Endotheliale cellen worden vaak gebruikt in onderzoek van vasculaire dysfunctie, diabetes, weefselregeneratie, transplantatie, en kankers14,15,16,17,18. Om de biologie en functie van ECs in deze ziekten te begrijpen en te karakteriseren, zijn er talrijke methoden om de ECs van verschillende zieke organen of weefsels te isoleren, gemeld8...

Het tekort aan beschikbare organen voor transplantatie is een uitstekend probleem wereldwijd1. Volgens de Rode Kruis vereniging van China, slechts een klein aantal patiënten met eindstadium orgaan falen kreeg een geschikt orgaan in China in 2018.
Xenotransplantatie is een veelbelovende manier om het probleem van orgaan tekorten op te lossen. Varkens organen worden beschouwd als de meest geschikte organen voor de mens vanwege anatomische en fysiologische gelijkenissen2,3. Het falen van een varkens xenotransplantaatmodellen is is grotendeels gerelateerd aan de primaat immuunafstoting en coagulatie responsen. Varkens endotheel cellen (ECs) zijn kritisch, omdat deze cellen zijn de eerste om te communiceren met het primaat immuunsysteem, waaronder antilichamen, complement, cytokines, en immune cellen (bv, T-cellen, B-cellen, en macrofagen)4,5. Varkens ECs spelen een vitale rol in varkens orgel en eilandje xenotransplantatie6,7. Dus, ECs zijn belangrijke cellen voor het onderzoeken van de afwijzing en coagulatie reacties op een varken Graft. Isolatie van hoge kwaliteit varkens ECs is vereist voor xenotransplantatie onderzoek.
In pogingen om ECs te isoleren van verschillende organen (bijv. hart, nieren, lever en aorta), zijn verschillende protocollen gerapporteerd in een xenotransplantaat instelling8,9,10,11,12. Echter, het houden van een ultrapuur cultuur van geïsoleerde ECs is een uitstekend probleem in de standaardprotocollen. De verhoogde concentratie van de verteerde oplossing, de ongepaste digestie tijd en de intensiteit van de Schraap kunnen bijdragen aan de verhoogde fibroblast besmetting in de huidige studies8,10,13. Trouwens, de methode van geïsoleerde pAECs van miniatuur varkens is minder bestudeerd. Hier beschrijven we een geoptimaliseerde enzymatische methode om hooggezuiverde pAECs te isoleren van miniatuur varkens (Wuzhishan of Bama). Verschillende stappen in het protocol zijn ontworpen om fibroblast besmetting te verminderen en hoge zuiverheid ECs te verkrijgen.

Xenotransplantatie is een veelbelovende manier om het tekort aan menselijke organen op te lossen voor patiënten met eindstadium orgaanfalen, en het varken wordt beschouwd als een geschikte orgaan bron. Immuun afwijzing en coagulatie zijn twee belangrijke hindernissen voor het succes van xenotransplantatie. Vasculaire endotheliale cel (EC) letsel en dysfunctie zijn belangrijk voor de ontwikkeling van de ontsteking en coagulatie reacties bij xenotransplantatie. Zo is isolatie van varkens aorta endotheel cellen (pAECs) noodzakelijk voor het onderzoeken van de immuunafstoting en coagulatie reacties. Hier hebben we een eenvoudige enzymatische aanpak ontwikkeld voor de isolatie, karakterisering en uitbreiding van zeer gezuiverde pAECs van miniatuur varkens. Eerst werd het miniatuur varken verdoofd met ketamine, en een lengte van aorta werd weggesneden. Ten tweede werd het endotheliale oppervlak van aorta blootgesteld aan 0,005% Collagenase IV digestieve oplossing voor 15 min. ten derde werd het endotheeloppervlak van de aorta in slechts één richting met een celscraper (< 10 keer) geschuurd en niet gecomprimeerd tijdens het proces van Schrapen. Tot slot, de geïsoleerde pAECs van dag 3, en na passage 1 en passage 2, werden geïdentificeerd doorstroming cytometrie met een anti-CD31-antilichaam. De percentages van CD31-positieve cellen waren respectievelijk 97,4% ± 1,2%, 94,4% ± 1,1% en 92,4% ± 1,7% (gemiddelde ± SD). De concentratie van Collagenase IV, de spijsverterings tijd, de richting, en de frequentie en intensiteit van het schrapen zijn van cruciaal belang voor het verminderen van fibroblast besmetting en het verkrijgen van hoge zuiverheid en een groot aantal ECs. Concluderend, onze enzymatische methode is een uiterst effentieve methode voor het isoleren van ECs van het miniatuur varken aorta, en de cellen kunnen in vitro worden uitgebreid om de immuunrespons en coagulatie reacties bij xenotransplantatie te onderzoeken.

