Detta förfarande ger upprepad optisk tillgång till dorsala hippocampus av levande möss att studera mekanismerna för minnesbildning och minns medan de utför inlärningsuppgifter. Denna teknik gör det möjligt att använda ljusmikroskopi för att studera dorsala hippocampus av levande möss på mikrometernivå rumslig upplösning longitudinellt under flera veckor. Minnesförlust är ett allmänt kännetecken för vissa hjärnsjukdomar som Alzheimers sjukdom.
Förstå mekanismen för minnesbildning och återkallande kan i slutändan hjälpa patienter som lider av dessa sjukdomar. Denna metod skulle kunna tillämpas på andra djursystem med en skalle som små däggdjur. Övervakning av vitala tecken under en överlevnadsoperation kan vara både störande och stressande.
Det kan vara bra att först utföra förfarandet på ett dött djur för att fokusera på de mer avgörande aspekterna av förfarandet. För att förbereda bildbehandling kanylen, först skära en tre millimeter diameter rostfritt stål rör i en 1,6 millimeter lång metallring. Om kanterna på ringen inte är trubbiga efter styckning, fil ut oegentligheterna.
Därefter använder ett par tlysen för att doppa ena sidan av metallringen i ett UV-härdande optiskt lim. Placera sedan metallringen i mitten av ett glasöverdrag med den sida av ringen som täcks av självhäftande röra vid täckslip. Slå på UV-härdnings-LED-förarenheten och lysa 365 nanometer våglängdsljus på limmet i en minut för att bota limmet.
Se till att ringens alla sidor är lika upplysta genom att ändra ljuskällans riktning. Efter att limmet har härdat i minst två timmar, håll fast kanylen från den öppna änden av metallringen med hjälp av en hemostat. Med hjälp av en tandborr med en roterande fil, fila bort överskottet glas coverslip tills den spolas med sidorna av ringen.
Förbered de instrument som krävs och bedöva musen enligt beskrivningen i det medföljande textprotokollet. Ta sedan bort håret och desinficera huden över mushuvudet. Därefter använder sax och tika för att göra ett litet snitt i hårbotten i positionen nära lambda.
Flytta i följd i riktning mot öronen sedan rostrally i riktning mot ögat kretsar för att bilda en triangel på sagittal axeln cirka fyra millimeter rostral till bregma. Applicera en enda droppe lidokain på skallen. Efter två minuter, rensa benhinna och torka skallen med hjälp av en bomullspinne.
Placera sedan öronstängerna för att fixera musens huvud. Med hjälp av en mikroborr med en 0,5 millimeter bredd burr, gör ett litet hål i frontalbenet mittemot den avbildade hippocampus cirka 1,5 millimeter från sagittal sutur och två millimeter distala från korona suturen. Skruva sedan en 0,86 millimeter bredd rostfritt stål benskruv i skallen hålet.
Säkra den på plats med hjälp av limcement. Låt cementen torka i 30 sekunder till en minut. Därefter använder en tre millimeter diameter trefinborr för att göra ett litet hål i parietalbenet.
Placera hålet ungefär 1,5 millimeter distala från sagittal sutur och två millimeter distala från lambdoid suturen. Ta försiktigt bort benfliken. Ta sedan bort hjärnhinnorna med hjälp av Dumont-forceps.
Ablate kortikal materia för att nå den yttre kapseln. Använd en 19 gauge trubbig nål ansluten till en vakuumpump och irrigering med saltlösning för att undvika uttorkning av den exponerade vävnaden och tvätta bort eventuellt kvarvarande blod efter att blödningen är löst. Sug kortikala vävnad långsamt ca 50 till 100 mikrometer i taget tills cortex lossnar från kapseln utsätta fibrerna i cingulum eller corpus callosum.
Sedan försiktigt skala dorsala fibrer tills de djupaste fibrerna, alveus av hippocampus är utsatta. När du är klar, skölj vävnaden med saltlösning. Därefter använder tunna tivnip för att doppa botten kanyl i saltlösning och placera den över skallen hålet.
Tryck sedan in kanylen i skallen tills glasöverdrag är i kontakt med fibrerna. Torka skallen och applicera en snabb klistercement över skallen för att hålla kanylen på plats. Var noga med att även applicera lim på kanylens fälg men var noga med att inte låta limmet köra in i kanylen.
