Interfacial Molecular-level Structures of Polymers and Biomacromolecules Revealed via Sum Frequency Generation Vibrational Spectroscopy

合計周波数生成振動スペクトルは、界面選択性を有する界面を調べるのに取り付けられています。ただし、フレネル係数法は、別のインタフェースが存在する場合の干渉効果の解決に役立ちます。SFGの主な利点は、界面選択性とサブモノレイヤー感度です。この非侵襲的な光学技術は固体液体、固体固体、液体液体、固体ガスおよび液体ガスのインターフェイスのようなさまざまなインターフェイスを投資するために適している。初心者は、光学系とSFGの専門家の助けを借りてSFGシステムを操作する多くの練習を得ることを示唆しています。ショートカットはありません。SFGは一般に強力な技術として認識されていないので、より多くの聴衆の関心を集め、より多くの研究者にこの技術を適用するよう促したいと考えています。始めに、ポリ-2-ヒドロキイェチルメタクリレートまたはPHEMAの2または4重量%溶液の約5ミリルターを無水エタノールで調製する。使用の 3 日前に、ソリューションをラボに保管してください。次に、4つのIRグレードフッ化カルシウム直角プリズムを分析グレード無水トルエンに少なくとも12時間浸す。その後、プリズムをエタノールの30ミリルターに浸し、約10分間脱脂綿で表面を拭きます。プリズムを超純水で2分間リンスし、窒素ガスで乾燥させます。次に、プリズムを酸素プラズマクリーナーに入れ、チャンバーを退出します。4分間酸素プラズマでプリズムを治療し、使用されるまでチャンバーに保管してください。膜製剤は、プラズマ処理の1時間以内に起こるべきです。本研究では、バリア界面を選択的に検出するためには、フレズナル係数モデルの計算結果に従って適切な膜厚を選択することが非常に重要である。PHEMAフィルムを準備する準備ができたら、スピンコーターのプリズムホルダーにきれいなプリズムを固定し、プリズムにエタノール中のPHEMAの溶液の1滴を適用します。スピンコーターを1,500RPMで1分間実行し、PHEMAフィルムを準備します。PHEMAでプリズムを同様に処理した追加のプラズマをコーティングする。少なくとも10時間摂氏80度に設定された真空オーブンでフィルムをアニーリングします。実験を行うためにPHEMA被覆されたプリズムの面を脱イオン水に入れる。10~20分待ってから、水とプリズムの界面PHEMA構造を、何らかの周波数生成分光法で評価します。FGSシステムを操作している間、光ビームが直接あなたの目に入らないでください。絹フィブロイン溶液の調製を開始するには、沸騰に0.02モル炭酸ナトリウムの3リットルを加熱します。この溶液中に7.5グラムのボンビクスモリシルクコクーンを沸騰させながら、30分間かき混ぜます。繊維状物質をきれいな容器に移し、2〜3リットルの脱イオン水で8〜10分間攪拌し、不要なセリシン分子を洗い流します。繊維状物質を60°Cの真空オーブンで少なくとも15時間乾燥させます。次に、9.3モル水性臭化リチウムの4ミリルターに乾燥したデグマドシルクフィブロインの1グラムを溶解する。摂氏60度で溶液を2時間かき混ぜます。その後、シルクフィブロイン溶液を3,500ダルトン透析袋に入れます。1リットルの脱イオン水に対して3日間透析し、1日に3回水を交換します。透析が完了したら、シルクフィブロイン溶液を摂氏4度で保存します。次に、前述の方法を使用してフッ化カルシウムプリズムを洗浄し、3.5重量%のポリスチレン溶液から薄いポリスチレンフィルムでコーティングします。ポリスチレンコーティングされたプリズムをシルクフィブロイン溶液と接触させ、ポリスチレンシルクフィブロイン界面をSFG分光法で調べます。オリゴヌクレオチド二重の調製を開始するには、10ナノモルの適切な一本鎖オリゴヌクレオチドを0.5ミリルターの超純水に溶解する。無料のオリゴヌクレオチドでも同様にしてください。溶液を組み合わせて、ミリリットル二重オリゴヌクレオチド溶液当たり10ナノモルを得る。次に、DPPCの2ミリグラム、重水素DPPCの2ミリグラム、クロロホルムの1ミリリットルを組み合わせて脂質溶液を得る。次に、クリーンおよびプラズマは、前述のようにフッ化カルシウムプリズムを治療する。ラングミュア・ブロジェット・トラフのサンプルホールドにこのプリズムを取り付けます。トラフを脱イオン水で満たし、プリズムの1面を1分あたり1ミリメートルでトラフに下げます。水面に数マイクロリットルの脂質溶液を注入し、表面圧が1メートル当たり約12ミリニュートンで安定するのを待ちます。その後、1分間に5ミリメートルで脂質単層を圧縮し始めます。表面圧が1メートルあたり34ミリニュートンに達すると、1分あたり1ミリメートルでトラフからプリズムを持ち上げます。次に、デュプレックスオリゴヌクレオチド溶液と脂質溶液の混合物の500マイクロリットルを1〜100モル比で調製する。その後、ラングミュア・ブロジェット・トラフを、脱イオン水で満たされた円筒形のポリテトラフルオルエチレン容器に交換します。表面圧が1メートル当たり34ミリニュートンに達するまで、オリゴヌクレオチド脂質混合物を水に注入する。プリズムの脂質単層被覆面を、脂質オリゴヌクレオチド混合物と接触して、最終試料を形成する。FSG分光法でキラルおよびアキラル水振動信号を評価します。この最初の例では、PHEMAヒドロゲルとフッ化カルシウムプリズムとの界面は、SFGスペクトルにおける明確な鋭いピークを示した。これは、フッ化カルシウムとヒドロゲルとの間の円滑な界面に起因した。ヒドロゲルと周囲の水との間の界面は、水分子がヒドロゲルの大部分に拡散する可能性があるため、より広く、より強い特徴を有していた。ここでシルクフィブロイン濃度が臨界重なり濃度を上回った場合、メタノールを誘導剤として添加しない限り、溶液ポリスチレン界面でキラル二次構造を検出した。臨界重なり濃度を下回ると、メタノールを添加しなくてもキラルSFG信号が検出され、溶液ポリスチレン界面で補助されずに形成された二次構造が形成されたことを示す。二重オリゴヌクレオチドではカルシウムイオン濃度を変化させる脂質二重層試料は、主にマイナー溝におけるキラル脊椎の水和後に対応するキラル水信号に有意な影響を及ぼさなかった。これに対し、カルシウム濃度が強いのは、主に二重鎖と二重層を取り巻く水層に対応するアキラル水信号に影響を与えた。このことから、水層のキラル脊椎がカルシウムイオンからオリゴヌクレオチドを保護できることを示唆した。フレネル係数を計算し、適切なフィルム厚さを選択すると、干渉の問題を解決できます。マルチ反射と屈折が考慮される場合は、インタフェースでの適切な電界分布を調整することができます。ステップコーティングや化学気相蒸着など、所望の厚みで薄膜を作製する代替方法があります。接着、ホワイティング、摩擦、その他の特性に関連する表面および界面構造を検出することは、研究者が基礎となるメカニズムを理解し、新しい機能材料を開発するのに役立ちます。この方法は、さまざまなインタフェースを調査する必要がある他のシステムにも適用できます。ただし、光ビームを伝播させる媒体は透明である必要があります。
