마이크로 엔지니어링 플랫폼은 물리적 및 화학적 미세 환경모두의동적 제어를 가능하게하기 때문에 분자, 세포 및 조직 수준 생물학을 연구하기위한 귀중한 도구입니다1,2,3 ,4,5,6,7,8. 따라서, 여러 가설은 동시에 엄격하게 통제된 방식으로 시험될 수 있다. 성장판 연골의 경우,성장판 연골9,10,11에대한 작용을 통해 뼈 성장을 조절하는 데 있어 압축 스트레스의 중요한 역할에 대한 증거가 증가하고있다. 12,13,14,15,16 ,17,18,19,20, 21,22,23,24,25. 그러나, 압축 응력의 작용의 메커니즘 - 특히, 스트레스가 성장 판에 연골 세포 기둥의 형성을 안내하는 방법 - 제대로 이해되지 않습니다.
이 프로토콜의 목적은 성장판 연골세포에서 메카노바이오틱의 메커니즘을 해명하기 위해 공압작동 미세유체 연골세포압축장치(26)를 만드는것이다(도 1a-c). 이 장치는 공압 작동 장치와 알긴산 겔 구조의 두 부분으로 구성됩니다. 미세 유체 공압 작동 장치는 사진 및 연리소그래피에 기초하여 폴리디메틸실록산(PDMS)을 사용하여 제작됩니다. 이 장치에는 직경에 따라 다르게 팽창 할 수있는 얇은 PDMS 멤브레인 풍선의 5 x 5 배열이 포함되어 있습니다. 상기 알긴산 겔 구조는 5 x 5 배열의 알긴산 겔 실린더에 내장된 연골세포로 구성되며, 전체 알긴산-콘드로시트 구조는 작동 장치와 함께 조립된다. 알긴산 겔 구문은 공압적으로 팽창된 PDMS 풍선에 의해 압축된다(도1b). 미세 유체 장치는 PDMS 풍선 직경의 차이에 따라 단일 플랫폼에서 동시에 다섯 가지 수준의 압축 응력을 생성할 수 있습니다. 따라서, 다중 압축 조건 하에서 연골세포 메카노생물학의 고처리량 시험이 가능하다.
본 프로토콜에 기재된 미세유체 장치는 외부픽서(14,21,23 및 거시적 압축장치)와같은 종래의 압축 장치에 비해 많은 장점을갖는다. 19세 , 27세 , 28 연골세포 메카노생물학을 연구하기 위한: 1) 미세유체 장치는 거시적 압축 장치보다 적은 양의 샘플을 소비하기 때문에 비용 효과적이며, 2) 미세 유체 장치는 여러 개의 샘플을 테스트할 수 있기 때문에 시간 효율적입니다. 압축 조건 동시에, 3) 미세 유체 장치는 마이크로 채널의 제한된 혼합에 기초하여 화학 물질의 농도 구배를 형성하여 기계적 및 화학적 자극을 결합 할 수 있으며, 4) 다양한 현미경 기술 (시간 경과) 현미경 검사법 및 형광 공초점 현미경 검사법)은 투명 PDMS로 만들어진 미세 유체 장치로 적용 할 수 있습니다.
