imaging integrin tension and cellular force at submicron resolution with an integrative tension sensor

このDNAベースの統合張力センサ(ITS)は、力信号を蛍光に局所的に変換することで、蛍光顕微鏡による細胞力の直接イメージングを可能にする。ITSは高い空間分解能と高感度を有する。活性化閾値は調整可能であり、異なるレベルでのインテグリン張力の選択的なイメージを可能にする。カスタマイズされた一本鎖DNA(ssDNA)を受け取って、RGDペプチドをチオラ化ssDNAクエンチャーに結合させた後、まず、400マイクロリットルの水で0.5モルEDTAの100マイクロリットルを、作製したTCEP溶液の約450マイクロリットルに加えます。次に、TCEP-EDTA溶液の10マイクロリットルをPBSの1ミリモルチオールDNAクエンチャーの20マイクロリットルに加え、室温で30分間インキュベーションします。RGDアミンをスルフォ-SMCCと結合するには、調製した23ミリモルスルフォ-SMCCを40マイクロリットル加え、100マイクロリットルの新鮮な11ミリモルRGDアミン溶液を室温で20分間インキュベーションします。RGDアミンSMCCをチオラ化ssDNAクエンチャーと共役にするには、チオラ化されたssDNAクエンチャー溶液の全容をRGDアミンSMCC溶液の全容と混合し、室温で1時間のインキュベーションを行います。インキュベーションの終了時に、溶液を一晩で摂氏4度の貯蔵に移します。翌朝、塩化3モルの塩化ナトリウムとチオール共役DNA溶液とマイナス20度の摂氏を混ぜ、冷やし、100%エタノールを1対10~25比で混合し、反応をマイナス20度で約30分間置きます。DNAが沈殿したら、遠心分離によってDNAを採取し、PBS中のペレットを再懸濁して分光器上のDNA濃度を測定する。統合張力センサー(ITS)の場合、合成は、上鎖と下の鎖DNA溶液を1.1モル比で混合し、反応を摂氏4度で一晩インキュベートしてから、混合物のアリコートをマイナス20°Cで長期保存します。ITS固定化のために、直径29ミリメートルのガラス底ペトリ皿に0.1ミリリットル当たり0.1ミリグラムのビチニ酸シウ血清アルブミン(BSAビオチン)を摂氏4度で30分間コーティングします。BSAビオチンがガラス表面に物理的に吸着した場合、洗浄ごとに200マイクロリットルの新鮮なPBSで皿を3回洗い、続いて4度で30分間、1ミリリットル当たり50マイクログラムの200マイクロリットルをインキュベートします。インキュベーションの最後に、示されているようにPBSで皿を3回洗い、0.1マイクロモルITSの200マイクロリットルを加えて、摂氏4度でさらに30分のインキュベーションを行います。インキュベーションの終わりに、PBSで皿を3回洗い、最後の洗浄を細胞メッキするまで皿に残します。魚の角質細胞培養を開始するには、平らな先端のツイーザーを使用して魚からスケールを摘み取り、スケールの内側がガラスに接触して、変更されていないガラス底のペトリ皿の井戸にスケールを軽く押し込みます。魚のスケールが皿のガラス表面に付着するまで約30秒待ってから、完全なIscoveの修正されたダルベッコの媒体の1ミリメートルでスケールをカバーします。その後、暗くて湿度の高い箱の中で、室温で6〜48時間スケールをインキュベートします。角質細胞をITS表面にプレートするには、ケサイトー細胞培養皿の培地を1X細胞剥離溶液に置き換え、室温で3分間インキュベーションします。インキュベーションの最後に、剥離溶液を新鮮な完全な媒体に置き換え、穏やかなピペットで細胞を解離します。次に、細胞をめっきに適した濃度に調整し、イメージング前に室温でITSコーティング皿に30分間播種します。生細胞での準リアルタイムインテグリンテンテグリン張力イメージングの場合、細胞を蛍光顕微鏡の段階に移し、目的の40倍または100倍を選択します。イメージングソフトウェアでは、露出時間を1秒に、フレーム間隔を20秒に設定して、ITS画像の映像を取得します。次に、適切な画像解析ソフトウェアプログラムを使用して、ITSビデオの前のフレームを現在のフレームから減算し、最新のフレーム間隔で新たに生成されたITS信号を計算します。新たに生成されたITS信号は、最新のフレーム間隔で発生したインテグリンテンションを表し、それによって細胞力を準リアルタイムで報告する。ここで、インテグリン張力マッピングは、角膜細胞移動中の2つの力の線路におけるインテグリン張力の誘導を示す。フォースマップの解像度は、0.4マイクロメートルに較正することができます。高いインテグリン張力は、モチル魚の角質細胞の後部マージンに集中する。非可動細胞では、高速移動角膜細胞で観察されるのとは全く異なる特定のインテグリンテンステンションパターンが形成される。魚の角質細胞の焦点接着とインテグリン張力を共像化して、対象細胞におけるインテグリン張力と細胞構造との関係を評価することができる。ITSでは、細胞接着力は、細胞構造イメージングに匹敵する分解能と感度で見えるようになります。この技術は、細胞における力構造相互作用の研究のための道を開きます。
