Aangeboren immuun afweer vormen de eerste lijn van verdediging tegen pathogene microben. Deze reacties kunnen functioneel worden onderverdeeld in humorale en cellulaire aangeboren immuniteit, die beide worden gemedieerd door germline-gecodeerde patroon erkenning receptoren (PRRs) die zin pathogen-geassocieerde moleculaire patronen (PAMPs)1. Veel van de signaalroutes en effector mechanismen van aangeboren immuniteit zijn bewaard in zoogdieren en ongewervelden, zoals de nematode, Caenorhabditis elegans, en de fruitvlieg, Drosophila melanogaster2. De fruitvlieg heeft ontpopt als een krachtig systeem te bestuderen host verdediging tegen infectieuze micro-organismen3. Drosophila is genetisch hanteerbare, gemakkelijk en goedkoop een ziek rund opgefokt in laboratoria, en heeft een korte generatietijd. Bovendien vertoont de fruitvlieg hoogefficiënte verdedigingen tegen een array van microben, waardoor het onderzoek van de immuniteit van de gastheer tegen virale, bacteriële, schimmel of parasitische ziekteverwekkers.
Drosophila immunologen hebben historisch gebruikt voorwaartse genetische schermen, genoom-brede RNA-gemedieerde interferentie (RNAi) screening van insecten cellijnen, en reeds bestaande mutant vliegen stammen te onderzoeken van aangeboren immuniteit – leidt tot de identificatie en karakterisering van verschillende evolutionair geconserveerde humorale immune trajecten4,5,6,7,8. De humorale ingeboren immune reactie is, misschien wel, de beste gekenmerkt immuunsysteem verdediging in fruitvliegjes. Na infectie leidt de humorale respons op de productie en de systemische release van antimicrobiële peptide (AMP) moleculen in de Hemolymfe, het bloed gelijk bij insecten. Versterkers zijn geproduceerd door zeer geconserveerde tol en Imd signalering trajecten. De tol-traject is homoloog aan zoogdieren TLR/IL-1R receptor signalering en de Imd-traject is homoloog aan zoogdieren Tumornecrosefactor-alfa signalering. In Drosophila, tol signalering wordt geïnduceerd door gram-positieve bacteriën, schimmels en Drosophila X virus6,9,10 en Imd signalering wordt veroorzaakt door gramnegatieve bacteriën11 ,12.
Cellulaire immuniteit, samengesteld uit inkapselen, melanization en fagocytose van invasieve pathogenen, uitgevoerd door gespecialiseerde bloedcellen genaamd hemocytes13. Er zijn drie klassen van hemocytes in de fruitvlieg: crystal cellen, lamellocytes en plasmatocytes13. Crystal cellen, die make-up van 5% van het circulerende hemocytes in larven, vrij proPhenoloxidase (tot) enzymen leidt tot de melanization van pathogenen en host weefsels op wond sites. Lamellocytes, die niet normaal te in gezonde embryo's of larven vinden, zijn Adherente cellen die vreemde voorwerpen kapselen. Deze cellen worden veroorzaakt bij pupariation of bij parasitizing wasp eieren worden afgezet in de larven. Fagocytische plasmatocytes, die 95 procent van de circulerende hemocytes in larven en alle overige hemocytes bij volwassenen, een rol spelen in de weefsel remodeling tijdens de ontwikkeling en, met name, dienen als de belangrijkste effector cel van Drosophila cellulaire immuniteit.
Fagocytose een onmiddellijke en cruciale lijn van het aangeboren immuunsysteem verdediging; microben die inbreuk op de host epitheliale barrière zijn snel overspoeld en geëlimineerd door fagocytische cellen van het bloed (zie referentie 14voor een algehele herziening van de celbiologie van fagocytose). Dit proces wordt gestart wanneer germline-gecodeerde patroonherkenning receptoren (PRRs) op hemocytes herkennen pathogen moleculaire patronen (PAMPs) van microben verbonden. Eenmaal gebonden aan hun doelstellingen, initiëren PRRs signalering cascades die tot de vorming van pseudopods via het remodelleren van actine cytoskelet leiden. De pseudopods rondom de microbe, die vervolgens overspoeld en geïnternaliseerd in een ontluikende organel, de phagosome. Microben zijn vernietigd zoals de phagosome het proces van rijping van de phagosome ondergaat wanneer de phagosome is verhandeld naar de binnenzijde van de hemocyte en door middel van een reeks interacties met lysosomen verzuurt. In vitro en cel zijn biologie onderzoek bij zoogdiercellen primaire instrumentale bij de identificatie en karakterisering van de factoren die fagocytose, zoals de zoogdieren Fc-gamma receptor en C3b receptoren15,16 regelengeweest. Echter de mogelijkheid grote schermen of in vivo onderzoek uit te voeren zijn beperkt in zoogdieren systemen.
