoir van rabiësvirus (rabv) in Puerto Rico en omvat meer dan 70% van dieren rabiës gevallen gerapporteerd per jaar. De controle van RABV circulatie in wild reservoirs wordt meestal bewerkstelligd door een strategie van orale rabiësvaccinatie (ORV). Momenteel bestaat er geen Wildlife ORV programma in Puerto Rico. Onderzoek naar orale rabiës vaccins en verschillende Bait types voor mangoest is uitgevoerd met veelbelovende resultaten. Het monitoren van het succes van ORV berust op het inschatten van de opname van aas door DOELDIERSOORTEN, wat meestal gepaard gaat met het evalueren van een verandering in RABV neutraliserende antilichamen (RVNA) na vaccinatie. Deze strategie kan moeilijk te interpreteren zijn in gebieden met een actief Wildlife orv-programma of in gebieden waar rabv enzoötische boviene is en achtergrondniveaus van rvna aanwezig zijn in reservoir soorten. In dergelijke situaties kan een biomarker die met het vaccin of de Bait matrix is verwerkt, nuttig zijn. We boden 16 gevangen mangoest placebo orv lokaas aan die ethyl-iopfenoxinezuur (et-IPA) bevatten in concentraties van 0,4% en 1% in het aas en 0,14% in de externe Bait matrix. We boden ook 12 gevangen mangoest orv lokaas aan die methyl-iopfenoxinezuur (me-IPA) bevatten in concentraties van 0,035%, 0,07% en 0,14% in de externe Bait matrix. We hebben een serummonster verzameld voorafgaand aan het aas en vervolgens wekelijks voor maximaal acht weken na het aanbieden. We hebben Iopfenoxische zuren uit sera geëxtraheerd in acetonitril en gekwantificeerd met behulp van vloeibare chromatografie/massaspectrometrie. We analyseerden sera voor et-IPA of me-IPA door vloeistofchromatografie-massaspectrometrie. We vonden adequate Markeer baarheid voor ten minste acht en vier weken voor et-en me-IPA, respectievelijk. Beide IPA-derivaten kunnen geschikt zijn voor veld evaluatie van de opname van ORV Bait in mangoest. Vanwege de levensduur van de marker in Mongoose sera, moet er voor gezorgd worden dat de resultaten niet worden verhaald door hetzelfde IPA-derivaat te gebruiken tijdens opeenvolgende evaluaties.

Analysis of Iophenoxic Acid Analogues in Small Indian Mongoose Herpestes Auropunctatus Sera for Use as an Oral Rabies Vaccination Biological Marker

We schetsten een procedure om iophenoxïnezuur in mongoose serum op te sporen en te kwantificeren. De resultaten suggereren dat iophenoxïnezuur kan worden gebruikt als een biologische marker om de bèta-opname bij de soort te controleren. Het belangrijkste voordeel van deze methode is dat het mogelijk maakt voor de kwantificering van iophenoxïnezuur analogen op delen per miljard niveaus, met behulp van een eenvoudig uit te voeren eiwit neerslag techniek.Voor mobiele fase A, combineer een milliliter mierenzuur met een liter ultra zuiver water. Voor mobiele fase B, combineer een milliliter mierenzuur met een liter acetonitril. Voor het verdunningsmiddel, combineer een milliliter trifluoroacetisch zuur met 200 milliliter acetonitril.Vervolgens weegt u ongeveer 10 milligram methyl-IPA op microbalans en registreert u de massa tot plus of min 0001 milligram. Breng de methyyl-IPA kwantitatief over naar een volumetrische kolf van 10 milliliter klasse A, met behulp van vier tot vijf milliliter acetonitril. Sonicate voor een minuut om alle vaste stoffen op te lossen en vervolgens te brengen tot het volume met acetonitril.Breng ongeveer acht milliliter van elke voorraad over op acht milliliter amberglazen flacons met PTFE-gevoerde doppen en bewaar de monsters op kamertemperatuur. Breng vervolgens de resterende voorraad over op gevaarlijk afval. Voor de 25X voorraad bereiden zeven methyl-IPA-voorraad een voorraad methyl-IPA en acetonitril voor op ongeveer 200 microgram per milliliter.Breng een milliliter van de voorraad over in een kolf van vijf milliliter en verdun het volume met acetonitril. Breng