1. 第0天: 鸡蛋生产和收集

4. 第3天:72 hpf 的细胞死亡和白细胞分析

using zebrafish larvae to study the pathological consequences of hemorrhagic stroke

使用双源泛素评估脑细胞死亡: Secanexinv-mvenus 在 atv 和 bbh 模型中产生明显的结论性死亡细胞簇, 而这些模型在所有 ich-vae 中都不存在 (图 3)。集群在 96 hpf 之前退去。通过图像分析, 出血与大脑中荧光信号的总强度显著增加两倍有关, 这表明细胞死亡明显。
通过大脑中mpeg1阳性巨噬细胞的显著增加, 在 ich + 幼虫中发现了神经炎症反应。总的 mpo阳性中性粒细胞的数量也增加了, 但这没有达到统计意义 (图 4)。Mpeg1阳性巨噬细胞的形态也可以在 ich + 幼虫中发生变化, 因为细胞采用活性的、圆形的、阿米体状。这些激活的圆形细胞也可以随着时间的推移进行监测, 以显示泛素的吞噬反应增加: SECANNEXINV-MVENUS 在 ich + 幼虫中表达死亡细胞 (图 5)。mpeg1显示成熟过程的阳性巨噬细胞被归类为非活性巨噬细胞。
与 bbh 和 ATV 模型中的 ichb 兄弟姐妹对照相比, 脑出血与72和 96 hpf 的运动力显著下降有关 (图 6)。120 hpf 的动力恢复到接近基线水平。鸡蛋离合器和菌株之间的基线运动往往存在差异, 因此每次都应与 ichh 控制进行比较。
