1. 材料和动物
<ol><li>准备所需材料: 75% 乙醇、胶带、脱毛剂、2毫升管、1.0 mL 注射器与针头连接 (26G)、电子秤、肝素和盐水 (见材料表)。</li>
	<li>动物: 准备10只昆明小鼠, 6-8 大, 体重21.6-28.3 克 (见材料表)。根据《实验动物护理和使用指南》对小鼠进行维护。</li>
</ol>2. 麻醉和动物定位
<ol><li>称重鼠标以计算所需的麻醉剂用量。</li>
	<li>通过腹腔注射戊巴比妥钠 (60 mg/kg) 诱导全身麻醉 14 (图 1).在手术开始时使用无菌眼液中的润滑软膏, 以防止对暴露的眼睛造成损害。 注: 当没有运动反应的踏板退出反射, 尾巴捏, 或腹部皮肤捏, 被认为是充分麻醉。</li>

</ol>3. 锁骨下静脉穿刺和采血
<ol><li>用手指识别锁骨骨和高级胸骨窝的位置。</li>
	<li>找到靠近锁骨骨中间的穿刺部位, 并将其尾端到上。将左手食指侧向穿刺部位, 固定皮肤和皮下组织 (图 3)。</li>

本报告是以往通过大鼠11层下静脉穿刺进行血液取样研究的延伸.由于老鼠是最常用的研究动物, 看看这种技术是否也能应用于老鼠, 这将是很有价值的。挑战来自于锁骨下静脉的直径相对较小。
在目前的研究中, 我们发现小鼠锁骨下穿刺是一种可行、可靠的采血方式。与传统的方法, 如尾静脉切割或眼眶血液收集相比, 目前的方法不存在伦理问题。所有动物在这个过程后生存了没有液体补充剂大约200μl 的血液提取, 并且动物可用于进一步的实验研究。从理论上讲, 每次总样本量提取不能超过循环血总量的 1 0%, 成年动物的总体积为 5 5 至 7 0 mlkg 体重5。对于本研究中使用的小鼠 (21.6-28.3 克), 最大体积提取量应在200μl 左右。因此, 我们将每只老鼠的血液采样量设置在200μl 左右。无法测量血压、心率和其他压力参数是本报告的主要局限性。
充足的全身麻醉是保证穿刺成功的另一个重要问题。小鼠在穿刺过程中应保持安静, 以避免被针头伤害血管的风险。我们发现, 五巴比妥钠在 60 mgkg 的全身麻醉可以满足理想的麻醉深度。此代理的缺点是恢复时间相对较长 (平均约 30分钟)。在一些研究所, 吸入异氟醚的恢复时间很短, 应考虑作为替代麻醉选择15。
锁骨下静脉的位置如图 4所示。由于静脉的薄壁和低压, 在针头向前移动时, 锁骨下静脉的前壁和后壁会在针头的压力下相互附着 (图 5A)。当慢慢向后取出柱塞时 , 连接的壁将自发分离 , 针尖可以进入血管的真正腔 , 血液可以在持续的负压入注射器 (图 5B) 。使用真空血液收集系统16也可能有利于血液采集。由于小鼠锁骨下静脉的直径相对较小, 即使是有经验的操作人员也可能出现故障。对于新手, 我们建议在大鼠中穿刺锁骨下静脉, 如报告11。在大鼠多次成功穿刺后, 小鼠锁骨下静脉穿刺成功率可显著提高。确保确切的穿刺位置只是锁骨骨中间的尾端。针头向上移动, 颅骨向上移动到上胸骨窝是保证成功穿刺的其他关键点。
总之, 锁骨下静脉穿刺是一种安全有效的小鼠采血方法。该方法除尾静脉切割和后眼眶丛吸血外, 还具有可行性、安全性, 适用于小鼠在多个时间点的观测研究。

在大多数研究实验室中, 从小鼠身上采集血液样本是必不可少的。小鼠采血的传统方法是在需要小于100μl 的样本时进行尾切1。然而, 如果在非末端时间点需要超过100μl 的血液, 眼眶后、下腔出血或胸下血液收集是最常用的技术2。在某些情况下, 采用颈静脉导管插入通过手术切口作为另一种方法3。
然而, 上述方法对小鼠有害。据我们所知, 由于潜在的并发症风险4,5,后眼眶法没有被广泛接受。操作相关的创伤不仅发生在可见区域6、7, 而且也发生在轨道深处6。此外, 下颌下采血压力为 8 , 可能与出血过多2,9有关。在我们之前的研究10,11的基础上, 我们介绍了一种新的策略, 从锁骨下静脉收集小鼠。介绍并讨论了该技术的安全性、可行性和获得的血量。

老鼠是研究人类疾病和人类健康的最重要的哺乳动物模型。然而, 在研究工作中, 从小鼠身上采集血液样本具有挑战性。当需要少量的血液样本时, 尾巴采血是一种流行的方法。如果需要大量的血液, 可以考虑轨道动脉, 但这种采血方法有伦理问题。以前, 我们通过大鼠锁骨下静脉穿刺法证明了采集血样的可行性和安全性, 并在此研究了该方法是否可用于小鼠。我们报告说, 这种方法对小鼠采血是安全和实用的。在日常研究工作中, 通过锁骨下静脉穿刺采血是一种简便的方法。

我们在10只昆明小鼠身上重复了这个过程 (雄性 n = 5, 雌性 n = 5, 体重25.4±2.0 克)。有九个程序在右侧成功。一个案件在3次尝试之内失败了在右边和血液收集成功在左边。时间过程 (从穿刺到获得所需的血量) 在 30-126秒 (平均684±26.4 秒) 之间。采血量设置为200Μl 左右 (平均 203±11.6μl)。血液收集在1到4次尝试之内成功了。血液采样成功在第一次尝试在两只小鼠和在一次小鼠的4次尝试以后;取样在其他小鼠身上进行了2-3 次尝试。所有数据见表 1。所有动物在血液取样后30分钟内存活和恢复。在所有观察到的参数中, 性别之间没有明显差异 (表 1)。
