このアッセイは、細菌構造および機能の調節を免疫細胞ベースの抗菌活性に結びつける。したがって、潜在的な治療の主要な免疫療法は、改善された細菌クリアランスの可能性を決定する際に評価することができる。眼球細胞性殺死アッセイは、伝統的に、細菌に対して提起された抗体の有効な機能を鈍化として評価するためのゴールドスタンダードとして機能する。このプロトコルは、既存の高スループットの標準化アッセイに対して、シンプルさと汎用性を提供します。このプロトコルでは、レンサ球菌肺炎に対する薬物治療の効果と、これらの効果が細菌の改善された食作用にどのように影響するかを調べる。ここでは、肺炎球菌にOPKAを適用する。しかし、このプロトコルは、免疫細胞による他の病原体の貪食性の殺害を評価するために採用することができる。細菌ストックを最適化し、カウントされる最終的なCFUsが最適な範囲内に収まるようにするために時間を捧げます。HL60細胞が生存可能で活性であることを保証するためにフローサイトメトリーを使用し、機能的に活性な細胞を確保するために異なる培養条件を試すことを強くお勧めします。視覚的なデモンストレーションでは、サンプルのメッキ方法と計数可能なサンプルの達成方法について、より深く理解できます。始めるには、L-グルタミンと15ミリリットルの熱不活化牛胎児血清を補充した500ミリリットルRPMIで構成されるHL60細胞培養培地を調製する。HL60細胞の増殖および維持のために、HL60細胞培養培地の10ミリリットルの500万個の細胞を摂氏37度および5%の二酸化炭素で75平方センチメートルのベントフラスコで培養する。HL60細胞の作業ストックを生成するには、HL60培養培地に1ミリリットル当たり約100万個の細胞を1ミリリットルの極低温チューブに10%ジメチルスルホキシドを添加した。HL60細胞を分化するために、培養1.5回10〜15ミリリットルのHL60細胞培養培地中の7細胞を、無菌フィルターで0.6%DMFを補い、75平方センチメートルのフラスコを摂氏37度、OPKAの3日前に5%の二酸化炭素を上限とした。細菌ストックサンプルを調製するには、WU2細菌株を37°Cで約2〜4時間適切なスープで成長させます。次に、6000回Gで2分間遠心分離して細菌をペレット化し、適切なスープ中の15%グリセロールの10〜30ミリリットルで細胞を再懸濁する。細菌培養物を無菌1.5ミリリットルの遠心分離チューブにアリコートし、摂氏80度で貯蔵する。次に、37°Cの水浴で細菌ストックの1バイアルを解凍します。細菌細胞をペレットにし、無菌条件下で500マイクロリットルのOBBで再懸濁した。血液寒天プレート上の未処理の細菌ストックのプレート希釈および30°Cで一晩プレートを培養する。インキュベーションステップの後、HL60細胞と共培養した未処理の細菌ストックの希釈ごとにコロニーを数える。どの細菌の希釈は、この細菌ストックと将来のOPKUのためのカウント可能なコロニーの最適な数をもたらす記録。細菌ストックの1本のチューブを解凍します。ペレット菌はGの6000倍2分間で、前工程で得られた最適希釈液でOBB中の細胞ペレットを再懸濁した。再懸濁された細菌希釈液のピペット10マイクロリットルは、ラウンドボトム96ウェル細胞培養板でウェルした。次いで、20マイクロリットルの適切な抗体または薬物処理を各実験によく重複して加える。コントロールウェルの場合は、治療用ウェルに使用するバッファーに応じて1XPBSまたはOBBを使用してください。室温でサンプルプレートを約90rpmで1時間振ります。3日前にDMFで処理されたHL60分化細胞を収穫するために、細胞をGの500倍で3分間ペレットした。上清を捨て、1XPBSの少なくとも10ミリリットルでペレットを洗います。洗浄した細胞をGの500倍で3分間ペレット化した。上清を捨て、OBBのセルを再中断します。1〜5最終ボリュームで無菌未希釈赤ちゃんウサギ血清を追加します。1時間の細菌培養の後、各サンプルを2つのグループの重複した井戸に分けます。次に、HL60補体混合物の50マイクロリットルを各実験セットの井戸に加え、50マイクロリットルのOBBを細菌の井戸だけに加えます。摂氏37度で96ウェルプレートを1時間振ります。サンプルをプレートするには、OBBを1~5個の最終体積で希釈し、各サンプルの体積が少なくとも50マイクロリットルになるようにします。次に、各サンプルのピペット50マイクロリットルを細菌培養プレートの指定領域に直接上にし、サンプル間の十分な間隔を確保する。蓋をして、サンプルを室温で約15分間乾燥させます。プレートを反転し、摂氏30度で一晩培養します。あるいは、嫌気性の瓶中の培養プレートは、嫌気性の状態が細菌の増殖に影響を与えるか、または形態を制御するかをテストする。一晩培養した後、各指定サンプル領域のコロニーをカウントし、各セット内の生細胞数を対応するコントロールと比較してデータの分析に進みます。この技術を初めて確立する最も困難な側面は、細菌ストックの開始CFUsを最適化することと、HL60細胞が効果的に分化されることを保証する可能性が高い。OPKAを開始する前に、HL60分化は、ヨウ化プロピジウムを有するフローサイトメトリーを生存マーカーとして使用して検証した。DMFを3日間処理した後、CD35の発現が増加し、CD71の発現が減少した。HL60処理群における細菌CFUsの平均割合は、対応する非HL60処置群と比較して、異なる治療の細菌細胞生存率を比較するために使用された。結果は、効果的な治療を行うと、コロニーの数がHL60共培養とHL60細胞の間で異なることを示し、より効率的な食細胞化を示した。結果は、細菌希釈が最適化されていないか、またはコロニーの成長がめっき後に注意深く観察されなかった場合、コロニーの過剰増殖がコロニーの正確なカウントを防ぐことができることを示した。この手順を試みる際に覚えておくべきことは、CFUsの過剰増殖を避けるために細菌のインキュベーションを注意深く監視することです。インビボアッセイは、宿主内で有効な細菌クリアランスをアセスするために使用することができる。これらのアッセイは、OPKAで観察される免疫有効性がヒトまたは動物モデルに翻訳可能であることを確認するために使用することができる。この眼球細胞性殺死アッセイは、抗菌療法をファゴサイト免疫応答に結びつける重要な研究で大きな効果を発揮するために使用されてきた。細菌病原体の使用は危険です。このアッセイ中は、常に保護用具を着用してください。
