注：对于这些实验，我们使用雄性C57Bl/6J小鼠，体重在25克至30克之间，年龄为8-12周。老鼠在常规条件下喂食小颗粒和水。外科设备以前已经消毒。实验者应戴上无菌手术手套和手术面罩，以限制污染和术后感染。
1. 麻醉和气管罐。

</ol>2. LAD冠状动脉的结扎
<ol><li>从胶带上松开左前爪，小心地将鼠标移到右侧的脱皮位置。一旦动物处于正确位置，固定左前肢。</li>
	<li>识别左胸肌小肌肉和主要肌肉之间的线，并在1厘米上用剪刀在线条后进行倾斜的皮肤切口。用解剖钝微剪刀，单独的胸肌筋膜没有切口。保持胸肌分开，用收缩器连接到弹性带。</li>
	<li>将呼吸机设置为 3 cm H2O 的正端呼气压力 （PEEP）。</li>
	<li>在第三和第四肋骨之间的第三个中间空间使用钝钳打开胸腔。避免接触内胸动脉，因为有出血的危险。不要触摸心脏或肺。将两个缩回器放入肋骨中，每个肋骨上一个（图2A）。</li>

	<li>找到左前下降（LAD）冠状动脉。LAD 动脉显示为一条浅红色的浅线，从左侧动脉边缘向顶点跑去。</li>

</ol>3. 术后程序和后续行动。

心肌缺血再灌注（I/R）是一种相关的手术模型，模仿在诊所对MI患者进行的冠状血流恢复。在I/R模型中，冠状动脉的瞬态闭塞是通过将一根管子拧紧到LAD上，持续20至45分钟8，13。然后释放闭塞，使心肌在所需的持续时间内重新灌注。这种简单的修改应用于我们的协议可以很容易地把它变成I/R模型4，8，14，15。心肌梗死可以通过心脏肌钙蛋白T8、10的血液测试或回声心电图15进行确认。
MI 与 I/R 模型不同，因为再灌注本身会导致损伤。MI诱导更多的组织坏死和凋亡在重新融合心肌5中更为明显。在MI和IR中，炎症细胞渗透的动力学也不同，MI7免疫细胞的心肌渗透延迟。在永久结扎和I/R模型15之间，梗塞区域的大小和位置也会有所不同。请记住，由于 I/R 和永久 MI 模型不等效，因此在选择相关模型时必须谨慎。心肌梗死的另一个动尿模型是低温心肌模型。在LV前壁上应用低温探针可诱导LAD动脉中心室组织和血流抑制的冻结。然而，这种技术不同于MI和I/R技术关于改造和炎症反应的时间和振幅16，17。
变异性是任何外科手术的一个限制。这种变异性依赖于生物差异。一个很好的例子是小鼠11的冠状动脉排列的变化。它还依赖于实验者的技能。值得一提的是，为了达到这一模型的稳定结果，必须对实验者进行适当的培训。训练有素的实验者可以很容易地产生可重现的梗塞尺寸（图3A-B）。模型的死亡率取决于LAD的位置、实验的持续时间（天、周）、小鼠菌株和基因型。麻醉和镇痛药物的类型也可能影响具有假定性心脏保护或心抑郁作用的实验结果。在我们手中，这种模式的全球死亡率为25-30%。亡率包括实验结束前的自发死亡和牺牲，无论菌株和实验持续时间如何。大多数死亡或牺牲是在手术后的第二天和第四天之间。对动物实行严格的疼痛管理和随访可以降低死亡率。
在这里，我们介绍使用TTC染色和表达蛋白质和基因在LV中分别由西方印迹和实时PCR参与的炎症或纤维化过程的梗塞大小代表性的结果（图3C-G）。也可以通过酶连结免疫吸附测定（ELISA）或酶测定法测量许多这些参数。当然，根据需要测试的假设，这种方法可以遵循任何功能分析，通过超声波，MRI或室内导管测量压力和体积。也可以提取心脏，并进一步研究分离细胞的心脏细胞生物学。总体而言，具有LAD冠状动脉永久结扎的MI模型对于评估炎症和纤维化过程、伤口愈合和心肌梗死后心脏功能的变化特别有用。

急性心肌梗死（MI）是缺血性心脏病（IHD）最严重的表现。IHD是全世界，特别是在西方国家，疾病和死亡的主要原因。因此，它对医疗体系产生了巨大的经济影响。MI的特点是动脉冠状动脉被动脉粥样硬化斑块遮挡，随后在心肌的大部分地区抑制血流。心肌缺氧导致心肌缺血死亡。这种病理状况触发心室组织的反应，最终导致心室功能缺陷，重塑和心力衰竭3。MI 是一种复杂的病理生理学条件，涉及多个复杂的生物过程，包括调节性细胞死亡、对氧化应激的反应、炎症、伤口愈合、纤维化和心室重塑。其中一些生物反应被建模为体外个体过程，如坏死引起的损伤相关分子模式和相关炎症反应释放4。这些简化的模型对于理解 MI 至关重要。然而，只有体内模型才能提供对MI反应的生物过程复杂性的真实图像。
尽管在大型动物（如猪）中的MI模型可能与MI的人类病理生理学有更密切的联系，但鼠模型的力量在于基因工程提供的可能性，这种可能性比任何其他哺乳动物物种都更先进。其他不可忽视的方面是相对低廉的成本和手术设置的简单性。
值得一提的是，心肌缺血再灌注的模型可以表现出与永久MI模型不同的结果。生物过程，如细胞死亡的类型，质量/振幅或动能的炎症和伤口愈合反应在心肌组织可能根据模型5，6，7有所不同。然而，这种永久冠状动脉闭塞方案可以很容易地适应，以获得缺血再灌注模型。
该方法与MI的理疗学相关研究不进行再灌注，允许监测从冠状动脉闭塞（分钟）到晚期心力衰竭（周）在当地心脏组织和全身发生的病理过程水平。

murine myocardial infarction model using permanent ligation of left anterior descending coronary artery

支持炎症信号通路和亲炎基因的表达通常在心肌梗死后的第一周内被激活。NF-βB p65转录因子的磷酸化是炎症的标志，在MI小鼠的整个心肌提取物中观察到（图3E）。通过实时PCR（图3G）对亲炎基因Il1b、Il6和Cxcl10（图3F）和单核细胞/巨噬细胞标记Cd14和Mertk的mRNA表达进行了分析。请注意，NF-βB p65 和 SMAD2 磷酸化的范围存在变异性（图 3C、E、通道 4-7）。这种变异性很大程度上取决于梗死的大小。
