マウスの裂肛は、目の大きさと形と、眼の部分の脆弱性のために、本当に繊細なプロセスです。効率的な方法とヒントを提供する詳細なプロトコルを持つことは、高品質のレティナル解剖とセクションの鍵です。研究者が共通の落とし穴を避け、免疫検査に適した高品質のレチナルセクションを得るのを助けるヒントとアドバイスを提供します。尾のコーテアライザーを使用して安楽死させたマウスの目の一時的な部分を非常に軽く触れ、角膜をわずかに燃やして穴を開けるのを避けるために1秒以下の部分をマークします。その直後に湾曲したデュモン545鉗子を使用してマウスの目を核とした。4%PFAとPBSの1ミリリットルの1.5ミリリットルのクライオチューブに目を入れ、氷の上で15分間インキュベートします。次に、曲面デュモン545鉗子を用いて固定眼を、PBSで満たされた改変された35ミリメートルの解剖皿に移し、解剖顕微鏡の下に置く。マイクロナイフを使用して、角膜の縁端のすぐ上と垂直なバーマークで小さな切開を行います。湾曲したはさみを切開に挿入し、手足に続く角膜の周回カットを行います。次に、薄いデュモン5鉗子を使用して、角膜を取り除き、レンズを抽出し、目の後部から繊細に離れて持ち上げます。ガラス体はレンズで出てきますし、後眼球の内側を覆う白い表面として、レチナが見えます。分離したアイカップをPBSの1ミリリットルで室温で10分間2回洗います。アイカップを凍結保護するには、15%のスクロースとPBSで一晩4度で平らにし、沈みます。その後、アイカップを30%スクロースに移し、2〜3時間PBSに移して沈みます。埋め込み用の10ミリメートルのクライオモールドで10月にアイカップを置きます。解剖顕微鏡の下で、火傷マークが上になるまで目を回転させることによって、その側側腹側軸に沿ってクライオモールドの眼を向けます。視神経は左の1つであり、カビの切り取られた部分は右に向いています。残存組織を凍結するには、イソペンタンを含む金属ビーカーに少なくとも5分間クライオモールドを浸し、ビーカーをビーカーの高さの3分の1まで液体窒素に入れます。冷凍ブロックを取り外し、アルミホイルで包み、マイナス80度で保管します。ペンまたはシャーピーを使用して、フリーズしたブロックに印を付け、その向きを記録します。クライオスタット温度をマイナス20度からマイナス25度の間で調整した後、埋め込まれたアイカップを含む金型を内部に入れ、クライオスタット温度に1時間平衡化できるようにします。側腹側の方向を持つクライオスタットにブロックを取り付け、10マイクロメートル厚いシリアルセクションの切断を開始します。注意深く、注意して、注意して、番号付きの顕微鏡スライドにレチンのセクションを置き、免疫染色の準備ができるまでマイナス20°Cまたはマイナス80°Cでスライドボックスに保存します。ロドプシンに対する抗体を用いた免疫組織化学、感光体マーカー、グルタミン酸デカルボキシラーゼ65、アマカリヌ細胞マーカー、グルタミン合成酵素およびミュラー細胞マーカー、およびカルビンビンジン、水平細胞マーカーを、残膜切片に対して行った。この結果は、このプロトコルが異なるプロトコルから得られた低品質のレティナルセクションとは異なり、高品質でよく保存されたレチンのセクションを生み出していることを示しています。ロドプシンの豊富な内側および外側のセグメントは、網膜色素上皮の上に置かからほとんどまたは全く分離されずに垂直および無傷のままである。Gad65陽性アマトリン細胞などのインターニューロンは、内部核層および内膜神経膜層内で適切に階層化されたままである。また、Muller細胞は、神経節細胞層から外核層に及ぶ細胞質プロセスにおいて適切に整列した相馬でよく保存されたままである。さらに、明確に定義された水平細胞は、内部核層および外型プレキシフォーム層境界で検出可能である。目のトッパー領域をマークし、埋め込み時に向きを維持することが重要です。常に適切な個人用保護具でフードの下にパラホルムアルデヒドを準備します。
