Desuden, Gut bakterier interagere med vært tarmceller og regulere grundlæggende vært processer, herunder metaboliske aktiviteter, næringsstof tilgængeliggørelse, immunsystem modulation, og endda hjernefunktion og beslutningstagning2,3 . Endobiotika har et betydeligt potentiale til at påvirke den bakterielle sammensætning og mangfoldighed af Gut microbiota. Således, interaktioner mellem endobiotika og human Gut mikrobiota har tiltrukket stigende forskning opmærksomhed4,5,6,7,8,9.
Det er vanskeligt at evaluere samspillet mellem endobiotika og humant tarm mikrobiota in vivo på grund af bioetik og økonomiske begrænsninger. For eksempel kan eksperimenter, der undersøger samspillet mellem endobiotika og humant tarm mikrobiota, ikke udføres uden tilladelse fra Food and Drug Administration, og rekruttering af frivillige er dyrt. Derfor anvendes dyremodeller ofte til sådanne undersøgelser. Brugen af dyremodeller er dog begrænset på grund af forskellige mikrobiota kompositioner og diversitet i dyr-vs. menneske-associerede samfund. En alternativ in vitro-metode til at udforske samspillet mellem endobiotika og human Gut mikrobiota er gennem brug af batch kultur eksperimenter.
Exopolysaccharider (Eps'er) er prebiotics, der bidrager væsentligt til opretholdelsen af menneskers sundhed10. Særskilte Eps'er, der består af forskellige monosaccharid kompositioner og strukturer kan udvise forskellige funktioner. Tidligere analyser har fastlagt sammensætningen af BIF Eps'er, som er det repræsentative xenobiotiske mål i den nuværende undersøgelse11. Men, Host-associerede metaboliske effekter er ikke blevet overvejet med hensyn til EPS sammensætning og mangfoldighed.
Den protokol, der er beskrevet her, bruger fækal mikrobiota fra 12 frivillige til gære BIF EPSS. Tyndtlags kromatografi (TLC), 16S rRNA-gensekvensering med høj gennemløb og gaskromatografi (GC) anvendes derefter i kombination til at undersøge samspillet mellem EPSs og humant tarm mikrobiota. analysis of interactions between endobiotics and human gut microbiota using in vitro bath fermentation systems

Humane tarm mikroorganismer er for nylig blevet et vigtigt mål for forskning i at fremme menneskers sundhed og forebygge sygdomme. Derfor er undersøgelser af interaktioner mellem endobiotika (f. eks. lægemidler og Prebiotika) og Gut mikrobiota blevet et vigtigt forskningsemne. Men in vivo eksperimenter med humane frivillige er ikke ideelle til sådanne undersøgelser på grund af bioetik og økonomiske begrænsninger. Som følge heraf er der blevet anvendt dyremodeller til at evaluere disse interaktioner in vivo. Ikke desto mindre er dyremodel undersøgelser stadig begrænset af bioetiske overvejelser, foruden forskellige sammensætninger og forskelle i mikrobiota hos dyr vs. mennesker. En alternativ forskningsstrategi er brugen af batch fermenterings eksperimenter, der muliggør evaluering af samspillet mellem endobiotika og Gut mikrobiota in vitro. For at evaluere denne strategi, bifidobacterial (BIF) Exopolysaccharider (EPS) blev brugt som en repræsentativ xenobiotic. Derefter, samspillet mellem BIF EPS og human Gut mikrobiota blev undersøgt ved hjælp af flere metoder såsom tyndtlagkromatografi (TLC), bakteriel samfund kompositoriske analyse med 16S rRNA gen High-gennemløb Sequencing, og gaskromatografi af kortkædede fedtsyrer (SCFAs). Produktionen af mucoid EPS kunne observeres i B. longum kulturer på PYG plader efter anaerob inkubation for 72 h (figur 1a). Vores protokol kan i vid udstrækning anvendes til høj-gennemløb og hurtig screening af samspillet mellem endobiotika og menneskelige tarm mikrobiota. Lave omkostninger og undgåelse af interferens effekter fra værtens stofskifte. Denne procedure har potentiale til at blive brugt i diagnosen.Først opløses BIF EPS og stivelse individuelt i varmt vand på en magnetisk omrører for at gøre både koncentrationen af otte gram pr. liter. I det forberedte basiskulturmedium VI skal du tilføje 100 milliliter BIF EPS-løsningen for at lave kultur VI BIF-løsning. Tilsæt 100 milliliter stivelsesopløsningen for at gøre kultur VI-stivelsesopløsningen.Autoklave de to medier med kulstofkilde, og mediet uden kulstofkilde som en kontrol, ved 121 grader Celsius i 15 minutter. Principal Coordinate Analysis viser kultur VI BIF, og kultur VI stivelse grupper, grupperet adskilt fra hinanden, hvilket indikerer, at EPS og stivelse tilgængelighed differentieret forme menneskelige fækale bakteriesamfund.Lineær Discriminant Analyse på effekt størrelse viser slægter collinsella, coprococcus, parabacteroides, og rhodopseudomonas var betydeligt mere rigelige i kulturen VI BIF prøver end i kultur VI stivelse prøver.
