単細胞レベルでの細菌細胞の培養が注目を集めています。限られた空間内の単細胞レベルで細菌細胞を培養すると、表現型変動性1、2、3、4、細胞挙動5などの細菌機能を解明できる。6,7,8,9、および抗生物質耐性10、11。近年の培養技術の進歩により、単一細菌の培養は、ウェルチップ4、7、8、ゲル液滴12、13などの限られた空間内で達成することができる。、および水中油(W/O)液滴5、11.単一細菌細胞の理解や利用を促進するためには、栽培技術のさらなる技術開発が必要である。
生体細胞膜を模倣する小胞は、両生類分子からなる球状の区画であり、様々な物質を保持することができる。小胞はサイズに応じて分類され、小さな小胞(SV、直径<100 nm)、大きな小胞(NEV、<1 μm)、および巨大な小胞(GV、>1 μm)が含まれます。SVまたはNEVは、生体細胞膜14に対する親和性のために薬物担体として一般的に使用される。GVは、原始電池15または人工細胞16の構築のための原子炉システムとしても使用されている。生体細胞をGVに封入すると17、18、したがってGVは、反応器系と組み合わせると細胞培養システムとしての可能性を示す。

我々は、巨大小胞(GV)内の単細胞レベルで細菌細胞を培養する方法を開発した。細菌細胞培養は、自然環境における細菌細胞の機能を理解するために重要である。技術の進歩により、限られた空間内の単細胞レベルで様々な細菌細胞機能が明らかになります。GVは、親和性脂質分子からなる球状の微小サイズのコンパートメントで、細胞を含む様々な材料を保持することができます。本研究では、液滴転写法により10~30μm GVに単一の細菌細胞を封入し、細菌細胞を含むGVをガラス基板上の支持膜上に固定化した。本手法は、GV内の単一細菌のリアルタイム増殖を観察するのに有用である。大腸菌(大腸菌)細胞をGV内のモデルとして培養したが、この方法は他の細胞型に適応できる。微生物学、生物学、バイオテクノロジー、合成生物学の科学・産業分野で活用できます。

このプロトコルは、巨大小胞内の単細胞レベルで細菌細胞を培養する手段を提供する。巨大な小胞を開発するのは簡単で、小胞を準備するために必要なサンプル溶液はごく少量です。この手順のデモンストレーションは、当研究室の博士候補生である太田百合です。まず、20マイクロリットルの新しく調製したPOPC溶液と4マイクロリットルの新たに調製したビオチン-PEG-DSPE溶液をガラスバイアルに添加します。脂質膜が形成されるまで空気流により有機溶媒を蒸発させる。フィルムを乾燥剤に1時間入れ、有機溶媒を完全に蒸発させます。その後、バイアルに200マイクロリットルのミネラルオイルを加えます。プラスチックパラフィンフィルムでバイアルの開口部をカバーし、少なくとも1時間の120ワットで超音波浴中のバイアル内容物を超音波処理します。小さな小胞の調製のために、ちょうど示したように、脂質膜を作成し、フィルムにLB培地で200ミリメートルのグルコースの200マイクロリットルを追加します。その後、少なくとも1時間120ワットでフィルムを超音波処理し、その後、100ナノメートルの孔サイズのミニ押出機とポリカーボネート膜を使用して、得られた巨大な小胞を小さな小胞に押し出します。細菌細胞前培養の場合、LBプレートから37°Cで一晩培養するためにLB培地のミリリットルに接種大腸菌。翌朝、培養物の上清の20マイクロリットルを1.98ミリリットルの新鮮なLB培地に移し、さらに2時間培養する。インキュベーションの終了時に、分光光度計上の培養前溶液の光学密度を600ナノメートル(OD600)で確認します。1~1.5のOD 600を含む培養前溶液を使用する必要があります。次に、作りたてのショ糖溶液と新鮮なLB培地と共にプレカルチャー溶液を混合する。巨大な小胞形成のために、1.5ミリリットル蓋付きプラスチックチューブに巨大な小胞の新しく調製された外水溶液の50マイクロリットルを追加します。巨大小胞の外水溶液の上に脂質含有油溶液の150マイクロリットルを穏やかに層化する。得られた溶液を室温で10~15分間インキュベートし、オイルと水溶液の界面が平坦であることを確認します。液滴形成を含む水および油菌細胞の場合、0.6ミリリットルの蓋付きプラスチックチューブに超音波処理された脂質含有油溶液の50マイクロリットルに、調製したばかりの内水溶液の2マイクロリットルを巨大小胞に加えます。次に、チューブをタップして2つの成分を乳化します。次に、ピペットを使用して、油および水溶液の界面に50マイクロリットルの水および油滴溶液を添加し、遠心分離によって細菌細胞含有巨大小胞を沈降させる。その後、トップオイル層を吸引し、巨大な小胞を収集します。手作りのチャンバーを準備するには、10対10で中空のパンチで7ミリメートルの穴を1ミリメートル両面シールで掘削し、2面シールを30~40ミリメートルのカバーガラスに貼り付けます。サポートされている二層膜を調製するには、手作りのチャンバーの穴に小さな小胞溶液の30マイクロリットルを加えます。室温で30分間小胞をインキュベートします。インキュベーションの終わりに、200ミリミラーグルコースを添加したLB培地20マイクロリットルで穴を2回静かに洗います。サポーターの二層膜に巨大な小胞を固定化するには、外水性巨大小胞溶液中の10マイクロリットルのノイトラビジンを室温で15分間インキュベーションする穴に導入する。インキュベーションの終わりに、20マイクロリットルのLB培地で2回、200ミリミラーのグルコースを添加し、巨大な小胞溶液の全容をチャンバーの穴に加えて、穴を2回静かに洗浄します。その後、18 18ミリメートルのカバーガラスで穴を密封します。巨大小胞内の細菌の増殖を監視するには、反転顕微鏡上で40Xの目的を選択し、顕微鏡加熱段階システム上にチャンバーを置きます。その後、37°Cで6時間静条件下でチャンバー内の細菌細胞含有巨大小胞をインキュベートし、30分ごとに細菌細胞増殖の画像をキャプチャして記録し、科学的相補的な金属酸化物半導体カメラで撮影します。細菌細胞を含む巨大小胞は、通常、10〜30マイクロメートルの大きさの範囲である。巨大小胞の両方の大きさについて、大腸菌細胞は伸びおよび分裂プロセスを経て、一部の大腸菌細胞は6時間の観察期間中に非常に多くの細胞に増殖する。単一の細胞レベルを含む巨大小胞の相対的な頻度は、得られた巨大小胞の約10%であり、単一細胞レベルでカプセル化された巨大小胞は、細菌細胞を含む巨大小胞の約50%である。油水界面の安定性は、細菌細胞を含む巨大な小胞を得るために重要である。巨大な小胞矯正の前に油を慎重に吸引するように注意してください。このビデオを見た後、あなたは個々の巨大な小胞内の単一の細胞レベルで細菌細胞を培養する方法をよく理解しているはずです。当社の細菌培養法は、未知の環境細菌を培養して代謝産物を得たり分析したりするための微生物学の新しいツールです。
