Om de structurele informatie op moleculair niveau te onderzoeken, heeft sfg zijn inherente voordelen (d.w.z. monolaag-of submonolayer-gevoeligheid en Interfaciale selectiviteit), die kunnen worden toegepast om verschillende interfaces te bestuderen, zoals de vaste/vaste, vaste/ vloeistof, vaste/gas, vloeistof/gas, vloeistof/vloeibare interfaces. Ontwikkeling van de som frequentie generatie (sfg) Vibrationele spectroscopie kan worden gedateerd terug naar het werk van Shen et al. dertig jaar geleden1,2. De uniciteit van de Interfaciale selectiviteit en sub-monolayer gevoeligheid maakt sfg Vibratie spectroscopie gewaardeerd door een groot aantal onderzoekers op het gebied van fysica, chemie, biologie, en materiaalkunde, etc3,4 ,5. Momenteel wordt een breed scala aan wetenschappelijke vraagstukken met betrekking tot oppervlakken en interfaces onderzocht met behulp van SFG, met name voor complexe interfaces met betrekking tot polymeren en biomacromoleculen, zoals de ketting structuren en structurele ontspanning in de begraven polymeer interfaces, de eiwit secundaire structuren, en de Interfaciale water structuren9,10,11,12,13,14, 15,16,17,18,19,20,21,22,23, 24,25,26.
Voor polymeer oppervlakken en interfaces worden dun-film samples over het algemeen bereid door een spin coating om de gewenste oppervlakken of interfaces te verkrijgen. Het probleem ontstaat door de signaalstoring van de twee interfaces van de as-prepared films, wat leidt tot ongemak voor het analyseren van de verzamelde sfg Spectra27,28,29. In de meeste gevallen is het trillationeel signaal alleen van een enkele interface, hetzij film/substraat of film/het andere medium, wenselijk. Eigenlijk is de oplossing voor dit probleemvrij eenvoudig, namelijk om de licht velden op de wenselijke interface te maximaliseren en de licht velden op de andere interface te minimaliseren. Vandaar dat de Fresnel-coëfficiënten of de lokale veld coëfficiënten moeten worden berekend via het dunne film model en moeten worden gevalideerd met betrekking tot de experimentele resultaten3,9,10,11, 12,13,14,15,30.
Met de bovenstaande achtergrond in het achterhoofd, kunnen sommige polymeer-en biologische interfaces worden onderzocht om de fundamentele wetenschap van het moleculaire niveau te begrijpen. In het volgende, het nemen van drie interfaciaal problemen als voorbeelden: indringende poly (2-hydroxyethyl methacrylaat) (phema) oppervlak en begraven interface met substraat9, vorming van zijde aan (SF) secundaire structuren op het polystyreen (PS) oppervlak en water structuren omringende model korte-keten oligonucleotide duplex16,21, we zullen laten zien hoe de sfg Vibrationele spectroscopie helpt om de interfaciaal moleculaire structuren te onthullen in verband met de onderliggende wetenschap.

interfacial molecular-level structures of polymers and biomacromolecules revealed via sum frequency generation vibrational spectroscopy

Als een tweede-orde niet-lineaire optische spectroscopie, is Sum Frequency Generation (SFG) Vibrationele spectroscopie op grote schaal gebruikt bij het onderzoeken van verschillende oppervlakken en interfaces. Deze niet-invasieve optische techniek kan de lokale informatie op moleculair niveau bieden met monolaag of submonolayer gevoeligheid. We bieden hier een experimentele methodologie voor het selectief detecteren van de begraven interface voor zowel macromoleculen als biomacromoleculen. Met dit in gedachten, worden interfaciaal secundaire structuren van zijde aan en water structuren rond model korte-keten oligonucleotide duplex besproken. De eerste toont een keten overlapping of ruimtelijke opsluiting effect en de laatste toont een bescherming functie tegen de CA2 + ionen die voortvloeien uit de chirale wervelkolom bovenbouw van water.

In de Fresnel coëfficiënt deel van protocol sectie, we hebben aangetoond dat, theoretisch, het haalbaar is om selectief te detecteren slechts één enkele interface in één keer. Hier, experimenteel, hebben we bevestigd dat deze methodologie in principe correct is, zoals weergegeven in Figuur 5 en Figuur 6.

