De groei van het ontwikkelen van axonen is hoofdzakelijk invasief en omvat ECM degradatie en het remodelleren. Met behulp van de hier gepresenteerde procedure, kunnen onderzoekers verkrijgen van een homogene 3-D matrix gevormd door de natuurlijke type I collageen waarin axonen (of cellen) kan reageren op een chemische gradiënt afgescheiden door genetisch gemodificeerde cellen zoals ze doen in vivo. Neuronale axonen, eindigend in axonale groei kegels, migreren lange afstanden door de extracellulaire matrix (ECM) van het embryo over specifieke wegen om hun juiste doelen te bereiken. De groei kegel is het distale gedeelte van Axon en het is gespecialiseerd om het fysieke en moleculaire milieu van cel te voelen1,2. Vanuit moleculair oogpunt, worden de groei kegels geleid door minstens vier verschillende moleculaire mechanismen: de aantrekkelijkheid van het contact, chemoattraction, de weerzin van het contact, en chemorepulsion veroorzaakt door verschillende axonale begeleidings cues3,4 , 5 , 6. contact-gemedieerde processen kunnen gedeeltelijk worden bewaakt in twee-dimensionale (2D) culturen op micro-patroon substraten (bijv. met strepen7,8 of spots9 met de moleculen). Nochtans, kunnen axons aan hun doel in een niet-Diffusive manier navigeren door verscheidene aantrekkelijke en weerzinwekkende molecules van guidepost cellen in het milieu4,5,10te ontdekken. Hier beschrijven we een eenvoudige methode van 3-D cultuur om te controleren of een afgescheiden molecuul induceert chemorepulsive of chemoattractive effecten op de ontwikkeling van axonen.
De vroegste studies gericht op het bepalen van de effecten van Axon begeleiding cues gebruikt explant culturen in drie-dimensionale (3-D) matrices te genereren gradiënten simuleren in vivo voorwaarden11,12. Deze aanpak, samen met in vivo experimenten, toegestaan voor de identificatie van vier grote families van oriëntatie cues: netrinse, spleten, semaphorins, en Ephrins4,5,6. Deze moleculaire cues en andere factoren13 zijn geïntegreerd door de groeiende axonen, triggering de dynamiek van adhesie complexen en transducing mechanische krachten via de cytoskelet14,15,16. Voor het genereren van moleculaire gradiënten in 3-D culturen voor axonale navigatie, baanbrekende onderzoekers gebruikt plasma stolsel substraten17, die ook werd gebruikt voor organotypic slice preparaten18. Nochtans, in 1958, werd een nieuw protocol om 3-D collageen hydrogels te produceren gerapporteerd voor het bestuderen met Maximow ´ s apparaten19, een cultuur platform, dat in verscheidene studies geschikt voor microscopische observaties20wordt gebruikt. Een andere pionier studie rapporteerde collageen gel als hulpmiddel om menselijke fibroblasten in te bedden voor het bestuderen van de differentiatie van fibroblasten in myofibroblasten in wond helende processen21. Parallel, Lumsden en Davies paste collageen van de boviene dermis toe om het vermeende effect van de factor van de zenuwgroei (NGF) op het begeleiden van sensorische zenuwvezels te analyseren22. Met de ontwikkeling van nieuwe cultuur platforms (b.v., multi-well platen) door verschillende bedrijven en laboratoria, werden de collageen culturen aangepast aan deze nieuwe apparaten6,23,24,25 ,26. Parallel, werd een uittreksel van ECM materiaal dat uit de Engelbreth-Holm-zwerm tumor cel lijn wordt afgeleid gemaakt commercieel beschikbaar om deze studies uit te breiden27.
Onlangs, zijn verscheidene protocollen ontwikkeld om moleculaire gradiënten met vermeende rollen in axon begeleiding te produceren gebruikend 3-D hydrogels (b.v., collageen, fibrine, enz.) 28. als alternatief kan de kandidaat-molecuul worden gemobiliseerd op verschillende concentratie in een poreuze matrix (BIJV. NGF29) of gegenereerd door kweken in een klein gebied van de 3-D hydrogel cel aggregaten afscheiden van het molecuul om een radiale genereren gradiënt4,23,24,25,26. De laatste mogelijkheid zal worden toegelicht in dit protocol.