Hier presenteren we een in vivo assay voor fagocytose in volwassen fruitvliegen, die is gebaseerd op een procedure voor het eerst geïntroduceerd door het laboratorium van David Schneider in 200017. Het Schneider-lab bleek dat sessiele hemocytes geclusterd langs de buik dorsale vaartuig gemakkelijk phagocytose polystyreen kralen en bacteriën. Om te visualiseren fagocytose, vliegen worden geïnjecteerd met fluorescently geëtiketteerde deeltjes (zoals E. coli aangeduid met fluoresceïne-isothiocyanaat (E. coli -FITC)), gedurende 30 minuten tot de hemocytes tijd te verzwelgen de deeltjes, geïncubeerd en vervolgens ingespoten met trypan blauw, die de fluorescentie van deeltjes niet phagocytosed tijdens de incubatieperiode lest. Vliegen dorsale schepen zijn dan beeld met behulp van een omgekeerde fluorescente microscoop. Deze baanbrekende papier, met behulp van een relatief eenvoudig experiment, aangetoond dat hemocytes phagocytose bacteriën en latex kralen, dat bacteriële fagocytose kunnen worden geremd door het injecteren van vooraf vliegt met latex kralen, en dat zonder zowel cellulaire en humorale vliegt immuunresponsen zijn gevoelig zelfs voor E. coli. De bepaling in dit verslag gepresenteerde bouwt voort op het werk van de Schneider-lab te kwantificeren in vivo fagocytose door het meten van de intensiteit van de fluorescentie van deeltjes opgeslokt door dorsale schip verbonden hemocytes.
Vergelijkbaar met de aanpak van zoogdieren systemen, Drosophila genetici aanvankelijk gebruikt genoom-brede in vitro RNAi schermen ter identificatie van genen die nodig zijn voor de cellulaire immuunrespons18,19,20 ,21,22,23. Echter de ontwikkeling van de volwassen in vivo fagocytose assay ingeschakeld follow-up experimenten gemakkelijk in hele dieren, waardoor onderzoekers om te controleren of de biologische de rol van factoren geïdentificeerd in in vitro onderzoek uit te voeren. Dit was het geval met de transmembrane receptor Eater, die werd voor het eerst geïdentificeerd als een bacteriële receptor in een RNAi-scherm met behulp van S224 cellen en vervolgens later getoond te bemiddelen van Escherichia coli (E. coli), Enterococcus faecalis, en Staphylococcus aureus (S. aureus) fagocytose in volwassenen25.
Onze lab gebruikt de bepaling van de in vivo fagocytose in voorwaartse genetische schermen en genoom-brede vereniging studies (met behulp van de Drosophila genetische referentie paneel (DGRP)) om nieuwe genen die fagocytose in volwassen hemocytes regelen te identificeren. Deze studie leidde tot de karakterisering van de receptoren PGRP-SC1A en PGRP-SA26, de intracellulaire vesikel handel van proteïne Rab1427, glutamaat vervoerder Polyphemus28en RNA-bindende eiwit Fox-129.
Wij verwachten dat toekomstige schermen nemen de in vivo fagocytose kunnen leiden tot de identificatie van extra genen die belangrijk voor de cellulaire immuunrespons in Drosophila zijn. Schermen met behulp van gesequenced ingeteeldestammen, zoals de DGRP of de Drosophila synthetische bevolking Resource (DSPR), kunnen het identificeren van natuurlijke varianten op het gebied van fagocytose of hemocyte ontwikkeling. Bovendien, de techniek kon worden aangenomen bij andere diersoorten Drosophila of gebruikt voor scherm nieuwe communautaire middelen, zoals de collectie van 250 Drosophila soorten onderhouden door de nationale Drosophila soorten voorraad Center (NDSSC ) op Cornell. Bij alle dieren biedt aangeboren immuniteit een onmiddellijke en robuuste verdediging tegen een breed spectrum aan pathogenen. Humorale en cellulaire immuunresponsen zijn de belangrijkste takken van aangeboren immuniteit, en veel van de regulering van deze reacties factoren evolutionair tussen ongewervelden en zoogdieren zijn bewaard. Fagocytose, de centrale component van cellulaire aangeboren immuniteit, wordt uitgevoerd door gespecialiseerde bloed cellen van het immuunsysteem. De fruitvlieg, Drosophila melanogaster, heeft ontpopt als een krachtige genetisch model om de moleculaire mechanismen en de fysiologische effecten van fagocytose in hele dieren te onderzoeken. Hier laten we zien een in vivo fagocytose injectie gebaseerde bepaling om te kwantificeren van de particle opname en vernietiging door de cellen van het bloed van de Drosophila , hemocytes. De procedure kan onderzoekers te regelen het deeltje concentratie en dosis, waardoor het kan zeer reproduceerbare resultaten te verkrijgen in een korte hoeveelheid tijd precies. Het experiment is kwantitatieve, gemakkelijk uit te voeren, en kunnen worden toegepast om te screenen op gastheer factoren die invloed pathogen erkenning, opname en goedkeuring.

