外傷性脳損傷(TBI)は、世界的な公衆衛生上の問題であり、若年成人の死亡および障害の主な原因である1,2.TBIは毎年多くの死を引き起こし、生存者は様々な身体的、精神的、感情的、認知障害を経験します。したがって、TBIは患者の家族や社会資源に大きな負担です。TBIは、外傷時に発生する一次脳損傷と、最初の損傷の後に数時間から数ヶ月に発症する二次脳損傷の両方を含む。二次脳損傷は、脳に有害であるだけでなく、消化器系3を含む様々な臓器系に重大な悪影響を及ぼすいくつかの生化学的カスケードによって媒介される。
現在、動物実験でTBIを誘導するモデルは、流体パーカッション損傷、制御皮質衝撃(CCI)、体重減少加速の3つのモデルがあります。横液パーカッション傷害(LFPI)は、拡散性脳損傷(DAI)4を確立するために最も一般的に使用されるモデルである。装置は無傷の硬膜に短い流体圧力の脈拍を加えることによって頭蓋切開を通して脳損傷を作り出す。このパルスは振り子のストライクによって作成されます。LFPIはTBI研究のための再生可能で制御可能なモデリング方法である。
マイクロバイオームは、人体に存在するすべての微生物の集合的ゲノムとして定義されます。特に腸内微生物は、腸恒常性および機能において重要な役割を果たすだけでなく、宿主生理学および他の器官の機能の多くの側面を調節する5。近年、腸内微生物叢が脳腸軸6を介して脳の発達と機能を調節することを示す証拠が増えている。腸内細菌叢の破壊は、パーキンソン病、気分障害、および自閉症7を含むいくつかの脳機能障害にリンクされています。最近、前臨床研究はまた、急性脳損傷が腸内微生物叢8、9の変化を誘発しうることも報告されている。
