1. 制备MCP-1负载生长因子-减少基底膜衍生解决方案

</ol>2. 注射基底膜衍生溶液

</ol>3. 收获基底膜衍生凝胶塞
<ol><li>为每只小鼠称重两个单独的 1.5 mL 微离心管,并标记它们。</li>
	<li>注射基底膜衍生溶液三天后,在麻醉室用3%异二苯基使小鼠安乐死,然后进行宫颈脱位。</li>

</ol>5. 使用单细胞西胸(scWB)分析确定细胞悬浮液中的细胞组成
<ol><li>在 Rt 在培养皿中以 15 mL 的 1x 悬浮缓冲液重新冻结 scWB 芯片至少 10 分钟。</li>
	<li>将稀释后的单个细胞悬浮液(10,000-100,000个电池/mL)加入1 mL的再水化芯片。将表面放在 10 厘米培养皿的底部。确保曲面设置平整。</li>

	<li>准备单细胞西式仪器。</li>

	<li>小心地将芯片加载到单细胞西式仪器的电泳单元中,凝胶面朝。小心不要损坏芯片的凝胶侧。</li>
	<li>将裂液/运行缓冲液倒入腔室,并完全覆盖整个单细胞西芯片。启动细胞莱沙。</li>

</ol>6. 剥离 scWB 芯片
<ol><li>将扫描的芯片存放在洗涤缓冲液中,直到准备剥离。</li>
	<li>将水浴放在烟罩中。</li>
	<li>将 50 mL 管架放在水浴中,水距机架仅 1 厘米,并将水温设置为 60°C。</li>
	<li>准备剥离缓冲区。对于1 L剥离缓冲液,在900 mL蒸馏水中溶解9.85克Tris-HCl(pH 6.8)和20克SDS,并调整pH。然后加入蒸馏水至1L。每次使用前加入0.8%(v/v)β-甲苯醇(β-ME;14.3 M)。</li>
	<li>初次扫描后,将芯片放入10厘米培养皿中,然后再剥离。</li>
	<li>对于每个芯片,取取40 mL的剥离缓冲液,并加入320μl的β-ME。 注意:β-ME对人类和环境有毒和有害。应在烟罩中执行以下步骤。</li>

为了获得血液单核细胞对化学吸引剂的响应能力,我们向每只小鼠的左右侧翼注射车载和MCP-1加载的基底膜衍生溶液。注射后1天、3天和5天,将基底膜衍生的凝胶塞取出,溶解并加入每个塞内细胞(图1A)。通过从 MCP-1 加载插头(闭合柱)中的单元计数中减去车载插头(开杆)中的单元计数,我们获得了响应 MCP-1(+ 细胞编号,图 1B)而专门招募的细胞数。我们观察到在5天期间加速MCP-1特定细胞的招募和积累,其比率从每天31,000个细胞(第1天)增加到78,000个细胞/天(第3天),第5天每天136,000个细胞(图1B)。
为了识别进入基底膜衍生凝胶塞的细胞类型,我们根据从每个插头分离的细胞进行单细胞西斑分析(scWB),探测CD45、CD11b和F4/80表达,以识别单核细胞(CD11b=F4/80)-), 巨噬细胞 (CD11b+F4/80+加上 CD11b-F4/80+) 和剩余的非单细胞白细胞 (CD45+CD11b-F4/80-) (图2)。然后,对scWB芯片进行探查,以测定每个凝胶上的总细胞数,并确认scWB芯片上微孔的单细胞占用率。MCP-1加载的基底膜衍生凝胶塞中单核细胞的百分比在第1天、20%时处于低位,在第3天达到47%的峰值,第5天降至14%(图2D)。MCP-1 加载的基底膜衍生凝胶塞内的巨噬细胞数量从第 1 天的 13% 稳步增加至第 3 天的 22%和第 5 天的 23%。对于 MCP-1 加载插头,隔离细胞群中单核细胞加巨噬细胞的百分比在第 3 天最高,车辆加载插头(图 3A)为 31%,MCP-1 加载插头为 58%(图 3B),表明 3 天后MCP-1依赖性化学细胞招募的绝大多数细胞是单核细胞和巨噬细胞。尽管单核细胞加巨噬细胞的总数在第5天比第3天(图3C和D)高,但由于单核细胞和巨噬细胞在细胞总数中所占的比例明显较高(图3A)和B),第3天的细胞计数更准确地反映了血液单细胞化学和招募。因此,我们通常在牺牲动物前三天注射基底膜衍生溶液。MCP-1招募的所有巨噬细胞是否都是单核衍生物还是从邻近组织招募的常驻巨噬细胞,尚未确定。

