Definir la conectividad entre las regiones cerebrales es necesario para entender los circuitos neuronales. Los métodos clásicos de rastreo anatómico permiten establecer la conectividad interregional, y los estudios de lesiones ayudan a entender la organización jerárquica del flujo de información. Por ejemplo, los circuitos cerebrales para la orientación espacial y la señalización de la dirección de la cabeza implican el flujo direccional de información desde el tálamo hasta el presubiculum. Esto ha sido demostrado por estudios de lesiones de núcleos talámicos antero-dorsales (ADN) que degradan la señal de dirección de la cabeza en el presubiculo dorsal aguas abajo, así como la señal celular de la red parahipocampal1,2.
La conectividad funcional entre las áreas cerebrales es más difícil de establecer a nivel celular y subcelular. En el hipocampo, una anatomía altamente organizada permite investigar conexiones sinápticas específicas de la vía utilizando la simulación eléctrica en la preparación de la rebanada. Los electrodos de estimulación colocados en el estrato radiato de CA1 se pueden utilizar para estimular específicamente la entrada colateral de Schaffer de CA33. Los electrodos estimulantes colocados en el lacunos molecular e estrato de CA1 activarán la entrada de trayectoria perforante a CA14,5. La estimulación eléctrica activa la liberación de neurotransmisores de los terminales de axón; sin embargo, activa las neuronas con somata cerca del sitio de estimulación, así como axones de paso. Por lo tanto, es de uso limitado para el estudio de afferents de regiones cerebrales definidas cuando las fibras de diferentes regiones de origen se entremezclan en la estructura objetivo, como es típicamente el caso en el neocórtex.
Las neuronas también pueden estimularse con luz. Los métodos ópticos incluyen la fotoactivación del glutamato enjaulado, que se puede combinar con el escaneo láser de uno o dos fotones. Múltiples sitios estrechamente espaciados pueden ser estimulados secuencialmente, sin daño mecánico al tejido6. Esto se ha utilizado con éxito para mapear receptores sinápticos, así como activar las neuronas individuales7. Mientras que el desafiamiento de glutamato se puede utilizar para el análisis de circuitos locales, no permite la activación específica de entradas de largo alcance.
Un método de elección para la investigación de la conectividad de largo alcance en circuitos neuronales es el uso de la expresión de canalizarpsina mediada por virus. Utilizando inyecciones estereotaxócicas in vivo como se describe aquí, la expresión de canales iónicos bloqueados por la luz puede ser dirigida y restringida espacialmente a una región cerebral deseada. De esta manera, las clasrodoninas son eficaces para mapear la conectividad excitatoria o inhibitoria de una región a su objetivo. Los terminales de axón transfectó pueden estimularse con luz en una preparación de corte cerebral, y las grabaciones de abrazaderas de parche como una lectura permiten el examen de las funciones y fortalezas de los componentes específicos del circuito en el cerebro8. El enfoque optogenético combinado con la inyección estereotaxia de un virus ofrece una especificidad sin precedentes y un control genético9. Estimular con la luz además permite una alta precisión temporal y espacial10,11.
El presubiculum es una estructura cortical de seis capas en la transición del hipocampo y la formación para-hipopótamo12,13. Recibe importantes aportaciones sinápticas del ADN11, pero también de varias otras regiones corticales y subcorticales14. Por lo tanto, la estimulación selectiva de terminales de axones talámicos dentro de una rebanada presubicular no es posible con la estimulación eléctrica ni el desatonación de glutamato. En este protocolo se describen métodos para determinar la conectividad funcional entre las regiones cerebrales (ADN y presubiculum) utilizando inyecciones estereoofócicas precisas de vectores virales que expresan canales de luz cerrada. También se describe la fotoestimulación de los terminales de axón de las neuronas que se proyectan en su región objetivo, junto con las grabaciones de abrazaderas de parche de células enteras de las neuronas postsinápticas en la preparación de la rebanada del cerebro.

El conocimiento de la conectividad sináptica específica de tipo celular es un requisito previo crucial para comprender los circuitos neuronales de todo el cerebro. La investigación funcional de conexiones de largo alcance requiere registros específicos de neuronas individuales combinadas con la estimulación específica de entradas distantes identificadas. Aquí se describe un conjunto de técnicas para la inyección estereoopónica precisa de vectores virales que expresan la cifrodonina en el cerebro del ratón, seguido de la fotoestimulación de los terminales de axón en la preparación de la rebanada del cerebro. Estos protocolos son simples y ampliamente aplicables. En combinación con la grabación de abrazaderas de parche de células enteras de una neurona conectada postsinánicamente, la fotoestimulación de los axons permite la detección de conexiones sinápticas funcionales, caracterización farmacológica y evaluación de su fuerza. Además, el llenado de biocitetina de la neurona registrada se puede utilizar para la identificación morfológica post-hoc de la neurona postsináptica.

El procedimiento presentado aquí se utilizó para expresar una canalización sensible a la luz azul (Chronos) fusionada a la GFP en el núcleo antero-dorsal del tálamo (ADN), por inyección estereotaxc de virus asociado al adeno anterogrado. El objetivo de este método es identificar la conectividad funcional de las entradas de largo alcance de regiones cerebrales distantes utilizando la fotosestimulación en las rebanadas cerebrales. La segmentación estereotáctica de una región cerebral específica para diversas expresiones mediadas de canales iónicos sensibles a la luz permite la estimulación selectiva de axones procedentes de esa región con luz.
