脾臓移植のマウスモデルは、炎症性疾患における局所および全身性免疫応答の調節における脾細胞の役割を明らかにする絶好の機会を提供する。このプロトコルの主な利点は、コンジェニックマウスを使用することにより、脾臓移植は、脾細胞の運命、長寿、および機能を確認することが可能になるということです。処置を開始する前に、外科用プラットホームの上に無菌の使い捨てカーテンを置き、麻酔されたドナーマウスのつまみつまみへの応答の欠如を確認する。腹部全体の毛髪を剃り、6~10倍の拡大で手術顕微鏡の下で無菌手術プラットフォームにマウスを置きます。アルコールの準備パッドで露出した皮膚を殺菌し、外科用テープで手足を固定します。恥骨からキシフォイドプロセスまでの3〜4センチメートルの正中垂直皮膚切開を介して腹部を入力します。皮膚血管を焼灼し、両側の側腹部に切開を拡張して、より良い露出を達成する。無菌綿棒を使用して、腹部の右脇腹に腸を移動し、脾臓を露出させ、無菌の低温焼灼を使用して脾臓に付着した短い胃静脈を焼灼する。脾臓の上に37度の摂氏生理食音を浸した滅菌ガーゼを置き、しっとりして膵臓組織からポータル静脈を分離して動員します。示されているように、ポータルの静脈の枝をリゲートします。脾静脈から遠位の門脈の周りに縫合糸を置き、脾臓を右側に反転させ、大動脈と脾動脈をセリアックトランクに露出させます。大動脈セリアック脾動脈を解剖し動員し、セリアック動脈に近位大動脈の周りに縫合を配置する。ヘパリンの100国際単位を下の大静脈またはIVCに注入して全身をヘパリン化する。3分後、大動脈の肝動脈と胃動脈をセリアック動脈に近い高位にリゲートする。門脈を透過し、腹部大動脈遠位からセリアックトランクまで摂氏4度ヘパリン化生理食塩水の10ミリリットルで全身を透過する。脾臓の静脈のセグメントと膵臓組織の小さな部分と一緒にポータル静脈に関連する大動脈セリアック脾臓セグメントと脾臓の移植片とブロックを収集します。その後、4度の生理食塩水の5ミリリットルで移植片を保存します。ドナーのために外科領域を準備した後、中線切開を介してレシピエント腹部を入力し、脾静脈と動脈を連結することによってレシピエントネイティブ脾臓を除去する。37°C浸したガーゼで腸組織を覆い、慎重に腸を左側に移動します。IVCの不フラレナル大動脈の腰部を解剖してリゲートし、IVC内の不フラレナル大動脈を2つの4ミリリットルの微小血管クランプでクロスクランプする。11-0ナイロン縫合糸を不全大高を通し、容器を引き込んでマイクロシザーで楕円形の大所切開術を作成できるようにします。30ゲージ針を使用してIVCを突き刺して楕円形毒毒を作成し、ドナーポータル静脈の長さに開口部を延長します。500マイクロリットルのヘパリン化生理食塩水で大動脈とIVCを洗い流し、血管から内アルミニウム血液または血栓を取り除き、ドナー脾臓移植片をレシピエント腹部の右脇腹に入れます。ドナー大動脈カフとポータル静脈を慎重に特定します。血管がねじれていないことを確認した後、4度の摂氏生理用ガーゼで脾臓移植片を覆う。2つの11-0の滞在縫合糸を使用して、ドナー大動脈カフをレシピエント大動脈切除術の近位および遠位頂点に接続し、ドナー大動脈カフとレシピエント大動脈の前壁との間の連続した11〜0ナイロン縫合糸の2〜3回の咬傷で解剖学を行う。脾臓移植片をレシピエントの左側に回し、ドナー大動脈カフとレシピエント大動脈切除術の後壁との間の吻合を行う。ドナー門脈とレシピエントIVCの間で静脈吻合を行い、両側に4~5回の連続縫合糸を付ける。最初に容器の内腔内の後壁を縫合し、次に同じ縫合糸を使用して前壁を閉じる。IVC の外側の下隅に結び目を配置して、吻合を完了します。今度は容器クランプを解放し、脾臓の色が回復するまで出血をタンコネードするために無菌綿棒を使用する。連続パターンで5-0合成吸収性Vicryl縫合糸で腹部を閉じ、中断されたパターンで5-0ナイロン縫合糸で皮膚層を閉じます。その後、250マイクロリットルの温かい生理食塩水を4つの別々の場所に注入し、動物が家庭ケージに移すのに十分な意識を取り戻すまで、最初の数時間の摂氏温度制御インキュベーターでマウスを30°Cの温度制御インキュベーターで温めます。移植後1日目と7日目の脾臓同種移植片のヘマトキシリンおよびエオシンは、脾臓同種移植片のアーキテクチャが術後最初の週中にそのまま残っていることを明らかにする。移植後のドナー脾臓細胞の移行を調べるフローサイトメトリック解析は、移植後1日に脾細胞の約50%がドナー由来であり、約46%がレシピエント由来であることを示している。移植から7日後、ドナー由来の白血球は脾臓細胞全体の約32%を占め、レシピエント由来細胞はリンパ節、血液、骨髄に早ければリンパ節、血液、骨髄に移行し、少なくとも7日目まで維持され、脾細胞密売研究に価値のあるユニークなキメラを生成する。遺伝子組み換えノックインマウスを脾臓ドナーとして使用することで、脾臓細胞由来メディエーターまたは疾患プロセスにおける主要なシグナル伝達経路の役割の調査が可能になる。このモデルは、病原体、傷害、炎症、または移植拒絶反応に応答して脾臓細胞集団のメカニズムを探索するための強力なツールである可能性があります。