Onze methode is bedoeld om een effectieve manier te bieden om zeer gezuiverde ECs te isoleren van de aorta's van miniatuur varkens. Het proces van aorta chirurgie wordt getoond in Figuur 1. De methode is een zeer effectieve methode voor het isoleren van endotheelcellen van miniatuurvarken. En de cellen kunnen ook worden uitgebreid, onderzoeken de immuun- en stollingsrespons bij xenotransplantatie. Het belangrijkste voordeel van techniek is dat het niet alleen gemakkelijk, zeer effectief is om zeer gezuiverde endotheelcellen te isoleren van varkens, maar ook beter om andere pAC's te houden wanneer deze worden doorgegangerd.Na bevestiging van een gebrek aan reactie op pijnreflex bij het verdoofde varken, was de varkensbuikwand drie keer met 75% medische alcohol en een tinctuur jodium. Na de laatste wasbeurt, gebruik een scalpel om een abdominale incisie te maken om de inferieure vena cava bloot te stellen, en gebruik een vijf milliliter spuit om 5000 eenheden heparine natrium te injecteren in de inferieure vena cava. Na vijf minuten, steek een katheter in de abdominale aorta, en gebruik een scalpel om het middenrif in te ringen.Verwijder met behulp van een tweede onderzoeker de linkerventrikel en gebruik bot tangen om de ribben uit te roeien om het hart en de longen bloot te leggen. Zoek de aorta achterste naar het hart en de longen, en klem de uiteinden. Gebruik een schaar om de aorta uit te roeien en was deze een keer met een voorgekoelde wasbuffer.Verwijder overtollig weefsel rond de aorta met steriele tangen en scharen en plaats het vat in een 50 milliliter centrifugebuis met verse voorgekoelde wasbuffer voor overdracht naar het laboratorium. Voor varkensatheek endotheelcelisolatie, breng de varkensaorta in een 150 millimeter petrischaal onder aseptische omstandigheden, en verwijder voorzichtig beide uiteinden van het vat. Snijd extra overtollig weefsel waar het vat was geklemd, met steriele tangen en scharen, en gebruik 20 tot 50 milliliter verse wasbuffer om de buitenkant en binnenkant van de aorta te spoelen.Vervolgens passeert u een chirurgische hechting door de aorta en bind losjes het ene uiteinde van het vat, waarbij de hechting in het lumen blijft. Bevestig de aorta voorzichtig in de buurt van het gebonden uiteinde met tangen en trek vervolgens de chirurgische hechting langzaam, om de aorta om te keren, totdat het endotheeloppervlak van de aorta wordt blootgesteld. Was het endotheeloppervlak van de aorta drie keer met 10 milliliter verse wasbuffer per wasmachine en plaats de aorta in een 15 milliliter centrifugebuis.Bedek het monster met 10 milliliter van 37 graden Celsius opgewarmd 0,005% collagenase IV spijsverteringsoplossing, en incubeer het weefsel op 37 graden Celsius gedurende 15 minuten. Breng aan het einde van de incubatie de gehele buisinhoud over in een 100 millimeter kweekschotel en arresteer de spijsvertering met 10 tot 15 milliliter vooraf gekoelde stopbuffer. Verwijder met behulp van een celschraper voorzichtig de aorta-endotheelcellen van het binnenoppervlak van het vat en was het vat drie keer met vijf milliliter wasbuffer per wasmachine.Na de laatste wasbeurt, breng de supernatant in een 50 milliliter centrifuge buis, en was de bodem van de cultuur schotel twee keer met vijf milliliter verse wasbuffer, per was, trekken de wast in de 50 milliliter buis. Verzamel de cellen door centrifugatie, en gooi alle, behalve de laatste 10 milliliter supernatant. Voeg 20 milliliter wasbuffer toe om de pellet opnieuw op te hangen, zorg ervoor dat bellen worden vermeden en verzamel de cellen met een andere centrifugatie.Resuspend de pellet in een milliliter van cel cultuur medium voor het tellen, en plaat de cellen op een keer tien tot de zes cellen per 25 centimeter vierkante kolf concentratie. Plaats vervolgens de cellen in de celkweek incubator op 37 graden Celsius in 5%kooldioxide, verfrissende het medium om de twee tot drie dagen. Na isolatie worden de endotheelcellen geïnspecteerd op dag nul, één en twee, en na passages één en twee op besmetting en hun morfologische evaluatie.Flow cytometrische analyse met behulp van een anti-CD31-FITC antilichaam geeft een typisch groter dan 97% endotheelcel marker positieve populatie op dag drie van de cultuur, dat wordt gehandhaafd op een meer dan 90%zuiverheid, zelfs na twee passages. Het endotheeloppervlak van de aorta werd in slechts één richting geschraapt, met een celschraper, minder dan 10 keer, en het werd niet gecomprimeerd tijdens het proces van schrapen. Beste antwoord, geoptimaliseerde methode van pAECs isolatie, kunnen we gezuiverde pAC's gebruiken om individuele experimenten uit te voeren en de immuunafstoting en stolling respons in xenotransplantatie te onderzoeken.
