1. Renilla-萤火虫双荧光酶记者

在考虑了测量误译的要求之后, 应当指出, 本议定书所述的功能遗传记录记者只能测量一种误译 (即一种氨基酸替代)密码子) 每个记者。因此, 为了测量不同的误译事件, 需要多名记者。例如, 要通过在一个位置通过 lysyl-tRNA 对近同源密码子的核糖体解码错误来测量误译, 至少需要16个记者 2,3,11。高精度质谱和生物信息学允许同时识别许多不同类型的误译事件 18;然而, 这些方法也伴随着警告。一般来说, 它们不如功能得分记者准确。此外, 如前所述, 它们不太适合检测某些类型的误译, 即可能与非酶脱胺23混为一谈的误译.最后, 质谱分析不适合作为中高通量筛选的读出。
对于这些记者和协议的改编, 以衡量误译在其他系统中, 进一步的考虑因素包括记者在所需的模型系统中的稳健表达。我们最初试图使用 Farabaugh 和我们的分枝杆菌系统中的同事开发的双荧光素酶系统, 但由于缺乏记者表达而没有成功。广泛的故障排除确定了代码优化和对记者的小序列修改都是为了允许鲁棒表达式 25 (见上文)。可以想象, 在其他系统中使用, 需要对记者进行类似的调整。
最后, 应考虑所衡量的误译事件的类型。例如, 如果需要测量停止密码读数 (无稽之谈抑制), 停止密码子的功能突变丢失需要引入主报告蛋白34 的非关键区域内。否则, 只有特定的无稽之谈抑制事件, 恢复关键残留物 (因此, 记者功能) 将通过分析测量, 这将有可能导致对真正的误译率的大量低估。越来越多的证据表明, 在大量的生物1、12、13、35中,平移误差可能起到适应性作用。然而, 对误译事件的测量仍然限于少数, 尽管模型物种越来越多。将敏感方法改编为测量误译, 有可能进一步增加科学家对翻译错误在生理和病理中的作用的认识。

分子生物学中的信息流要求将遗传信息转化为功能蛋白。与所有生物系统一样, 基因翻译也涉及可测量的错误。对翻译错误率的估计通常为每个密码子约为 1/10000 (由 Ribas de Pouplana 等人审查).然而, 错误率差别很大, 从不到10-5到超过 0.05/密码子1、2、3、4、5。误差率范围之广, 跨越三个数量级以上, 是由于翻译路径中的多个步骤可能产生的误差: 从随机、突变或压力引起的氨基酰化误差6,7.,8,9,10、天冬氨酸和谷氨酰胺-rna 5 的生理不均匀化, 或核糖体解码错误2,3,11。明显的高错误率, 代表超过 0.01. 1, 建议翻译错误可能执行生理功能1,12和误译可能是特定情况下的13。
我们和其他人已经证明, 基因翻译中自然发生的错误可能是适应性的, 特别是在环境压力1、5、12、14、15、16的情况下 ,17,18。在分枝杆菌中, 两步间接谷氨酰胺/天冬氨酰胺-氨基酰化途径19、20产生的误差显著提高了对一线抗结核抗生素利福平的耐受性5. 因此, 我们推测, 减少分枝杆菌误译与一个小分子可能有潜在的杀死利福平。我们筛选并鉴定了天然存在的氨基糖苷卡苏霉素, 将其作为一种化合物, 可以减少分枝杆菌的双译, 增强利福平介导的分枝杆菌在体外和体内的杀灭 21, 并限制利福平耐药 21的出现, 威胁到全球对结核病的控制 , 结核病是世界上最致命的病原体。
为了研究平移误差, 必须采用误译的测量方法。有多种方法已经开发的误译的测量, 每一种都有优点和缺点。简单地说, 基于精确质谱的方法有几个优点, 其中最重要的是, 使用较新的算法来检测多种类型的平移误差, 可以对误译进行相对无偏见的测量。执行了 18。然而, 质谱分析并不十分适合测量去酰胺化事件--正是由于间接 tRNA 酰化途径容易出错而在分枝杆菌中发生的误译类型。这是因为在处理样品进行质谱23 时发生了高频非酶脱胺, 从而产生了极高的背景信号。因此, 对于检测该途径中的错误, 遗传功能的记者提供了明显的优势。具体而言, 合适的功能增益记者可以有极低的背景率, 允许测量非常低的错误率11。