Sum frequentie generatie trillingsspectra zijn uitgerust om interfaces met interfaciale selectiviteit te onderzoeken. De Fresnel-coëfficiëntenmethode kan echter helpen om het interferentie-effect op te lossen wanneer er een andere interface bestaat. Het belangrijkste voordeel van SFG is de interfaciale selectiviteit en submonolayer gevoeligheid.Deze niet-invasieve optische techniek is geschikt voor het investeren van verschillende interfaces, zoals vaste-vloeistof, vast-vast, vloeibaar-vloeistof, vast-gas, en vloeistof-gas interfaces. We raden aan dat beginners veel praktijk krijgen met het SFG-systeem met de hulp van experts in optica en de SFG. Er zijn geen snelkoppelingen.SFG is over het algemeen niet erkend als krachtige techniek, dus we willen meer publiek belangstelling te trekken en meer onderzoekers aan te moedigen om deze techniek toe te passen. Om te beginnen, bereiden ongeveer vijf freeilters van een twee of 4% in gewicht oplossing van poly-2-hydroxyethyl methacrylaat of PHEMA in watervrije ethanol. Bewaar de oplossing drie dagen voor gebruik in het lab.Vervolgens weken vier IR-grade calciumfluoride rechte schuine prisma in analytische kwaliteit watervrije tolueen voor ten minste 12 uur. Dompel vervolgens de prisma's onder in 30 millilters ethanol en veeg de oppervlakken af met ontvetterend katoen gedurende ongeveer 10 minuten. Spoel het prisma twee minuten af met ultrazuiver water en droog ze met stikstofgas.Plaats vervolgens de prisma's in een zuurstofplasmareiniger en evacueer de kamer. Behandel de prisma's met zuurstofplasma gedurende vier minuten en bewaar ze in de kamer totdat ze worden gebruikt. Filmvoorbereiding moet plaatsvinden binnen een uur na de plasmabehandeling.Om de barrière-interface selectief op te sporen, is het in deze studie van groot belang om de juiste filmdikte te kiezen op basis van het berekende resultaat van het Fresnal-coëfficiëntmodel. Wanneer u klaar bent om de PHEMA-folies voor te bereiden, bevestigt u een schoon prisma in een prismahouder op een spincoater en breng u één druppel van de oplossing van PHEMA in ethanol aan op het prisma. Voer de spincoater op 1, 500 RPM gedurende een minuut om de PHEMA-film voor te bereiden.Coat extra plasma behandeld prisma's met PHEMA op dezelfde manier. Anneal de films in een vacuüm oven ingesteld op 80 graden Celsius voor ten minste 10 uur. Om het experiment uit te voeren plaats de PHEMA gecoate gezicht van het prisma in gedeïmiseerd water.Wacht 10 tot 20 minuten en evalueer vervolgens de interfaciale PHEMA-structuur op de water- en prisma-interfaces met enige frequentiegeneratiespectroscopie. Tijdens het gebruik van het FGS-systeem laat de lichtbundel niet direct in uw ogen komen. Om te beginnen met de voorbereiding van de zijde fibroin oplossing warmte drie liter 0,02 molaire waterige natriumcarbonaat aan het koken.Kook 7,5 gram Bombyx mori zijde cocons in deze oplossing terwijl roeren gedurende 30 minuten. Breng de vezelige materie naar een schone container en roer het in twee tot drie liter gedeïoniseerd water gedurende acht tot 10 minuten drie keer af te wassen van de ongewenste sericine moleculen. Droog de vezelige materie in een vacuümoven op 60 graden Celsius gedurende ten minste 15 uur.Los vervolgens een gram droge, ontgemeen zijdefibroine op in vier millilters van 9,3 molaire lithiumbromide. Roer de oplossing op 60 graden Celsius gedurende twee uur. Plaats vervolgens de zijdefibroinoplossing in een 3,500 Dalton dialysezak.Dialyze de oplossing tegen een liter gedeïmiseerd water gedurende drie dagen, het veranderen van het water drie keer per dag. Zodra de dialyse is voltooid, bewaar de zijdefibroinoplossing op vier graden Celsius. Gebruik vervolgens de eerder beschreven methoden om een calciumfluoride prisma schoon te maken en het te coaten met een dunne polystyreenfilm van een 3,5% in gewicht polystyreenoplossing.Plaats het met polystyreen gecoate prisma in contact met de zijdefibroineoplossing en onderzoek de polystyreenragragfibroineinterface met