薬物アフィニティ応答性標的安定性、DARTSは、新規の低分子タンパク質標的の検出のための堅牢な方法である。既知の小分子タンパク質相互作用を検証し、天然物の潜在的なタンパク質標的を見つけるために使用することができます。本研究では、タンパク質安定性の変化をモニタリングし、タンパクリガンド相互作用の親和性を推定することで、DARTS実験のデータ解析能力をさらに強化しました。タンパク質とリガンドの相互作用は、タンパク質分解曲線と用量依存曲線の2つの曲線に対してプロットすることができます。プロトコルの確立のための模範的なケースとして mTOR-ラパマイシン相互作用を使用しました。10%のウシ胎児血清、2ミリモルグルタミン、および1%の抗生物質を用いてDMEMを使用して293T細胞を成長させます。5%の二酸化炭素の下で摂氏37度で培養をインキュベートする。培養が80~90%合流した場合、細胞を冷たいPBSで2回洗浄する。細胞スクレーパーを使用して、細胞を適切な量の冷たい細胞のライシスバッファーに集め、1.5ミリメートルのチューブに細胞を移します。反転して、細胞をライシスバッファーとライシングをよく混合し、チューブを氷の上に10分間インキュベートします。チューブを摂氏4度で10分間18,000倍に遠心します。上清を新しい1.5ミリリットルチューブに移し、氷の上で冷やします。BCAアッセイを実行して、溶解物のタンパク質濃度を近似し、タンパク質濃度に基づいてプロナーゼの希釈を計算します。まず、ライセートを2つの1.5ミリリットルチューブ、それぞれ99マイクロリットルに移します。溶解液の各アリコートに小分子ストック溶液の1マイクロリットルを追加し、揺れで室温で30〜60分間2つのチューブをインキュベートします。氷の上で、1x TNCで解凍した新しいプロナーゼ溶液の連続希釈を確立する。小分子でのインキュベーションに続いて、各アリコートを5本のチューブで20マイクロリットルのサンプルに分けます。同じ消化時間を確保するために、30秒間隔で、調製されたプロナーゼ溶液の2マイクロリットルをそれに応じてサンプルに加える。1つの制御グループに対して、2マイクロリットルのTNCバッファを追加します。5~20分後、30秒間隔で2マイクロリットルのコールド20xプロテアーゼ阻害剤カクテルを添加して6本のチューブの消化を停止する。よく混ぜて氷の上で10分間インキュベートします。5倍のSDS-PAGEローディングバッファの6マイクロリットルで各サンプルを希釈し、10分間摂氏70度の水浴でサンプルを沸騰させます。さて、ウェスタンブロットを実行するには、適切な分子量マーカーと一緒に、8%SDS-PAGEゲルのウェルに等量のタンパク質をロードします。80ボルトで30分間ゲルを実行し、電圧を120ボルトに調整し、1〜2時間稼働し続けます。小分子が潜在的な標的タンパク質に直接結合できることを確認します。このプロトコルでは、小分子とのインキュベーションは、タンパク質分解に対する保護を確認した。3つのバンドは、車両制御上のラパマイシンとのインキュベーションによって保護されていることが判明した。ウェスタンブロッティングは、タンパク質比に対する低プロナーゼにおけるmTORタンパク質の存在と、その減少および損失を増加比率と共に示した。プロナーゼによるmTORのプロテオライシスはラパマイシンの存在によって明らかに阻害され、ラパマイシンの添加はタンパク質分解曲線の明らかな変化を生じた。ラパマイシン用量依存的にmTORのレベルを高め、ラパマイシン治療によるmTORの安定性の上昇を示唆した。標的タンパク質バンド強度の定量化は、mTORがラパマイシンの標的タンパク質であることを示唆した。最良のステップワイズ効果を得るためには、この実験は、タンパク質比に対して適切な範囲のプロナーゼを得るために数回繰り返さなければならない場合がある。