de voorraad vervolgens over op een acht milliliter amberglas flesje met een met PTFE omzoomde dop en bewaar op kamertemperatuur. Met behulp van een herhaalde pipettor, bereid de zes extra 25X methyl-IPA voorraden beschreven in het tekstprotocol in acht milliliter amber glazen flacons met PTFE-gevoerde doppen en op te slaan bij kamertemperatuur.Voor de 25x draagmoeder voorraad, breng 0,1 milliliter van de geconcentreerde ethyl-IPA voorraad naar een 10 milliliter klasse A volumetric kolf met behulp van een 100 microliter glas spuit en vervolgens verdunnen tot volume met acetonitril. Breng ongeveer acht milliliter van de draagmoeder voorraad naar een acht milliliter amber glazen flesje met PTFE-gevoerde dop en op te slaan op kamertemperatuur. Breng de resterende voorraad over op gevaarlijk afval.Bereid vervolgens 4X-voorraden voor die zowel methyl-IPA als ethyl-IPA bevatten in 10 milliliter schroeftop glazen auto sampler flacons, zoals beschreven in het tekstprotocol. Bijvoorbeeld, om voorraad 4X7 voor te bereiden, voeg 0.2 milliliter van de 25X voorraad zeven methyl-IPA voorraad aan een twee milliliter flacon met behulp van de herhaalde pipettor met een 0,5 milliliter capaciteit tip. Voeg 0,2 milliliter van de 25x surrogaat ethyl-IPA voorraad met behulp van een herhaal pipettor met een 0,5 milliliter capaciteit tip.Voeg 0,85 milliliter acetonitril toe met behulp van een herhaalde pipettor met een punt van één milliliterinhoud. Dop de flacon veilig en omkeren vijf keer om te mengen. Bereid hierna de standaardcurve voor in twee milliliter schroef-top auto sampler flacons zoals beschreven in het tekstprotocol.Bijvoorbeeld, om standaard zeven voor te bereiden, voeg 0,2 milliliter van de 4X7 voorraad aan een twee milliliter flacon, met behulp van een herhaalde pipettor met een 0,5 milliliter capaciteit tip. Voeg 0,6 milliliter ultra zuiverheid geïoniseerd water met behulp van een herhaalde pipettor met een een milliliter capaciteit tip. Dop de flesjes stevig af en keer vijf keer om te mengen.Bereid voor elk monster een 1,5 milliliter microcentrifugebuis met 200 tot 300 milligram natriumchloride en schik de buizen in een plastic rek van 80 plaatsen. Label voor elk monster één 1,5 milliliter microcentrifugebuis als A en een tweede microcentrifugebuis als B.Schik de buizen in een plastic rek van 80 plaatsen. Plaats de volgende materialen en apparatuur die nodig zijn voor serumextractie in een bioveiligheidskast van klasse twee: de microcentrifugebuizen; een vortex mixer, een herhaalde pipettor met 0,5 milliliter en vijf milliliter capaciteit tips, een 100 tot 1000 microliter luchtverplaatsing pipet met 1000 microliter tips, containers met een ongeveer 100 milliliter elk van verdunnings- en ultrazuiverheid geïoniseerd water, en biohazard afval container.Verwijder vervolgens serummonsters uit bevroren opslag en warm tot kamertemperatuur in de bioveiligheidskast. Vortex meng elk serummonster voorafgaand aan de bemonstering. Met behulp van een herhaalde pipettor met een 0,5 milliliter capaciteit tip, afzien van 05 milliliter mongoose serum in buis A en voeg 05 milliliter van de 25x surrogaat voorraad.Voeg vervolgens 0,95 milliliter verdunningsmetje toe aan buis A met behulp van een herhaalde pipettor met een punt van vijf milliliter capaciteit. Dop de buizen stevig vast en vortex mix voor 10 tot 15 seconden. Voeg vervolgens het controlemongoose serum toe aan buis A.Fortify elk van de 4QC monsters met me-IPA met behulp van een herhaalde pipettor met een 0,5 milliliter capaciteit tip.Voeg 25x draagmoedervoorraad toe aan de QC-monsters. Het ionenchromatogram van het representatieve monster uit de studie toont een waargenomen serumconcentratie van 33,5 microgram per microliter methyl-IPA.De nauwkeurigheid en precisie resultaten voor controle mangoest serum versterkt met methyl-IPA worden hier getoond.
