L’EVI est un très accessible et pourtant une technique puissante pour cellulaire forcer en termes de synthèse et application. Avec tout le matériel prêt, le STI peuvent être synthétisé dans 1 jour. Cellules s’appuient sur les intégrines adhérer et d’exercer des forces cellulaires à la matrice extracellulaire. Transmission de force et adhérence cellulaire intégrine sont cruciales pour cellule diffusion1,2, migration3,4et survie5,6,7. À long terme, signalisation biomécaniques intégrine influe également sur cellule prolifération8,9,10 et différenciation11,12. Les chercheurs ont mis au point différentes méthodes pour mesurer et cellulaire de l’intégrine-transmis de carte oblige à l’interface de la cellule-matrice. Ces méthodes sont basées sur substrat élastique13, tableau de micropost14, ou atomique force microscopy (AFM)15,16. Méthodes élastiques de substrat et micropost reposent sur la déformation de substrats pour signaler le stress cellulaire et ont des limites en termes de résolution spatiale et forcer la sensibilité. AFM a force de haute sensibilité, mais il ne peut pas détecter la force à plusieurs endroits en même temps, compliquant la carte cellulaire force transmises par les intégrines.
Ces dernières années, plusieurs techniques ont été développées pour étudier la force cellulaire au niveau moléculaire. Un ensemble de capteurs de tension moléculaires basés sur polyéthylène glycol17,18, araignée de soie peptide19et ADN20,21,22,,23 ont été développés pour visualiser et surveiller la tension transmise par les protéines moléculaires. Parmi ces techniques, l’ADN a été adopté comme le matériau de synthèse dans la tension jauge d’attache (TGT), un linker cassables qui module la limite supérieure de l’intégrine tensions dans des cellules vivantes22,24. Plus tard, technique de transfert de résonance ADN et fluorescence ont été combinés pour créer en épingle à cheveux capteurs basés sur l’ADN fluorescente tension tout d’abord par Chen groupe23 et groupe20 de Salaita. Le capteur de tension basées sur l’ADN en épingle à cheveux intégrine tension en temps réel des rapports et a été appliqué avec succès à l’étude d’une série de fonctions cellulaires21. Par la suite, laboratoire de Wang combiné un TGT avec la paire de fluorophore-extincteur à tension intégrine rapport. Ce capteur est appelé une ITS25,26. La STI sont basée sur l’ADN à double brin (dsDNA) et a une plage dynamique plus large (pN 10-60) pour l’étalonnage de l’intégrine tension. Contrairement aux capteurs de basées sur l’ADN de l’épingle à cheveux, la STI ne signalent pas de force cellulaire en temps réel mais enregistre tous les événements de l’intégrine historique comme l’empreinte de force cellulaire ; ce processus d’accumulation de signal améliore la sensibilité cellulaire force d’imagerie, rendant possible à force cellulaire de l’image, même avec un microscope à fluorescence bas de gamme. La synthèse des STI est relativement plus pratique, car elle est créée par l’hybridation des deux ADN simple brin (ssDNA).
L’EVI est un ADN double brin 18-base-jumelé, conjugué à la biotine, un fluorophore, un désactiveur (Black Hole Quencher 2 [BHQ2])27et un acide (RGD) peptide cyclique arginylglycylaspartic28 comme un ligand de peptide intégrine (Figure 1). Le brin inférieur est conjugué avec le fluorophore (Cy3 est utilisé dans ce manuscrit, tandis que d’autres colorants, par exemple, série Cy5 ou Alexa, ont également fait leurs preuves possible dans notre laboratoire) et la balise de biotine, avec laquelle l’EVI est immobilisé sur un substrat par liaison biotine-avidine. Le brin supérieur est conjugué avec le peptide RGD et l’extincteur de trou noir, qui assouvit Cy3 avec environ 98 % trempe efficacité26,27. Avec le protocole présenté dans cet article, la densité du revêtement de l’ITS sur un substrat est d’environ 1 100/µm2. Il s’agit de la densité qui que nous a été calibrés pour 18 bp biotinylé dsDNA enduit sur le substrat Neutravidine fonctionnalisés en suivant le même protocole29de revêtement. Lorsque les cellules adhèrent au substrat recouvert de l’EVI, intégrine lie l’ITS par RGD et transmet la tension à la STI. La STI a une tolérance de tension spécifique (Ttol) qui est définie comme le seuil de tension qui sépare mécaniquement l’ADN double brin de l’ITS au sein de 2 s22. SA rupture par l’intégrine tension mène à la séparation de l’extincteur de la teinture qui émet par la suite par fluorescence. Par conséquent, la tension de l’intégrine invisible est convertie en un signal de fluorescence et la force cellulaire peut être mappée par imagerie de fluorescence.