Deze in vivo fagocytosetest stelt onderzoekers in staat om genetische schermen en genoombrede associatiestudies uit te voeren om nieuwe genen te identificeren die fagocytose reguleren in volwassen bloedcellen. Dit experiment is kwantitatief, gemakkelijk uit te voeren en kan worden toegepast op de screening van levende dieren op gastheerfactoren die de herkenning, opname en klaring van ziekteverwekkers beïnvloeden. Na het trekken van dunne wand glazen haarvaten met een naaldtrekker, gebruik maken van een micrometer om de naald onder een microscoop te houden, en gebruik nummer vijf, fijn-punt, roestvrijstalen pincet om de tip te breken tot een 100-micrometer tip diameter.Om het volume van de vloeistof die zal worden geïnjecteerd in elke vlieg te meten, laad een capillaire naald met steriele 5%voedsel kleuring in PBS, en verdrijft de vloeistof op een druppel minerale olie op een 0,01-millimeter stadium micrometer. Doseer 10 microliter van 1,6 milligram per milliliter deeltjes op een klein vierkantje Parafilm en trek de vloeistof in de naald. Monteer de naald in de injector mondstuk, en lijn de verdoofde vliegen langs hun aangewezen gebied op het vliegkussen, ventrale kant omhoog, met de hoofden gericht op de voorkant van het pad.Plaats de flesjes in overeenkomstige gebieden op de bank, en injecteer de vliegen in de bovenhoek van de buik met vijf, 100-milliseconde pompen vloeistof om ongeveer 10 nanoplegers van deeltjes in totaal te leveren. Breng elke vlieg in de juiste flacon als het wordt geïnjecteerd, wijzend op de tijd op de flacon. Laad vervolgens een nieuwe naald met 0,4%Trypan Blue-oplossing en stel de pneumatische injector op gated, zodat een constante luchtstroom de vloeistof uit de naald kan duwen.30 minuten na de eerste injectie, injecteer elke vliegbuik met Trypan Blue totdat de buiken vol en opgezwollen zijn. Monteer de vliegen op microscoop dia's met elektrische tape, ventrale kant naar beneden, duwen de vleugels aan de zijkant van de vlieg om ze te beveiligen aan de tape. Duw vervolgens het hoofd voorzichtig in de tape om ervoor te zorgen dat de vlieg niet beweegt.Onmiddellijk nadat alle vliegen zijn beveiligd, beeld de insecten, een voor een, op een 25 of 32 keer vergroting op een omgekeerde fluorescentie microscoop bevestigd aan een digitale camera en computer, gericht op de dorsale schip van elke vlieg met behulp van de computer software voor de digitale camera. Noteer vervolgens de belichtingstijd en vergroting tussen experimenten. Om de fluorescentie te kwantificeren, opent u een geschikt beeldanalyseprogramma en opent u één afbeelding.Om de fluorescentieintensiteit van het dorsale vat te meten, trekt u een veelhoek rond het dorsale vat en selecteert u Meten om de fluorescentieintensiteit in de veelhoek vast te leggen. Om de intensiteit van de achtergrondfluorescentie te bepalen, kopieert u de eerste veelhoek en verplaatst u deze naar een gebied dat grenst aan het dorsale vat van elke vlieg. Selecteer vervolgens De fluorescentieintensiteit van het achtergrondgebied en noteren.Om de dorsale vat fluorescentie te normaliseren door de achtergrond fluorescentie, na het meten van de fluorescentie intensiteit van de rest van de vliegen, verdeel de dorsale vat fluorescentie door de achtergrond fluorescentie, en bereken de gemiddelde genormaliseerde dorsale vat fluorescentie intensiteit van alle vliegen in een stam. Na te zijn gemonteerd ventrale kant naar beneden op een stuk elektrische tape, de eerste twee segmenten van de buik, waar de rug vat is gevestigd, zijn duidelijk zichtbaar. Belangrijke bronnen van experimentele fout doen zich voor bij de injectie- en beeldvormingsstappen van de procedure.Met behulp van dezelfde naald om meerdere vliegen te injecteren kan leiden tot het verstopt raken met vliegweefsel of deeltjes. Vliegen die niet genoeg Trypan Blue fluorescentie krijgen in hun hele buik, wat de verhouding van het dorsale vat tot de achtergrondfluorescentie kan verminderen, waardoor de werkelijke fluorescentieintensiteit van het dier afneemt. Vliegen moeten worden gefotografeerd terwijl geïmmobiliseerd door kooldioxide, als actief bewegende vliegen produceren wazige beelden die niet kunnen worden gekwantificeerd.Een gemuteerde lijn uit de Zuker-verzameling methanesulfonaat behandelde vliegen is niet in staat om gramnegatieve en gram-positieve bacteriën te fagosten en vertoont bijna geen dorsale vaatfluorescentie in de in vivo fagocytosetest, gekoppeld aan de isogene zuker-stam op de achtergrond en een andere veel voorkomende laboratoriumcontrolestam. Houd de tijd en de volgorde bij waarin vliegen worden geïnjecteerd om ervoor te zorgen dat vliegen zich in vergelijkbare stadia van pathogene herkenning en opname bevinden wanneer ze worden afgebeeld. De moleculaire mechanismen die ten grondslag liggen aan fagocytische defecten kunnen worden bepaald door het bestuderen van enkele hemocyten met confocale microscopie of na fluorescentie-geactiveerde celsorling of door magnetische kraalisolatie van volwassen hemocyten.
