1. 素材と動物
<ol><li>必要な材料を準備:75% エタノール、粘着テープ、epilating 剤、2ml チューブ、1.0 は、針 (26G)、電子スケール、ヘパリンおよび生理食塩水に接続されたシリンジ (材料の表を参照してください)。</li>
	<li>動物:10 昆明マウスを準備し、6-8 週齢、重さ 21.6 ~ 28.3 g (資料表参照)。実験動物13のケアおよび使用のためのガイドに従ってマウスを維持する。</li>
</ol>2. 麻酔と動物のポジショニング
<ol><li>麻酔薬の必要な投与量を計算するために、マウスを計量します。</li>
	<li>全身麻酔14を誘導するために腹腔内注射を通じて、ペントバルビタール (60 mg/kg) のナトリウムを注入する (図 1)。露出した目への損傷を防ぐためにプロシージャの開始で生殖不能の目の潤滑の軟膏を加えなさい。 注: マウスは、ペダル離脱反射、尾ピンチ、または腹部の皮膚ピンチのテストに対してモータ応答を示さない場合に十分に麻酔されると考えられる。</li>

</ol>3. 鎖骨下静脈穿刺と採血
<ol><li>指で鎖骨骨と優れた胸骨窩の位置を特定します。</li>
	<li>鎖骨の真ん中に近い穿刺部位を見つけます。左の人差し指を穿刺部位に置き、皮膚および皮下組織を固定する (図 3)。</li>
	<li>針を上方に移動し、優れた胸骨窩に向かって cranially ます。針が皮下組織に入ると、右手の薬指を後方に動かして陰圧を形成する (図 3)。 注: このステップでは、オペレータの右手の位置は、針を水平面より上方に移動させるために、操作テーブルよりもわずかに低くする必要があります。動物がテーブルの端の近くに配置されている理由です。</li>
	<li>シリンジを前方に 3-4 mm 移動しますが、シリンジに血液が流れない場合は停止します。その後、ゆっくりとシリンジを引き戻し、その中に負圧を保持します。ほとんどの場合、血液は、戻って描画するときにシリンジに入ります。</li>

この報告は、ラット11における鎖骨下静脈穿刺を介した血液採取に関する以前の研究の延長を表している。マウスは、最も一般的に使用される研究動物であるように、この技術はまた、マウスに適用することができるかどうかを確認することは有益であろう。課題は、鎖骨下静脈の比較的小さい直径に由来する。
本研究では、マウスの鎖骨下穿刺が血液を採取するための実現可能で信頼できる方法であることを発見した。尾静脈切断や軌道採血などの従来の方法と比較して、本方法の倫理的問題はない。すべての動物は、約200μ l の血液抽出後に液体サプリメントなしでこの手順の後に生き残った、そして動物はさらなる実験的研究のために使用することができた。理論的には、合計サンプル量の抽出は、毎回の総循環血液量の 10% 以上に達することができず、成人動物の総体積は55〜 70 mL/kg 体重5である。この研究で使用されるマウス (21.6-28.3 g) については、最大量の抽出は約200μ l でなければなりません。このようにして、各マウスの血液サンプリング量を200μ l 前後に設定した。血圧、心拍数、その他のストレスパラメータを測定できないことは、このレポートの主な制限事項です。
十分な全身麻酔は、穿刺の成功を保証するもう一つの重要な問題である。マウスは、針によって血管を傷つけるリスクを避けるために、穿刺手順の間は静かなままでなければならない。我々は、全身麻酔に 60 mg/kg でペントバルビタールナトリウムが麻酔の理想的な深さを満たすことができることを発見しました。このエージェントの欠点は、比較的長い回復時間 (平均約30分) です。一部の研究所では、イソフルラン吸入が短い回復時間のために使用され、代替麻酔選択15として考慮されるべきである。
鎖骨下静脈の位置を図 4に示す。細い壁と静脈の低圧のために、鎖骨下静脈の前と後壁は、針が前方に移動している間に針の圧力の下で互いに付着するであろう (図 5a)。プランジャーをゆっくりと後方に引き出す一方で、アタッチされた壁は自発的に分離し、針の先端が血管の真の空洞に入り、血液がシリンジに持続的な負圧の下で流出する可能性があります (図 5b)。真空採血システム16の使用はまた、血液採取を支持してもよい。マウス鎖骨下静脈の比較的小さい直径のために、経験豊富なオペレータに対しても障害が生じることがある。初心者のために、報告された11としてラットに鎖骨下静脈穿刺を推奨する。ラットにおいていくつかの成功した穿刺の試みの後、マウスにおける鎖骨下静脈穿刺の成功率は、有意に増強され得る。正確な穿刺位置が鎖骨骨の中央のちょうど尾部であることを確認してください。上方および頭蓋を優れた胸骨窩に移動する針は、穿刺の成功を保証するための他の重要なポイントである。
結論として、鎖骨下静脈穿刺は、マウスにおける血液採取のための安全で効果的な方法である。尾静脈の切断およびレトロ軌道神経叢の血のコレクションのほかに、この方法は可能、安全、およびマウスの複数の時点の観察的な研究のために適している。

