リサウイルス属内のウイルスは人獣共通病原体であり、少なくとも7種のリサウイルス種がヒトの症例に関連している。コウモリはほとんどのリサウイルスの自然貯蔵所であるため、コウモリのリッサウイルス監視プログラムは、コウモリのこれらのウイルスの生態を理解するために2008年から台湾で行われています。このプログラムでは、非政府のコウモリ保護団体と地元の動物病対策センターが協力して、衰弱や病気で死んだコウモリやコウモリを集めました。コウモリの脳組織は壊死を介して得られ、リッサウイルス抗原および核酸の検出のための直接蛍光抗体試験(FAT)および逆転写膜ポリメラーゼ連鎖反応(RT-PCR)を行った。FATの場合、少なくとも2つの異なる狂犬病診断コンジュゲートが推奨されます。RT-PCRの場合、2組のプライマー(JW12/N165-146、N113F/N304R)を使用して、リッサウイルスヌクレオタンパク質遺伝子の部分配列を増幅します。この監視プログラムは、コウモリのリッサウイルスや他の人獣共通性を監視します。台湾コウモリのリサウイルスは、2016年から2017年にかけて日本のピピストレルス・アブラムス(ピピストレルス・アブラムス)の2例で発見された。これらの知見は、コウモリや他の野生動物と接触する潜在的なリスクについて、一般市民、医療専門家、科学者に知らせるべきである。

リサウイルスは動物園の病原体です。台湾のコウモリ集団におけるリサウイルスの存在を評価するために、我々は監視プログラムを開始した。そして、2016年から2018年にかけて4つのリッサウイルスの同定に成功しました。このプロトコルは、台湾におけるビアル・リサウイルス監視のステップバイステップの紹介です。私たちは、vi-al Lyssavirusの監視に興味を持っている研究に役立つことを願っています。正しいタックケーグシンボルを接続することが重要です。死んだコウモリや死ぬコウモリは、リサウイルス監視のためのライブバットよりも適しています。研究所がある国のバイオ安全規則に応じて;適切なバイオ安全レベルの研究室で次の手順を実行します。そして、研究者が現在資格を持っていることを確認し、適切な個人的な保護具を着用してください。弱いコウモリや病気のコウモリや死体の収集時に、コウモリ生態学者に標本の形態学的特徴によってコウモリ種を特定してもらいます。各コウモリの標本に関する収集サイト、種、臨床徴候、その他の関連データを含む情報シートを研究室に提出します。壊死検査を開始する前に、すべての外部オリフィスを調べ、コウモリの外部特徴、特に種分化のための頭、耳、翼を撮影してください。経口綿棒サンプルを収集するには、無菌解剖板の腹側の精通にバットを置き、ピンセットで頭を固定します。メスを使用してふくらはぎ領域の正中線に沿って皮膚を切断し、皮膚を側面に引っ張り、ふくらはぎ領域の正中線に沿って頭蓋骨を切開します。頭蓋で頭蓋骨を開いて脳組織を露出させ、ピンセットを使用して脳組織を接続された神経組織から穏やかに分離します。脳幹や小脳を含む脳の断面を切断し、ガラススライドで脳組織の切断面に軽く触れて、スミア印象を与えます。レンズ組織のスライドを押して過剰な組織を取り除き、スミア印象スライドをマイナス20°Cのアセトンで30分間固定します。その後、組織病理学的検査のためにホルマリンで、新鮮な脳組織の小片を修正します。そして、残りの組織をemi-em 10に入れて核酸抽出を行う。次に、マニホールドから体の中線に沿ってアヌスに皮膚を切開し、ピンセットを使用して皮膚と下線筋組織を持ち上げ、分離し、下顎骨の近くに位置する唾液腺の採取を可能にする。ピンセットで胸骨を少し持ち上げ、メスを使って正中線に沿って胸骨と腹壁を切断し、鎖骨を切断します。針を使用して、左と右の肋骨ケージを解剖板に固定し、胸腔を開きます。そして、総軍団と死後の変化の程度を記録します。次に、ピンセットとメスを使用して、計画された下流分析に適した内臓組織を除去します。固定の終了時に直接蛍光抗体検査を行うには、スミア印象スライドを乾燥し、分析のために少なくとも2つの蛍光コンジュゲート抗狂犬病抗体を選択します。0.45マイクロメートルのストレーナーを通して各スライドに1つの抗体をフィルターします。そして、湿度の高いチャンバーで摂氏37度で30分間スライドをインキュベートします。インキュベーションの終わりに、余分なコンジュゲートを排出し、PBSでスライドを洗います。その後、10%グリセロールの少量を使用して、カバースリップをスライドに取り付け、蛍光顕微鏡でスライドを画像化します。蛍光抗体検査またはRT-PCR分析のいずれかが陽性を示す場合、emi-en10の10%懸濁液中の脳標本を均質化し、遠心分離によって組織をフルーリーに沈殿させる。スーパーナットエンの200マイクロリットルを、第6マウス神経芽細胞の3倍から6回目のマウス神経芽細胞細胞、1ミリリットルのエミエン10を摂氏37度と1%の二酸化炭素で1時間接種する。そして、脳の均質な細胞懸濁液を25センチメートルの2乗フラスコに移し、新鮮なエミエン10の6ミリリットルを含む。脳均質細胞懸濁液の1ミリリットルで4ウェルテフロンコーティングガラススライドに、直径6ミリメートルで、スライドを摂氏37度、2酸化炭素1%で3〜4日間置きます。100%アセトンで固定した後、蛍光抗体検査に使用した2つの蛍光コンジュ化抗狂犬病抗体でスライドを染色する。蛍光顕微鏡でスライドを可視化し、イントラシダプラス-マイク含有物の数を決定します。標準的なプロトコルに従って接種細胞培養をトリプシン化およびサブ培養した後、100%感染性に達するまで実証したように、感染細胞数を繰り返してさらに3〜4日間細胞培養インキュベーターに戻す。2014年1月から2017年5月にかけて、監視のために13種から332のコウモリの死骸が集められました。この2つの陽性コウモリ症例の代表的な分析では、脳の印象は、市販のフィットsy共役抗狂犬病抗体の1つを用いて蛍光抗体検査を使用して陰性を試験し、RT-PCRは2つの異なるプライマーセットのそれぞれを採用し、肯定的な結果をもたらした。Genバンクデータベースでのブラストクエリに対する得られたアンプリコン配列の主観は、検出されたリッサウイルスのアイデンティティを支持する79%未満のアイデンティティを有するリッサウイルスに配列が類似していることを明らかにした。その後、2つのリサウイルスが、蛍光抗体検査およびシーケンシングによって確認された2つの脳から正常に単離された。リサウイルスの陽性度を決定するための重要なステップは、蛍光抗体検査とRT-CPR分析です。プライマーセットにおける2つ以上の抗狂犬病抗体の使用が強く推奨される。リサウイルスに加えて、他の動物園ではないコウモリ病原体は、閉じた診断方法を使用して監視することができます。データは、コウモリとの接触を避けることについて一般の人々に知らせるために使用することができます。コウモリのリッサウイルスを研究すればするほど、その進化と公衆衛生への影響を理解できるようになります。