De hier gepresenteerde procedure is een eenvoudige, snelle en zeer reproduceerbare methode gebaseerd op de analyse van de axonale groei in 3-D hydrogel culturen van de embryonale muis hersenen. In vergelijking met andere methoden, is het protocol zeer geschikt voor niet-opgeleide onderzoekers en kan volledig worden ontwikkeld na een korte opleiding (1-2 weken). In dit protocol, isoleren we eerst collageen van volwassen rat staarten om verder te genereren 3-D matrices waarin genetisch gemodificeerde cel aggregaten worden gekweekt in de voorkant van de embryonale neuronale weefsel. Deze cel aggregaten vorm radiale chemische gradiënten van een kandidaat-molecuul dat een reactie voor de groeiende axonen ontlokt. Ten slotte kan de evaluatie van de effecten van het molecuul op de groeiende axonen gemakkelijk worden uitgevoerd met behulp van een fasecontrast microscopie of, alternatief, immunocytochemische methoden.

Dit protocol maakt gebruik van natuurlijke type I collageen te genereren drie-dimensionale (3-D) hydrogel voor het toezicht op en het analyseren van de axonale groei. Generation of 3-D Collagen-based Hydrogels to Analyze Axonal Growth and Behavior During Nervous System Development

Deze methode is nuttig om de rol van sommige moleculen in axonale begeleiding en neuronale migratie tijdens de ontwikkeling van het neurosysteem te analyseren. Het belangrijkste voordeel van deze techniek is dat de analyse van de resultaten is snel en eenvoudig en vereist geen complexe software die veel training voor onderzoekers zou impliceren. Demonstreren van de procedure zal worden Mirian Segura-Feliu, een technicus van ons laboratorium en ikzelf.Om plaat 200, 000 COS-1 cellen in 35 millimeter Petri schotels te beginnen en met volledig cultuurmiddel in een couveuse van de celkweek te broeden om 70 tot 80% samenvloeiing 's nachts te lezen. Plaats de schotel in de couveuse op 37 graden Celsius en 5% kooldioxide totdat de gelatie wordt waargenomen.Bereid een tweede strook met controlecellen in een tweede kweekschaal en plaats deze in de couveuse. Wanneer gelatie is voltooid, voeg drie tot vier milliliter van 37 graden celsius opgewarmd COS-1 complete cultuur medium aan elk gerecht met de gelled collageen cel strips en leg ze in de couveuse. Gebruik een fijne scalpel of een weefsel chopper om de collageen cel strips snijden om kleine vierkante stukken te genereren, 400 tot 500 micrometer in lengte.Breng alle secties van dezelfde transfectie conditie naar een petrischaaltje met drie tot 3,5 milliliter van neuronale cultuur media. Controleer onder een ontleedmicroscoop de kwaliteit van de stukken en plaats de gerechten in de couveuse. Gebruik een schaar om embryohoorns te snijden op embryonale dag 16,5 uit de buikholte van een geofferde zwangere vrouwelijke rat en plaats ze in een grote petrischaal met koude HBSSg buffer.Plaats de schotel in de laminaire stroomkap en gebruik rechte tangen om de embryo's te extraheren en plaats ze in een nieuwe schotel met koude HBBSg. Met kleine tangen, verwijder de huid, en vervolgens met gebogen en rechte tangen, zorgvuldig ontleden de hersenen en plaats in een schotel met koude HBSSg. Onder een ontleden microscoop, met behulp van een scalpel of fijne schaar, snijd de hersenen in de helft langs de middellijn om zowel de hemisferen te scheiden.Verwijder vervolgens de diencephalon en verwijder met fijne tangen hersenvliezen en bloedvaten uit de hersendelen. Gebruik 100% ethanol om alle delen van de weefselhakselaar schoon te maken, met name PTFE snijplaat en het scheermesje en houd de weefselhakselaar gedurende 15 