1. 未分化人类胚胎干细胞的维持和培养
<ol><li>在无进纸器条件下,通过预涂带特定细胞外基质的洗碗,对 hSEC 进行维护和扩展。<ol>
<li>在 4°C(典型范围浓度 18-22 mg/mL,保持冰上)下解冻 300 μL 的细胞外基质,并轻轻与 15 mL 的冷 DMEM 介质混合,以避免细胞外基质过早凝固。在两个T150烧瓶中加入7.5 mL的细胞外母体。</li>

</ol>2. 用于 GBM 研究的 hESC 衍生神经器官

	<li>在12.5 mL的无血清培养基中制备28.2 x 106个细胞,辅以10μM ROCK抑制剂。分配1000个细胞/微井。在300 x g下将细胞离心5分钟,并将板置于37°C的培养箱中过夜(最大36小时)。例如:要获得30个人类神经器官,在汇合70%-80%(约3000万个细胞)处使用一个T150烧瓶。</li>
	<li>第二天(第 1 天),收集球体(带 P1000),并将其放入 6 孔板中。在每口井中,加入2 mL的B27介质和DMEM-F12谷氨酸和神经碱基(混合在1:1),辅以1%B27补充剂和1%非必需氨基酸(NEAA)。为了促进快速神经诱导,补充介质双SMAD抑制鸡尾酒,由10μM TGF®/Activin/诺达尔抑制剂和0.5μM骨形态蛋白(BMP)抑制剂组成。从这一步开始,球体以旋转方式培养(60 rpm,轨道摇床)。旋转对于防止球体粘在一起或粘在板上至关重要。</li>

		<li>从第21-25天起,在同一神经成熟培养基培养人类神经器官(参见步骤2.7.4)。</li>
		<li>从第25-28天起,只需用1μMβ-分泌酶抑制剂补充B27介质。</li>
		<li>从第28-39天起,停止添加β-分酶抑制剂,仅在B27培养基中继续人体神经器官培养。 注:3周后,神经器官可用于GIC植入。在神经成熟时,观察到神经不成熟标记Nestin的减少和成熟神经标记+3-tubulin和GFAP的增加。</li>
	</ol></li>
</ol>3. 胶质细胞瘤启动细胞的分离和培养
<ol><li>通过分割高等级人体 GBM 活检来分离 GIC。将肿瘤在含有0.25%胰蛋白酶的烧杯中转移到0.1 mM EDTA (4:1)中,并在37°C下缓慢搅拌30-60分钟(取决于肿瘤大小)。</li>
	<li>在GIC培养基中,在2cm 2cm2中,在75 cm2组织培养瓶中镀上分离的细胞:DMEM/F-12介质(1:1),含有1%的N2、1%B27和1%的G5补充剂(以利于GIC生存),辅以bFGF和EGF(均为10分)ng/Ml,促进茎)和1%的青霉素/链霉素。</li>