SFG-spectroscopie. Los om te beginnen met de voorbereiding van de oligonucleotide duplex, los 10 nanomol van de juiste enkelstrengs oligonucleotide op in 0,5 millilters ultrazuiver water. Doe hetzelfde voor de gratis oligonucleotide.Combineer de oplossingen om een 10 nanomol per milliliter duplex oligonucleotide oplossing te verkrijgen. Combineer vervolgens twee milligram DPPC, twee milligram deuterated DPPC en een milliliter chloroform om de lipideoplossing te verkrijgen. Vervolgens, schoon en plasma behandelen van een calciumfluoride prisma zoals eerder beschreven.Bevestig dit prisma aan de monstergreep van een Langmuir-Blodgett trog. Vul de trog met gedeïmiseerd water en laat een gezicht van het prisma in de trog met een millimeter per minuut. Injecteer verschillende microliters van de lipideoplossing op het wateroppervlak en wacht tot de oppervlaktedruk stabiliseert met ongeveer 12 millinewton per meter.Dan beginnen met het comprimeren van de lipide monolayer op vijf millimeter per minuut. Wanneer de oppervlaktedruk bereikt 34 millinewton per meter lift het prisma uit de trog op een millimeter per minuut. Bereid vervolgens 500 microliter van een mengsel van de duplex oligonucleotide-oplossing en de lipideoplossing in een verhouding van één tot 100 molar.Vervang vervolgens de Langmuir-Blodgett-trog door een cilindrische polytetrafluorethyleencontainer gevuld met gedeïoniseerd water. Injecteer het oligonucleotide lipidemengsel op het water tot de oppervlaktedruk 34 millinewton per meter bereikt. Zet de lipide monolayer gecoate gezicht van het prisma in contact met de lipide oligonucleotide mengsel om het uiteindelijke monster te vormen.Evalueer de chirale en achirale watertrilsignalen met FSG-spectroscopie. In dit eerste voorbeeld vertoonde de interface tussen PHEMA hydrogels en het calciumfluoride prisma duidelijke scherpe pieken in het SFG-spectrum. Dit werd toegeschreven aan de gladde interface tussen calciumfluoride en de hydrogel.De interface tussen de hydrogel en het omringende water had bredere, minder intense eigenschappen omdat watermoleculen zich in het grootste deel van de hydrogel konden verspreiden. Hier toen de zijdefibroineconcentratie boven de kritische overlappende concentratie lag, werden er chirale secundaire structuren gedetecteerd op de oplossingsplystyreeninterface, tenzij methanol als inducerende agent werd toegevoegd. Onder de kritische overlappende concentratie werden chirale SFG-signalen gedetecteerd, zelfs zonder methanol toe te voegen, wat aangeeft dat geordende secundaire structuren zich niet ondersteunden bij de oplossingspolystyreeninterface.In de duplex oligonucleotide verankerde lipide bilayer monster variërend van de calcium-ionconcentratie had geen significant effect op de chirale water signaal dat voornamelijk overeenkomt met de hydratatie later van de chirale wervelkolom in de kleine groef. Daarentegen, de calciumconcentratie sterk uitgevoerd de achirale water signalen die voornamelijk overeenkomen met de waterlaag rond de duplex keten en de bilayer. Al met al suggereerde dit dat de chirale wervelkolom van de waterlaag de oligonucleotide kon beschermen tegen calciumionen.Het berekenen van de Fresnel-coëfficiënt en het kiezen van een geschikte filmdikte kunnen interferentieproblemen oplossen. Als multi-reflectie en breking worden beschouwd als de juiste elektrische veldverdeling bij de interfaces kan worden aangepast. Er zijn alternatieve methoden die kunnen worden gebruikt om dunne films met de gewenste diktes te bereiden, zoals stap-coating en de chemische damp afzetting.Het detecteren van het oppervlak en interfaciale structuren met betrekking tot hechting, wijting, wrijving en andere eigenschappen kan onderzoekers helpen de onderliggende mechanismen te begrijpen en nieuwe functionele materialen te ontwikkelen. Deze methode kan ook worden toegepast op andere systemen waar verschillende interfaces moeten worden onderzocht. Het medium dat u wenst dat de lichtstralen zich voortplanten, moet echter transparant zijn.