マウスからの血液サンプル採取は、ほとんどの研究室において不可欠である。マウスにおける採血のための従来のアプローチは、試料の100μ l 未満が必要な場合のテール切断である1.しかし、非終端の時点で100μ l 以上の血液が必要とされる場合、眼窩後方、顎下出血または submental 採血は、最も一般的に考えられる技法2である。いくつかの機会において、外科的切開を通して頸静脈のカテーテル法が、代替方法3として採用された。
それにもかかわらず、上記の方法は、マウスに有害である。私たちの知識の最大のために、眼窩後方法は、合併症の潜在的なリスクのために広く受け入れられていません4,5.操作関連の外傷は、可視領域6、7だけでなく、軌道6内でも深く起こる。また、顎下血液採取はストレス8であり、過剰な出血2,9に関連している可能性があります。我々の先行研究10,11に基づいて、ここではマウスの鎖骨下静脈からの血液採取のための新しい戦略を紹介する。この技術を用いて安全性、実現性、および得られた血液量を提示し、議論する。

マウスは人間の病気と人間の健康を研究するための最も重要な哺乳類モデルです。しかし、マウスからの血液サンプル採取は、研究作業において困難である。尾血のコレクションは少量の血のサンプルが必要とされるとき普及した方法である。大量の血液が必要な場合には、軌道動脈を考慮することができますが、この採血方法には倫理的問題があります。以前は、ラットの鎖骨下静脈穿刺による血液検体採取の実現性と安全性を実証し、マウスでこの方法が使用できるかどうかを調べた。我々は、この方法は、マウスの採血のための安全かつ実用的であることを報告する。マウスの鎖骨下静脈穿刺による採血は、日常の研究作業において簡便な方法となり得る。

我々は、10昆明マウス (雄 n = 5、メス n = 5、重量25.4 ± 2.0 g) でこの手順を繰り返した。右側には9つのプロシージャが成功しました。1つのケースが右側で3回の試行で失敗し、左側に採血が成功しました。時間経過 (穿刺から必要な血液量を得るまで) は35-126 秒 (平均68.4 ±26.4 秒) の範囲であった。採血量は、約200μ l (平均203±11.6 μ l) に設定した。採血は 1 ~ 4 回以内に成功した。血液サンプリングは、2匹のマウスでの最初の試みと1匹での4回の試みの後に成功した。サンプリングは、他のマウスで2-3 の試みを要した。すべてのデータを表 1に示した。すべての動物は生き残ったし、血液サンプリング後30分以内に回復しました。すべての観察されたパラメータについて、男女間に有意差はなかった (表 1)。