När du är torr, använd en stereotaxisk arm för att placera en huvudhållareplatta över kanylen i kontakt med skallen. Förbered en blandning av dental akryl och sedan tillämpa den över hela kraniet. Täck hela exponerade skallen, skruven och den öppna huden med dental akryl för att stabilisera preparatet.
Efter 15 minuter, applicera en avtagbar självhäftande film på huvudhållarplattan för att förhindra att skräp kommer in i kanylen. När redo att bilden hjärnan, återigen följa med i den medföljande textprotokoll för detaljer om anesthetization. När tillräckligt sövd, använd tivetter för att försiktigt ta bort den självhäftande filmen från huvudhållarplattan.
Ta bort filmen försiktigt för att undvika att preparatet skadas. Därefter placerar du musen under mikroskopet över värmemattan och fäst huvudplattan till hållaren. Applicera ögonsalva på djurets ögon.
Rengör sedan avbildningskanylen genom att använda en spruta och en tunn nål för att släppa avjoniserat vatten i kanylen följt av borttagning med en vakuumpump. Använd låga förstoringslånga arbetsavståndsmål för att visuellt kontrollera kanylen för restvatten, smuts, integritet och förekomst av fluorescens. Rikta sedan in kanylen mot den optiska axeln genom att justera huvudhållararmarnas vinklar.
Byt till ett 25X-mål med en 1,0-numerisk bländare och ett fyra millimeters arbetsavstånd. Tillsätt sedan tillräckligt avjoniserat vatten till kanylen för att fylla det och underhålla överflödigt vatten ovanpå kanylen. Slutligen, använd två foton excitation och bild fluorescenssignalerna.
Visas här är en 2P bild stack från en thy1-GFP transgena mus hjärnan. Thy1-GFP möss uttrycka cytoplasmatisk förbättrad GFP under kontroll av Thy1 promotorn i en gles slumpmässig population av pyramidala nervceller. Generellt är den stora axeln av pyramidala nervceller i dorsala hippocampus CA1 ungefär vinkelrätt mot XY imaging planet.
Dessa regioner mappas manuellt till en låg förstoring tredimensionell stack som visar mönstret för GFP-uttryck i volymen under avbildning kanyl. För longitudinell spårning definieras flera hjärnregioner inom kanylfältet. Varje region motsvarar en yta på cirka 240 gånger 240 mikrometer och innehåller mellan ett och sju dendritiska segment.
Den avbildade serien här visar en mikrometer z-steg bild staplar av CA1 pyramidala nervceller och basala dendrites förvärvats vid olika tidsintervall i 14 dagar. Längre avbildningsnvaraktighet och intervaller är dock möjliga. Även om de flesta bilder är av dendritiska taggar i stratum oriens, är det också möjligt att bilden dendritiska spines i sneda dendriter av stratum radiatum.
En annan förlängning av denna teknik är att bilden aktivitet-evoked plasticitet i CA1 pyramidal nervceller genom att injicera dorsala CA1 hippocampus område med en viral vektor. Detta möjliggör plasticitet hundratals CA1 pyramidala nervceller i varje djur som skall mätas visas här som en 2P bild stack. Det är avgörande att förhindra skador på hippocampus när du tar bort den överliggande vävnaden.
Dessutom bör man placera kanylen korrekt för att säkerställa en stabil förberedelse. Den beskrivna beredningen möjliggör användning av olika slags optisk avbildning såsom miniatyrmikroskop som kan implanteras på djurets huvud. Detta gör det möjligt för forskaren att följa neuronal aktivitet i fritt rörliga djur.
Ursprungligen användes tekniken för att studera strukturell plasticitet i hippocampus nervceller. Idag, Det genomförs i att studera neuronal aktivitet även i andra områden hjärnan.
Denna metod beskriver en kronisk beredning som tillåter optisk tillgång till Hippocampus levande möss. Denna beredning kan användas för att utföra longitudinell optisk avbildning av neuronala strukturella plasticitet och aktivitet-framkallat cellulär plasticitet under en period av flera veckor.