Pour illustrer l’application de la STI, nous utilisons des poissons keratocyte ici, un modèle de cellule largement utilisée pour cell migration étude30,31,32, cellules CHO-K1, une lignée cellulaire non motile couramment utilisés et fibroblastes 3 t 3 NIH. Coimaging des structures de tension et de la cellule intégrine est également effectué.

imaging integrin tension and cellular force at submicron resolution with an integrative tension sensor

Moléculaire tension transmise par obligations intégrine-ligand est le signal mécanique fondamental dans la voie de l’intégrine qui joue un rôle important dans de nombreuses fonctions cellulaires et les comportements. Pour calibrer et image intégrine tension avec force haute sensibilité et une résolution spatiale, nous avons développé un capteur de tension intégrative (STI), un capteur de tension fluorescent basées sur l’ADN. L’EVI est activé pour une fluorescence si maintient une tension moléculaire, convertissant ainsi la force de signal fluorescent au niveau moléculaire. Le seuil de tension pour l’activation de la STI est accordable dans la fourchette de 10 à 60 pN qui couvre bien la plage dynamique de l’intégrine tension dans les cellules. Sur un substrat greffé avec une ITS, la tension de l’intégrine des cellules adhérentes est visualisée par fluorescence et image à résolution submicronique. L’EVI est également compatible avec l’imagerie structurale cellulaire dans les cellules vivantes et les cellules fixes. La STI a été appliquée avec succès à l’étude de la contraction de plaquettes et la migration cellulaire. Cet article détaille la procédure pour la synthèse et l’application de la STI dans l’étude de force cellulaire intégrine-transmis.

Ce capteur de tension intégrative à base d’ADN, ou ITS, convertissant le signal de force en fluorescence permet localement l’imagerie directe de la force cellulaire par microscopie de fluorescence. ITS a une haute résolution spatiale et une sensibilité élevée. Le seuil d’activation est tunable, permettant l’image sélective de la tension d’intégrine à différents niveaux.Après avoir reçu l’ADN mono-échoué personnalisé, ou SSDNA, d’intérêt, pour conjuguer le peptide RGD au quencher thiolated de ssDNA, ajoutez d’abord 100 microlitres de 0.5-molaire EDTA dans 400 microlitres d’eau à environ 450 microlitres de solution fraîchement préparée de TCEP. Ensuite, ajoutez 10 microlitres de la solution TCEP-EDTA à 20 microlitres d’ADN de thiol d’un millimlaire quencher dans PBS pour une incubation de 30 minutes à température ambiante. Pour conjuguer l’amine RGD avec du sulfo-SMCC, ajoutez 40 microlitres de sulfo-SMCC de 23 millimlaires fraîchement préparés à 100 microlitres de solution d’amine RGD fraîchement préparée de 11 millimolaires pour une incubation de 20 minutes à température ambiante.Pour conjuguer rgd amine SMCC avec le quencher thiolated ssDNA, mélanger l’ensemble du volume de la solution thiolated ssDNA quencher avec l’ensemble du volume de la solution SMCC RGD amine pour une incubation d’une heure à température ambiante. À la fin de l’incubation, transférer la solution à un stockage de quatre degrés Celsius pendant la nuit. Le lendemain matin, mélanger le chlorure de sodium trois molaire avec la solution d’ADN conjuguée au thiol et moins 20 degrés Celsius, réfrigéré, 100% éthanol à un rapport de 1 à 10 à 25, et placer la réaction à moins 20 degrés Celsius pendant environ 30 minutes.Lorsque l’ADN s’est précipité, recueillir l’ADN par centrifugation et réutiliser la pastille dans PBS pour mesurer la concentration d’ADN sur un spectromètre. Pour le capteur de tension intégrative, ou ITS, synthèse, mélanger les solutions d’ADN du brin supérieur et inférieur à un rapport de 1,1 à un molaire, et incuber la réaction pendant