enrichment of native lipoprotein particles with microrna and subsequent determination of their absolute/relative microrna content and their cellular transfer rate

脂蛋白颗粒是原生运输工具。外国物质的浓缩便于它们用作履带载体。分子或整个颗粒的特定标记使得测量细胞吸收成为可能。两种浓缩方法快速且易于使用，可适用于各种物质。演示这个程序将是马库斯·阿克斯曼和安德烈亚斯·卡纳，两个博士后从我的实验室。要开始在烟气罩中脱脂，在锥形离心管中混合一至两毫升含有5毫克高密度脂蛋白颗粒的预制高密度脂蛋白溶液，并加入50毫升预冷却的三至两种乙醇二乙醚混合物。在负20摄氏度下孵育两小时。在负10摄氏度下以2500倍g离心10分钟。丢弃上清液，将颗粒重新放入50毫升预冷却乙醇二乙醚混合物中，并短暂地旋转。在负20摄氏度下再孵育两小时。在负10摄氏度下，以2500倍g再次离心10分钟。然后插入供应氮气流量的油管，以干燥颗粒。并在250微升缓冲液 A.中重新暂停，使用布拉德福德蛋白质测定或其他适当的蛋白质测定确定蛋白质浓度。去除乙醇二乙醚混合物的任何残留物至关重要，因为这些溶剂会抑制非脂蛋白的重新生长。接下来，稀释至每250微升缓冲液中1毫克蛋白质的最终浓度。如果需要，在惰性气体大气中将溶液在四摄氏度下过夜。为了开始重组，在干净的玻璃管中，混合100微升CO、13.5微升C和500微升PC。然后旋转玻璃管，同时冲洗管内的氮气以干燥混合物，以产生均匀的表面层。在0.5毫升反应管中，在缓冲液中准备一个新的30毫升精氨酸溶液，然后在两毫升反应管中将100微升10微摩尔合成微RNA与100微升精氨酸溶液混合。在30摄氏度下孵育30分钟。接下来，将孵育的200微升溶液转移到玻璃管中，以补充准备好的PC-CO-C主混表面层。然后加入50微升，每毫升30毫克脱氧钠溶液。在四摄氏度下搅拌两个小时。之后，将250微升的脱脂高密度脂蛋白溶液加入玻璃管。并在四摄氏度的一夜之间搅拌。要开始透析，首先在800毫升双蒸馏水中加入50克吸附珠。并使用磁力搅拌器搅拌一分钟。等待 15 分钟，让珠子沉淀下来，然后将上清液去。使用预冷却的 PBS 重复此过程。PBS 中的预湿透析盒。根据制造商的说明，使用注射器将先前准备的混合物添加到盒式磁带中。将 PBS 处理的吸附珠添加到三升 PBS 中，并将盒式磁带放入 PBS 中，在 4 摄氏度下进行拨号。珠子保持透析膜的密度梯度恒定。一小时两小时后更换缓冲液和珠子。24小时后，用注射器将溶液从盒式中提取到1.5毫升反应管中，以回收重组后的HDL颗粒溶液。使用布拉德福德测定确定蛋白质浓度。向反应管提供惰性气体，将其密封，并在四摄氏度的惰性气体大气中储存重组的 HDL 颗粒溶液。首先，在无RNAAse水中准备一个新的30毫升精子溶液。在两毫升反应管中，将100微升的10微摩尔合成微RNA与100微升的精氨酸溶液混合。在30摄氏度下孵育30分钟。然后将100微升DMSO和1.2毫升1X LDL缓冲液添加到准备好的微RNA精子溶液中。使用 PBS 稀释以前准备的 LDL 颗粒溶液，最终浓度约为每毫升 4 毫克。然后将稀释溶液的450微升抽入1.5毫升反应管中，与50微升10X LDL缓冲液混合。在冰上孵育10分钟。孵育后，将500微升LDL颗粒溶液和1.5毫升微RNA-精氨酸-DMSO溶液结合，在40摄氏度下孵育两小时。执行与前面描述类似的透析，并在四摄氏度的惰性气体气氛下将标有 LDL 颗粒溶液的存储。首先，在PBS中稀释1到100和1-1，000之间的HDL或LDL颗粒溶液。将云母压在云母基材上，然后拔下胶带以去除上部云母层，将云母切割。使用移液器将两个微升剂沉积在刚切割的云母上，孵育五分钟。脂蛋白颗粒往往在表面上形成连续粘膜。因此，调整云母表面的颗粒浓度以获得单独的浓度非常重要。孵育后，用PBS冲洗样品。用 PBS 填充高速 AFM 液体电池，将携带云母的扫描仪安装到高速 AFM 级。在控制软件中，开始悬臂到云母表面的接近过程。使用小于一平方千米的扫描尺寸，并保持成像力尽可能低。在攻丝模式下对样品进行成像。将数据加载到 Gwydion 中，然后通过标记颗粒按阈值函数检测颗粒。调整高度阈值以屏蔽单个粒子。通常，50%左右的水平是一个很好的起点。删除多项式背景以拼合图像，并激活排除蒙版区域选项。使用各种颗粒特性函数的分布导出检测到粒子的最大高度值，并对所有记录的图像重复这些步骤。在这个实验中，通过去除高密度脂蛋白颗粒，然后进行重新皮化和透析，对HDL粒子进行重组。可达到50%的重组高密度脂蛋白颗粒的产生率。然而，由于脂蛋白B100蛋白的疏水性，与微RNA相同的标签对于LDL颗粒的标签是不可行的。因此，DMSO 用于LDL颗粒的脂质单层渗透，在质量控制期间，其产量接近100%，稀释对于分析至关重要。顶部图像显示的粒子密度过高。底部图像适合分析。计算粒子高度的概率密度函数，以便比较原生、标记和控制脂蛋白粒子的大小分布。标签程序之前和之后的相对变化是不容忽视的。处理RNA寡核苷酸时，无RNA酶工作。使用新鲜的一次性塑料耗材，并始终戴上手套。仅使用无核酸酶溶液。高速AFM只是确定脂蛋白一般形状的一种方法。另一种方法是电子显微镜。佩戴适当的个人防护设备，在处理二乙醚时在烟罩中工作。