マウスの鎖骨下静脈穿刺による採血。マウスからの血液サンプル採取は、ほとんどの研究室で不可欠です。マウスにおける採血のための従来のアプローチは、100マイクロリットル未満のサンプルが必要な場合に、尾切断である。しかし、非終末期の時点で100マイクロリットル以上の血液が必要な場合、レトロ眼窩下出血、下顎下出血、または心外の採血が最も一般的に考えられる技術である。いくつかの場所では、頸静脈切開またはカテーテル法が代替方法として採用された。それにもかかわらず、上記の方法はマウスに傷ついています。正式な研究に基づいて、ここではマウスの鎖骨下静脈からの採血のための新しい経済的で穏やかな戦略を紹介します。安全性と実現可能性、およびこの技術で得られた血液量が提示され、議論される。全身麻酔後、仰向けの位置にマウスを横たえる。75%エタノールで破砕空間を洗浄してください。穿刺前に鎖骨と上耳窩を確認します。鎖骨の真ん中に穿刺部位を特定します。針を上方に移動し、鎖骨の後部に向かって上向きに動かします。3~4ミリメートル前方に移動した後、注射器に負圧を入れて、ゆっくりと針を引き戻します。血液は撤退の過程で注射器に排出されます。採血後、穿刺部位を1~2分間柔らかく押して出血を止める。75%エタノール、粘着テープ、上着剤、チューブ、針で接続された1.0ミリリットルのシリンジ、電子スケール、ヘパリン、および生理食塩水を含む必要な材料を準備します。満足のいく全身麻酔の後、手術台の上にマウスを置き、テーブルの端から2〜4センチメートル離れて静脈穿刺を容易にします。図 2 に示すように、手足を快適な位置に固定します。全手順で挿管や機械的換気は必要なかった。綿棒で破膜腔の周りに上膜剤を塗布する。3分後、上流剤を湿った綿棒で洗い、毛皮と目に見える汚れを取り除きます。75%エタノールで破包領域を殺菌し、清潔なガーゼで乾燥させます。鎖骨の骨と上耳のフォッサエの位置を1本の指で特定します。穿刺部位を見つけ、鎖骨の真ん中に近づき、その上に引っ掛かります。皮膚および皮下組織を固定するために穿刺部位に左人差し指を横に置く。頭蓋の上方に針を移動し、上の胞子窩に向かって置きます。針が皮下組織に入ると、右手の薬指は後方に移動して陰圧を形成する必要があります。注射器を3~4ミリメートル進めるが、注射器に血液が流出しない場合は停止する。その後、ゆっくりとシリンジを引き戻し、その中に負圧を保ちます。ほとんどの場合、血液は引き戻しの過程で注射器に入ります。血液が注射器に入ったら、注射器を固定し、注射器に採取された血液量が必要になるまで負圧を保ちます。採血後、穿刺針を引き出し、綿棒で穿刺部位を1~2分間わずかに押して出血を止め、マウスをケージに送り返します。血液サンプルをヘパリンチューブに移します。我々は、10個のキミマウスでこの手順を繰り返した。9つの手順が右側で成功しました。1つのケースは右側の3回の試みで失敗し、左側で採血に成功した。タイムコースの範囲は35~126秒でした。採血量は約200マイクロリットルに設定した。採血は1~4回の試みで成功した。血液採取は2匹のマウスの最初の試みで成功し、1匹のマウスで4回の試みで、他のマウスでは2〜3回の試みで成功した。すべてのデータを表 1 に示します。すべての動物は生き残り、血液サンプリング後30分以内に回復した。すべての観察されたパラメータの性別に有意差はなかった。結論として、鎖骨下静脈穿刺は、マウスの採血のための安全で効果的な方法である。尾静脈切断およびレトロ眼窩神経叢採取に加えて、この方法は、マウスのいくつかの時点での観察研究に適しており、適当である。