la nuit à quatre degrés Celsius avant de placer aliquots du mélange à moins 20 degrés Celsius pour le stockage à long terme. Pour l’immobilisation its, enrober une boîte petri de 29 millimètres de diamètre à fond de verre de 200 microlitres de 0,1 milligramme par millilitre d’albumine de sérum bovin biotinylé, ou BSA-biotine, pendant 30 minutes à quatre degrés Celsius.Lorsque le BSA-biotine a physiquement adsorbé sur la surface de verre, laver le plat trois fois avec 200 microlitres de PBS frais par lavage, suivie de l’incubation avec 200 microlitres de 50 microgrammes par millilitre de protéines avides pendant 30 minutes à quatre degrés Celsius. À la fin de l’incubation, laver le plat trois fois avec le PBS comme démontré, et ajouter 200 microlitres de 0,1 micromolaire ITS pour une autre incubation de 30 minutes à quatre degrés Celsius. À la fin de l’incubation, laver le plat trois fois avec du PBS, en laissant le dernier lavage dans le plat jusqu’au placage cellulaire.Pour initier une culture de kératocyte de poisson, utilisez une pince à épiler à bout plat pour arracher un morceau d’écaille d’un poisson, et appuyez doucement sur la balance sur le puits d’une boîte petri à fond de verre non modifié, le côté intérieur de l’échelle contactant le verre. Attendez environ 30 secondes que l’écaille de poisson adhère à la surface vitrée du plat avant de couvrir l’écaille d’un millimètre de dulbecco modifié d’Iscove. Ensuite, incuber l’écaille dans une boîte sombre et humide pendant six à 48 heures à température ambiante.Pour plaquer les kératocytes sur la surface ITS, remplacez le milieu du plat de culture des kératocytes par une solution de détachement cellulaire 1X pour une incubation de trois minutes à température ambiante. À la fin de l’incubation, remplacer la solution de détachement par un milieu complet frais et dissocier les cellules par un tuyautage doux. Ensuite, ajustez les cellules à la concentration appropriée pour le placage et ensemencez-les sur le plat enduit its pendant 30 minutes à température ambiante avant l’imagerie.Pour l’imagerie par tension d’intégrine en temps quasi réel dans les cellules vivantes, transférez les cellules au stade d’un microscope à fluorescence et sélectionnez l’objectif 40 ou 100 fois. Dans le logiciel d’imagerie, réglez le temps d’exposition à une seconde et l’intervalle d’image de 20 secondes pour obtenir une vidéo des images ITS. Ensuite, utilisez un logiciel d’analyse d’image approprié pour soustraire un cadre précédent de la vidéo ITS à partir d’un cadre actuel pour calculer le signal ITS nouvellement produit dans le dernier intervalle d’image.Le signal ITS nouvellement produit représente la tension intégrine générée dans le dernier intervalle d’image, signalant ainsi la force cellulaire de manière quasi-en temps réel. Ici, la cartographie de tension d’intégrine démontre l’induction de la tension d’intégrine à deux voies de force pendant la migration de keratocyte. La résolution de la carte des forces pourrait alors être calibrée pour être de 0,4 micromètre.La tension élevée d’intégrine se concentre à la marge arrière dans les kératines motiles de poissons. Dans les cellules nonmotiles, un modèle spécifique de tension d’intégrine qui est tout à fait différent de celui observé dans le kératocyte migrant rapide est formé. Les adhérences focales et la tension intégrine des kératines de poissons peuvent être co-imaged pour évaluer la relation entre la tension d’intégrine et la structure cellulaire dans les cellules d’intérêt.Avec l’ITS, la force d’adhérence cellulaire devient visible avec une résolution et une sensibilité comparables à l’imagerie structurale cellulaire. Cette technique ouvre la voie à l’étude d’une interaction force-structure dans les cellules.
