سيساعد هذا البروتوكول الباحثين على توليد نوع جديد من نموذج الماوس لتكميل تطور سرطان الثدي البشري من مجموعات فرعية مختلفة من الخلايا الظهارية الثديية. هذه التقنية تسمح للباحثين لتوليد نماذج فأر سرطان الثدي في الأنسجة مرحلة النمو على نوع الخلية بطريقة محددة ودون الحاجة إلى تربية واسعة الماوس. لبدء هذا الإجراء، قم بإنشاء ماوس مزيل الماوس، والاحتفاظ به، ومزيل التخدير كما هو موضح في بروتوكول النص.جلب الماوس إلى منطقة منفصلة عن حيث الجراحة التي يتعين القيام بها. بعد التأكد من التخدير عن طريق إجراء قرصة إصبع قدم، حقن ميلوكسيكام كما مسكن تحت الجلد بجرعة من خمسة ملليغرام للكيلوغرام الواحد للتحضير لإجراء العمليات الجراحية. فضح موقع الحلمة الجراحية عن طريق تطبيق عدة قطرات من كريم إزالة الشعر.إزالة كريم المفرط والشعر فضفاض باستخدام المناشف الورقية الناعمة. لا ينصح الحلاقة لتجنب تلف الحلمات. تطهير موقع الجراحة مع iodophors تليها لأول مرة 70٪ الكحول وتنتهي مع التطبيق النهائي من iodophors فرك.القيام بذلك في حركة دائرية من وسط منطقة العمل نحو المحيط باستخدام إسفنج الشاش أو قضيب القطن يميل. كرر الدورة من ثلاث إلى أربع مرات. استخدام تقنيات معقمة طوال العملية الجراحية.جعل شق على الجلد على طول ما يقرب من سنتيمتر واحد بين الغدد الشعاعية الإربية الرابعة. فصل بعناية رفرف الجلد من الصفاق الجداري وذلك لبصر شجرة القناة الثديية. أمسك الحلمة بعناية مع أجهزة صنع الساعات لإزالة الحلمة الخارجية دون قطع أي جلد قريب باستخدام مقص microdissection.تحميل ما يقرب من ثلاثة إلى خمسة ميكرولترات من خليط حقن فيروس Cre-adeno في حقنة هاملتون 25 ميكروليتر مع إبرة محور معدنية قياس 33 الملصقة. تقدير حجم خليط الحقن في الحقنة على أساس الصبغة الزرقاء المدرجة في الخليط. أمسك بلطف حافة رفرف الجلد مع ملاقط منحني ناعم وحقن خليط حقن الفيروس القرين ببطء في الحلمة أثناء مراقبة انتشار الصبغة الزرقاء في شجرة القناة الثديية.الحفاظ على معدل الحقن في بطء ممكن لتجنب الأضرار التي لحقت تجويف القناة. يشار إلى الحقن الناجح داخل الدوكتال عن طريق حقن السوائل المنتشرة في جميع أنحاء شجرة القناة بأكملها دون تسرب في مقصورة ستروما. سحب بلطف إبرة من الحلمة لتجنب أي تسرب من السائل المحقون.فحص الجانب القاصي من الغدة الثديية أو المنطقة المحيطة بالحلمة المحقونة. صبغ نشرها في ستروما القريبة كما هو مبين هنا يشير إلى حقن وسادة الدهون ماماري بدلا من حقن ناجحة داخل المستقيم. أغلق الجروح الجراحية في الجلد بمقاطع الجروح.إزالة الماوس من التخدير ووضعها على وسادة التدفئة داخل قفص نظيف للانتعاش. مراقبة تطور ورم الثديي كما هو موضح في بروتوكول النص. أصغر الفئران الإناث التي يمكن إجراء الحقن داخل المستقيم بنجاح هي تلك في ما يقرب من ثلاثة أسابيع من العمر.على الرغم من أن معظم التجارب التعريفي ورم ماماري, يتم استخدام الفئران الإناث الشباب البالغين. يمكن إجراء الحقن داخل الرحم مع حجم أكبر من خليط الحقن في الفئران الإناث خلال فترة الحمل المبكرة إلى المتوسطة لاستهداف الخلايا السنخية. لاستهداف مختلف الخلايا الظهارية الثديية الفرعية للتجنيد الورم mammary، تم استخدام الفيروسات Cre-adeno تحت سيطرة مختلف الخلايا الظهارية الثديية فرعية المروجين محددة للحقن.على سبيل المثال تم استخدام فيروس Cre-adenovirus تحت سيطرة كيراتين 8 المروج لاستهداف الخلايا الظهارية الظهارية المامية اللماحية. استهدف فيروس Cre-adeno تحت سيطرة كيراتين 5 المروج النسب القاعدي مما أدى إلى وضع علامات وراثية للخلايا الظهارية الشعاعية القاعدية فقط. لأنثى P53 الفئران المثلية المخفقة مع مراسل ROSA26-YFP، أدى حقن داخل الكبد من الكيراتين 8 فيروس Cre-adeno إلى تطوير أورام الثديي بعد عدة أشهر من الحقن.بسبب إدراج مراسل R26-Y الشرطي ، تم وضع علامات على الخلايا الظهارية الورمية عادة من قبل YFP ويمكن اكتشافها عن طريق قياس التدفق. يمكن إثراء الخلايا عن طريق فرز تدفق الخلايا الإيجابية YFP لمزيد من التحليل. أهم الأشياء التي يجب تذكرها عند محاولة هذا الإجراء هي استخدام حجم حقن صغير ، وحقن ببطء ، وكذلك سحب الإبرة ببطء.بعد هذا الإجراء، يمكن للمرء أن يراقب تطور الخلايا الظهارية الظهارية الإيجابية YFP المستهدفة إلى مراحل ما قبل الخبيث والسرطان عن طريق قياس تدفق الخلايا أو التلطخ المناعي المشترك للغدة المحقونة. هذه التقنية سوف تمهد الطريق للباحثين لدراسة خلايا المنشأ من سرطان الثدي، وكيف أنها تستجيب لمختلف الأحداث autogenic وكيف أنها تتقدم إلى الخلايا السرطانية. كل من الفيروس الغدي والإبرة للحقن هي خطرة محتملة.لذلك، ينبغي للمرء دائما ارتداء القفازات واستخدام الإبر والمحاقن السلامة عند إجراء الحقن داخل المستقيم.