minuten in de laminaire fluxkap onder UV-verlichting. Breng elk weefselstuk over op de snijplaat van de weefselhakselaar.Nadat het weefsel is gehakt, breng de stukken van de 35 millimeter petrischaaltjes gevuld met drie tot vier milliliter van volledige NCM. Met behulp van fijne wolfraamnaalden, maak de weefseldissectie in volledige NCM af. Controleer de kwaliteit van de verkregen plakjes onder de ontledenmicroscoop, zorg ervoor dat de lagen duidelijk herkenbaar zijn in de donkere veldoptica en ontleed het gebied van belang met wolfraamnaalden.Bewaar de stukken van goede kwaliteit in een compleet NCM-medium in de koolstofdioxide-incubator. Visuele demonstratie is van cruciaal belang in de stappen in verband met de voorbereiding van de co-culturen die de woorden vergemakkelijkt om het proces te begrijpen. Plaats verschillende steriele vier goed cultuur platen in de laminaire flow kap en bereiden een collageen werken mengsel als voorheen.Voeg 15 tot 20 microliter van het hydrogelmengsel toe aan de bodem van elke put om een cirkelvormige collageenbasis te produceren. Plaats de gerechten in de couveuse totdat volledige gelatie is waargenomen. En controleer de kwaliteit van het gelled collageen.Breng met een pipet een klein stukje COS-1-celaggregaat over op de hydrogelbasis en gebruik vervolgens een pipet om een weefselstuk op dezelfde basis te plaatsen, dicht bij het stuk cellen op een afstand van één explantgrootte. Na het bereiden van een nieuw werkend collageenmengsel op ijs, voorzichtig pipette 15 tot 20 microliter van dit nieuwe mengsel om de explant en het celaggregaat te bedekken, waardoor een sandwich als hydrogelcultuur wordt gemaakt. Gebruik een fijne wolfraam naald om de explant te heroriënteren, zodat het gezicht van de cel aggregaat op 500 tot 600 micrometer.Breng de plaat terug naar de couveuse totdat de gelatie wordt waargenomen en voeg vervolgens 0,5 milliliter complete NCM toe aangevuld met 2%B27-supplement en bewaar culturen gedurende 36 tot 48 uur in de couveuse. Toen hippocampal axonen werden geconfronteerd met Netrin-1, groeiden ze bij voorkeur naar de bron van Netrin-1, wat aangeeft dat Netrin-1 fungeert als een chemo aantrekkelijk molecuul voor deze axonen. In de controlevoorwaarde, of mock transfection, groeiden alle axonen radiaal, zonder enige directionele voorkeur.Toen hippocampal axonen werden geconfronteerd met Sema3E afscheidende cellen, de meeste van hen groeide tegenovergesteld van de cel aggregaat, wat aangeeft dat Sema3E fungeert als een chemo weerzinwekkende molecuul voor deze axonen. Deze schematische weergave van de axonale respons en kwantificeringsmethode toont aan dat in controleomstandigheden de axonen gelijkelijk werden verdeeld in zowel proximale als distale kwadranten, weergegeven met radiale uitgroei. Dit duidde op een proximale distale of PD-verhouding van één.Toen explants een toename van het aantal axonen in het proximale kwadrant vertoonden in vergelijking met het distale, wat duidt op chemo-aantrekkingskracht, was de PD-verhouding groter dan één. Toen het aantal axonen hoger was in het distale kwadrant dan in het proximale kwadrant, wat duidt op chemoafstoting, was de PD-verhouding lager dan één. Volgens deze methode kunnen onderzoekers essentiële informatie verkrijgen over de rol van sommige moleculen tijdens de neuronale ontwikkeling, waardoor we een startpunt kunnen maken om de functies verder te verkennen.Hoewel deze techniek nuttig is in neuronale ontwikkeling als dit is, kan het ook worden toegepast in farmacologische screening, angiogenese, en weefsel engineering als strategieën.
