Se han realizado progresos significativos hacia la comprensión de la composición y las actividades de la microbiota intestinal humana en la última década. La microbiota intestinal desempeña un papel importante en el funcionamiento de los intestinos humanos y en la salud del huésped. En consecuencia, la microbiota intestinal se ha convertido recientemente en un objetivo importante para la prevención y la terapia de enfermedades1. Además, las bacterias intestinales interactúan con las células intestinales anfitrionas y regulan los procesos fundamentales del huésped, incluidas las actividades metabólicas, la disponibilidad de nutrientes, la modulación del sistema inmunitario e incluso la función cerebral y la toma de decisiones2,3 . Los endobióticos tienen un potencial considerable para influir en la composición bacteriana y la diversidad de la microbiota intestinal. Así, las interacciones entre endobióticos y microbiota intestinal humana han atraído una creciente atención de la investigación4,5,6,7,8,9.
Es difícil evaluar las interacciones entre los endobióticos y la microbiota intestinal humana in vivo debido a la bioética y las limitaciones económicas. Por ejemplo, los experimentos que investigan las interacciones entre los endobióticos y la microbiota intestinal humana no se pueden realizar sin el permiso de la Administración de Alimentos y Medicamentos, y el reclutamiento de voluntarios es costoso. En consecuencia, los modelos animales se utilizan a menudo para tales investigaciones. Sin embargo, el uso de modelos animales es limitado debido a las diferentes composiciones de microbiota y diversidad en las comunidades asociadas a los animales frente al ser humano. Un método in vitro alternativo para explorar las interacciones entre los endobióticos y la microbiota intestinal humana es a través del uso de experimentos de cultivo por lotes.
Los exopolisacáridos (SEP) son prebióticos que contribuyen significativamente al mantenimiento de la salud humana10. Los SAE distintos que consisten en diferentes composiciones y estructuras monosacáridos pueden exhibir funciones distintas. Los análisis anteriores han determinado la composición de los SAE Bif, que son el xenobiótico representativo al que se dirige en el actual estudio11. Sin embargo, efectos metabólicos asociados al huésped no se han considerado con respecto a la composición y diversidad EPS.
El protocolo descrito aquí utiliza la microbiota fecal de 12 voluntarios para fermentar Bif EPS. La cromatografía de capa delgada (TLC), la secuenciación de alto rendimiento del gen rRNA 16S y la cromatografía de gases (GC) se utilizan en combinación para investigar las interacciones entre los SSA y la microbiota intestinal humana. Las ventajas distintivas de este protocolo en comparación con los experimentos in vivo son su bajo costo y evitar los efectos de interferencia del metabolismo del huésped. Además, el protocolo descrito se puede utilizar en otros estudios que investigan interacciones entre endobióticos y microbiota intestinal humana.

Los microorganismos intestinales humanos se han convertido recientemente en un objetivo importante de la investigación en la promoción de la salud humana y la prevención de enfermedades. En consecuencia, las investigaciones de interacciones entre endobióticos (por ejemplo, fármacos y prebióticos) y la microbiota intestinal se han convertido en un importante tema de investigación. Luego, las interacciones entre Bif EPS y la microbiota intestinal humana se investigaron utilizando varios métodos como la cromatografía de capa delgada (TLC), el análisis de composición de la comunidad bacteriana con secuenciación de alto rendimiento del gen rRNA 16S y la cromatografía de gases de ácidos grasos de cadena corta (SCFA).
