この空気作動性マイクロ流体圧縮装置は、成長プレート軟骨細胞の3Dヒドロゲル培養におけるメカノバイオロジーを研究するために使用することができる。マイクロ流体デバイスは、少量のサンプルに対して複数の大きさの圧縮を同時に生成できるため、コストと時間効果が高く、さまざまな顕微鏡イメージングをデバイスと共に適用できます。我々の方法は、他の細胞型のメカノバイオロジーの研究にも応用できる。マイクロチャネル層をセットアップするには、まず10個の前重合体を1つの硬化剤に対して1つの硬化剤に対して1つの分子量比でPDMSを5分間混合する。次に、フォームパッドに金型を置き、マスター金型に混合物を注ぎます。PDMSを真空チャンバー内で30分間脱気し、脱気PDMSを透明フィルムで挟みます。サンドイッチされたアセンブリをガラス板、フォームパッド、プレキシガラスプレートでクランプします。PDMSを摂氏80度で6時間硬化させ、PDMS層と透明フィルムを挟み構造から分離します。PDMSの表面とプラズマクリーナー付きのクリーンガラス板を1分間アクティブにしてから、PDMSを80°Cのオーブンでガラス板に30分間接着します。その後、透明フィルムを除去する。薄いPDMS膜の調製のために、スピンコートは1分間に1,000回転毎に透明膜上でPDMSを硬化させず、60マイクロメートル厚いPDMS層を得た。プラズマクリーナーを使用してガラス板上のマイクロチャンネルPDMS層と薄いPDMS層を1分間活性化する前に、スピンコートPDMSを80〜30分間、80〜30分間部分的に硬化させます。その後、薄いPDMS膜層をマイクロチャネルPDMS層に接着し、80°Cのオーブンに一晩置きます。チューブブロックの準備のために、金属チューブをシャーレの上に垂直に置き、チューブの約3/4が水没するまで、未硬化のPDMSを皿に静かに注ぎます。真空チャンバーでPDMSを30分間脱気した後、60°Cで6時間以上オーブンでPDMSを硬化させます。硬化工程の終わりに、1本のチューブを含むPDMSブロックを切り取り、入口用の薄いPDMS層に穴を開けます。PDMSブロックとプラズマクリーナーで薄いPDMS層をアクティブにします。1分後、PDMSブロックを薄いPDMS層の入口穴に取り付け、作動ユニット全体をオーブン内に一晩で摂氏80度に置きます。アガロースゲル型製剤の場合は、脱イオン水に5%アガロースと200ミリモルの塩化カルシウムを混ぜ、沸騰するまでアガロースゲル溶液を加熱します。ゆでたアガロースゲル溶液をアルミニウムモールドに注ぎ、ガラス板でモールドを挟みます。5分後、固化したアガロースゲルをアルミニウムモールドから取り出します。150マイクロリットルのアルギネート軟骨細胞溶液をアミノシラン化ガラス板にプレートし、アガロースゲルモールドで3分間溶液を覆う。インキュベーションの終わりに、カミソリの刃を使用して過剰なアルギン酸ゲル溶液を除去し、プレートからアガロースゲル型を除去する。その後、アルギネート軟骨細胞の構成体を、50ミリモルの塩化カルシウムと140ミリモルの塩化ナトリウムを1分間脱イオン水に含む架橋溶液に入れます。装置を組み立てるには、4つの1ミリメートル厚いPDMSスペーサーを、作動部の薄いPDMS層の4つのコーナーに配置する。軟骨細胞培養培地の700マイクロリットルで薄いPDMS層の空気室をカバーします。ステレオマイクロソペを使用して、アルギネート-コンドロサイト構造を薄いPDMS層に配置し、構造を空気室に合わせるように注意し、3Dプリントクランプでデバイスをクランプします。装置の作動のために、ソレノイド弁の入口と空気ポンプの出口を接続するためにシリコン管の部分を使用する。追加のチューブを使用して、ソレノイドバルブの出口を組み立てられた装置の入口と接続します。ソレノイドバルブを関数ジェネレータに接続し、関数ジェネレータによって生成された 1 つのヘルツ方波を使用してソレノイドバルブを操作します。その後、空気ポンプをオンにして、デバイスを空気圧的に作動させます。マイクロ流体軟骨細胞圧縮装置は、5つの配列の円筒アルギネート-コンドロサイト構築物を含み、これらの構造は5つの異なる大きさの圧縮で圧縮することができる。この例では、ゲルカラムを最大PDMSバルーンによって高さ33.8%圧縮し、ゲル構築物の結果として生じる圧縮歪みは0.2ミリメートル増分あたり約5%増加し、PDMSバルーン直径が増加した。軟骨細胞の圧縮変形は、ゲル構築センター付近の体積を613で613で613で40~55マイクロメートルの体積で撮像することによって求めた。ここでは、最大PDMSバルーンによって16%圧縮された軟骨細胞の画像が示されている。このグラフでは、測定された細胞圧縮歪み値の分布が観察できる。この分析では、より大きなPDMSバルーンによって細胞をより全体的に圧縮した。これらのデータをまとめて、アルギン酸ゲルおよび軟骨細胞圧縮の量は、14キロパスカルの一定の圧力下でPDMSバルーンの直径によって制御されることを示唆している。デバイスの性能の再現性を確保するためには、一定の厚さと弾力性を持つ薄いPDMS膜を作成することが重要です。この手順に従って、免疫蛍光およびその蛍光中のハイブリダイゼーションのような種々の生物学的アッセイが行うことができる。この技術を用いて、成長板や関節軟骨細胞におけるメカノトランスダクションに関する多くの疑問に答えることができる。