</ol>4. hESC衍生多巴胺类有机物,用于PD研究

</ol>5. TH和Nurr1基因表达的定量,用于验证多巴胺能分化

</ol>6. 高压液相色谱 (HPLC) 检测

human neural organoids for studying brain cancer and neurodegenerative diseases

在再生医学中，一个针对特定神经细胞类型的多能干细胞分化的稳健协议是一个有价值的工具。在我们的案例中，这个协议对于研究胶质母细胞瘤发育和帕金森病是非常有用的。因此，首先，三维器官的生成具有密切模仿生理发育的优点，而且多能干细胞产生大小校准神经球具有很高的可重复性。以下程序将由来自我们实验室的博士生马农·洛卡泰利演示。在开始三D培养前24小时，用无血清介质代替HESC培养基，辅以10微摩尔 ROCK抑制剂。细胞应为60%的汇合。第二天，通过去除介质和用不含钙和无镁的PBS冲洗，分离hESC菌落作为单细胞。然后，加入5毫升的酶溶解溶液，在37摄氏度下孵育一至两分钟。之后，用10微摩尔 ROCK抑制剂收集无血清介质中的细胞，并在300倍g下将细胞离心5分钟。离心后，检查细胞颗粒的大小。去除上清液，在10毫升无血清介质中计数细胞，并辅以10微摩尔 ROCK抑制剂。同时，每口井用两毫升无血清介质冲洗微井板，并在1，200次g下离心5分钟，以清除所有气泡，以防止神经球的形成。接下来，在12.5毫升无血清介质中准备2820万个细胞，并辅以10微摩尔 ROCK抑制剂。每微井分配1000个细胞。将300倍g的细胞离心5分钟，并在显微镜下检查微井板中细胞的均质重新分离。将板在37摄氏度的培养箱中过夜。第二天，检查微井板，检查每个微井中形成球体的大小。使用辅二SMAD抑制鸡尾酒的介质将球体转移到六井板，促进快速神经感应。从这一步前进，球体以60转/分的转速旋转，以防止它们粘在一起或粘在板上。第一天到第四天，在进行神经感应之前，每两到三天更换一次介质。从第四天到第11天，通过添加EGF和bFGF以每毫升10毫微克添加到B27介质辅以双SMAD鸡尾酒，促进HESC衍生神经玫瑰花环的增殖。从第11天到第13天，培养B27介质中的球体，辅以0.5微摩尔BMP抑制剂。从第13天到第21天，培养B27介质中的球体，以每毫升10毫微克和1微摩尔伽马-分泌酶抑制剂为补充GDNF和BDNF。在第21天，将一个培养板插入到新的六井板的一个井中。之后，在膜插入物下的每孔中加入一毫升B27介质，并辅以生长因子和抑制剂。随后，将球体板放在沉积在培养板上的亲水PTFE膜上，并在随后三周的分化中每两到三天更换一次介质。停止此步骤的旋转。玫瑰花环的存在表明神经分化的开始。从第21天到25日，在同一神经成熟媒介中培养人类神经器官。然后，从第25天到28日，只补充B27介质与一个微摩尔伽马-分泌酶抑制剂。从第28天到第39天，停止添加伽马-秘密酶抑制剂，继续在B27介质中继续人类神经器官培养。三周后，神经器官准备用于GIC植入。在第 0 天，在双 D 培养中放大 60% 的合合。然后，用含有双SMAD抑制鸡尾酒的无血清培养基取代用于维持HESCs多能特性的干细胞介质，开始神经诱导和10微摩尔 ROCK抑制剂，以提高第二天通过过程中细胞的存活率。第一天，准备微井板，每井含有2.5毫升的无血清介质，含有相同浓度的 ROCK 抑制剂和双SMAD抑制分子。为了区分细胞对神经管腹型，添加SHH和FGF8在100南克每毫升和两个微摩尔平滑激动。加入介质后，将板以 1，200 倍 g 离心 5 分钟，以清除微井中的气泡。一旦微井板准备好使用半分型介质，取出hESC的介质，并迅速用不含钙和氯化镁的PBS清洗。在T75烧瓶中加入7.5毫升重组酶溶液，将菌落分离成单细胞悬浮液。在37摄氏度下孵育细胞2分钟，然后加入7.5毫升DMEM F12。随后，收集电池悬浮液和离心机在300倍g5分钟。然后，取出上清液，并计算用于准备微井板的相同介质中的细胞。调整介质体积，通过制备2.5毫升介质中的470万个细胞，并添加到先前已经放入的2.5毫升的培养中，获得细胞悬浮液，从而形成每个微井包含1000个细胞的神经球。为了正确分配每个微井中的细胞，轻轻摇动板，以300倍g离心5分钟。接下来，在37摄氏度下孵育板24小时，生成球体。在第二天，用介质轻轻冲洗微井，并在组织处理的六井板中转移球体。将球体旋转至37摄氏度，每两到三天更换一半介质，使用新鲜介质。第三天至第13天，为了增强神经感应，用中脑识别物转化为神经祖细胞，用三微摩尔葛兰素史克-3-β抑制剂补充该介质。将样品分成两个新的组织处理六孔板，以减少球体密度，避免球体聚集。第8天，开始神经成熟，切换到新的媒介与生长因子，并改变介质每两到三天。在第21天，将一个培养板插入到新的六孔板的一个孔中，并在膜插入下加入1.2毫升用于神经球分化的神经成熟介质。然后，将 PTFE 膜放在培养板插入件上。最后，在PTFE膜的空液界面条件下，产生神经器官，在100个神经球中播种。停止此步骤的旋转，每两到三天更换一次介质，直到达到所需的区分时间点。这里是用于生成多巴胺神经器官的标准化协议的示意图。这些图像中显示的多巴胺神经器官的免疫荧光分析表明TH免疫反应细胞共同表达Nurr1，一种中脑特异性标记。这两个图形表示由定量RT-PCR评估的TH和Nur1基因表达的动力学。下面是使用 MEA 平台记录的原始数据的示例。每个尖峰都用垂直线显示，而其余轨迹是噪声。这张照片代表一个沉积在 MEA 上的神经球。这些图表显示了从原始数据检测到的典型峰值的叠加。黑色粗体曲线表示相应红色曲线的平均值。此栅格图显示与检测到的每个峰值关联的时间戳。不同的颜色突出显示不同的电极。该协议允许研究人员回答有关癌细胞与神经器官相互作用的问题，以及直接筛选与多巴胺机器官。