这种方法使我们能够量化活体动物中血液单细胞的化疗活性，并研究不良饮食对单细胞功能及其基本机制的影响。单细胞衍生巨噬细胞可以使用各种方法进行分离和分析，包括单细胞和在小鼠模型中提供血液单细胞位点健康状况的奥米学技术。今天的手术将由我实验室的助理教授杨俊博士和实验室的博士后王露茜博士进行。在开始手术之前，用消毒剂清洁细胞培养罩，并完全解冻冰上生长因子减少的地下室膜衍生溶液。用 26 个测针为单个无菌单毫升注射器配备，并将针头放在冰上。溶液解冻后，用酒精拭子擦拭地下室膜衍生溶液小瓶的顶部，然后将 500 微升溶液装入每毫升注射器中，并辅以或不带化疗吸引剂。然后清除地下室膜衍生溶液中加载的注射器的任何气泡，以确保插头形成均匀。在确认对头趾捏缺乏反应后，缓慢地将整个体积的地下室膜衍生溶液注入麻醉小鼠的右侧，而不添加 MCP-1，以约 100 微升/秒的皮下作用。整个500微升的地下室膜衍生溶液辅以MCP-1到动物的左侧。注射后 20 至 30 秒将注射器固定在就位，以防止溶液泄漏，并允许形成单个光滑的凝胶塞，然后将鼠标放在加热垫上，并进行监控，直到完全恢复。注射三天后，从插入注射的动物中去除后侧毛，并用钳子抓住下胸椎和上腰椎周围的皮肤。将两毫米长的切口切入皮肤，沿着小鼠背部的中线从颈椎向下切到椎骨结上方一厘米。将皮肤与肌肉层分离，将皮肤固定在聚苯乙烯泡沫平台上。接下来，在解剖显微镜下，仔细抓住含有基膜衍生凝胶塞的纤维胶囊，并使用细钳去除纤维胶囊。然后用细剪刀清洁插头。然后将清洁的地下室膜衍生凝胶插头转移到贴有适当标签且重1.5毫升的微型离心管中。重新加压管子以计算地下室膜衍生凝胶塞的重量。对于地下室膜衍生凝胶塞的消化，使用干净的细剪刀将地下室膜衍生凝胶塞切碎，并加入800微升置换插头。以最高速度旋转 10 秒，以中断插头，将微型离心管在 37 摄氏度下以每分钟 1400 次旋转的速度在热混合器中放置两个小时，从而完全溶解地下室膜衍生凝胶塞。在孵化结束时，通过离心两次沉淀塞片，并在300微升PBS中重新悬浮颗粒。将每个细胞悬浮液的50微升转移到新的微离心管中，用0.5微升钙化物标记新管中的细胞。在二氧化碳培养箱中，在37摄氏度下孵育10分钟后，在自动细胞计数器上计算活的绿色荧光阳性和死非荧光细胞的数量。对于单细胞西印迹分析，在10厘米培养皿底部的一块重新加水化的单细胞西印片中加入一毫升稀释的单细胞悬浮液。5 到 15 分钟后，检查亮场显微镜下的芯片。大约 15 到 20% 的微孔应由单个细胞占据，少于 2% 的孔应包含两个或更多细胞。如果芯片已正确加载，将盘式倾斜 45 度，用悬架缓冲液清洗芯片三次，以清除任何未捕获的电池。最后一次洗涤后，小心地将芯片加载到单细胞西式仪器的电泳细胞中，凝胶侧向上，用解合运行缓冲液覆盖整个单细胞西印片。启动细胞解解，并根据适当的实验参数运行仪器。在运行结束时，在室温下用每次洗涤的新鲜洗涤液用两个 10 分钟的洗涤器冲洗芯片。第二次洗涤后，将80微升的原抗体溶液加入抗体检测室，并将芯片凝胶侧向下降低，使抗体溶液在芯片上芯。在室温下两小时后，在洗涤缓冲液中清洗芯片三次，在水中清洗一次。在室温下用适当的二次抗体孵育芯片一小时，防止光线。在孵育结束时，用洗涤缓冲器清洗芯片三次，并在滑动微调器上旋转芯片以去除剩余的洗涤缓冲液。要剥离单细胞西印片，请将一个 15 毫升的管架放在 60 摄氏度的水浴中，水在机架上方一厘米处。在烟罩中，在罐中加入40毫升脱衣缓冲液和320微升β莫塞塔乙醇。将芯片放在罐内 10 厘米的培养皿中，然后用 Parafilm 密封罐。然后将罐放在水浴的管架内。90分钟后，小心地将芯片转移到新的培养皿中，用洗涤缓冲液短暂清洗一次，然后将15毫升的新鲜洗涤缓冲液加入培养皿，在摇床上清洗15分钟。在这个具有代表性的实验中，每个插头中招募的细胞在注射后一天、三天和五天被计算在内。通过从 MCP-1 加载插头中的单元计数中减去车辆加载插头中的细胞计数，然后计算为响应化疗吸引剂而专门招募的细胞数。在五天期间观察到MCP-1特异性细胞的加速招募和积累，注射一天后，细胞的发病率从每天3.1万个细胞增加到注射后五天的13.6万个细胞。然后，根据单个细胞对感兴趣的免疫细胞标记的表达，使用单细胞西部印迹分析来识别地下室膜衍生塞中招募的不同细胞类型。例如，MCP-1 中加载的基底膜衍生凝胶塞中的单核细胞百分比在第一天较低，在第三天达到峰值，然后在第五天再次下降，而巨噬细胞数量在整个研究期间稳步上升。对于 MCP-1 加载的插头，隔离细胞群体中单细胞加上巨噬细胞的百分比在第三天最高，表明三天后，MCP-1 依赖性化疗所招募的绝大多数细胞是单核细胞和巨噬细胞。尽管单核细胞和巨噬细胞的总数通常高于第三天，但第三天的细胞计数更准确地反映了单核细胞化疗的外周血和招募，因此建议第三天进行地下室膜直接插头分析。成功采集用于下游分析的单细胞取决于注射的准确性和插头的完全去除。流细胞学、单细胞西印迹、体积或单细胞RNA测序等奥米学程序可用于评估所招募的单细胞和单细胞衍生巨噬细胞的特征和表型。
