De ziekte van Parkinson (PD) wordt veroorzaakt door degeneratie van Dopaminerge (DA) neuronen bij de Substantia nigra pars Compacta (SNpc) in de ventrale Mesencephalon (VM). Cel vervangende therapie heeft een grote belofte voor de behandeling van PD. onlangs, geïnduceerde neurale stamcellen (iNSCs) zijn ontstaan als een potentiële kandidaat voor cel vervangende therapie als gevolg van het verminderde risico van tumorvorming en de plasticiteit te geven aanleiding tot regiospecifieke neuronen en glia. iNSCs kunnen worden geherprogrammeerd vanuit autologe somatische cellulaire bronnen, zoals fibroblasten, perifere bloed mononucleaire cellen (Pbmnc's) en verschillende andere soorten cellen. In deze studie beschrijven we een protocol waarin iNSCs worden verkregen door het herprogrammeren van Pbmnc's, en onderscheiden in gespecialiseerde VM DA neuronen door middel van een tweetraps methode. Dan worden DA-precursoren getransplanteerd in eenzijdig 6-hyroxydopamine (6-OHDA)-lesioned PD-Muismodellen om de veiligheid en werkzaamheid voor de behandeling van PD te evalueren. INSC kolonies ontstond op dag 12.Na het plukken en overbrengen van iNSC klonen voor uitbreiding voor een aantal passages, toonden ze een goede morfologie en konden ze zichzelf stabiel vernieuwen in iNSC-medium, hetzij in een monolaagvorm of als sferen. Na 24 dagen van differentiatie, iNSCs werden geïdentificeerd als dopaminerge neuronen als een meerderheid van hen uitgedrukt tyrosine hydroxylase, vorkhoofd doos A2, en neuron-specifieke klasse III beta-tubuline markers. Drie maanden na transplantatie, onder overlevende cellen, ongeveer 13.84%waren TH-positieve dopaminerge neuronen.Ongeveer 91,72% van de TH-positieve cellen uitgedrukt weeskern receptor, ongeveer 86,76%uitgedrukt FOXA2, en ongeveer 98,77%werden mede-gelabeld met G-eiwit gekoppelde inkomende rectifier kalium. Bij het leggen van 30 mils verdund perifeer bloed op het dichtheidsgradiëntmedium, doe het dan langzaam en voorzichtig, niet om de gradiëntinterface te verstoren. Zorg moet worden genomen om te voorkomen dat herhaalde ontdooien en bevriezing van Sendai virus om een hoge virusactiviteit te garanderen.Bij het herprogrammeren van perifere bloed mononucleaire cellen aan geïnduceerde neurale stamcellen, gelieve nauwlettend observeren de morfologie van cellen elke dag. Ook is 6-hydroxydopamine licht- en temperatuurgevoelig. Wees voorzichtig om de oplossing te beschermen tegen licht, en houd het op ijs voor gebruik.Na deze procedure kan men doorgaan met het bestuderen van de ziektemechanismen met behulp van familiale PD patiënt-afgeleide dopaminerge neuronen in de cultuur. Men kan ook blijven onderzoeken hoe de getransplanteerde cellen de beschadigde neuronale circuits beïnvloeden.
