この技術は、クリケットの胚または幼虫の発達をアッセイするように設計された実験の基礎的な方法として機能する。この技術の主な利点は、柔軟性があり、RNA干渉やゲノム操作を利用するものなど、いくつかのタイプの実験的アプローチをサポートすることです。手順のデモンストレーションは、研究室の主任研究者であるハドリー・ウィルソン・ホーチです。この手順を開始するには、滴水でベベラーの回転グラインダー面を濡らします。引っ張られた針をベベラーに入れ、20度の角度に変えます。砥石に触れるまで針を下げ、2~3分ベベルします。ガラス顕微鏡スライドに付着した二重スティックテープにベベル針を置くことによってベベルを評価します。カメラと画像取得ソフトウェアを搭載した複合顕微鏡のステージ上にスライドを置きます。20倍の目的を使用して針の画像を取得します。イメージ投射ソフトウェアを使用して、ベベリング開口部に近い針の内腔径を測定します。8マイクロメートル以下または12マイクロメートルより大きい開口部を持つ針を捨てます。まず、水に1%アガロースの40ミリリットルを作り、10センチメートルのペトリ皿に注ぎます。アガロースの表面に卵のスタンプを置き、固めます。アガロースが固まったら、切手を取り除き、井戸を明らかにします。次に、アガロースウェルプレートに1%ペニシリンストレプトマイシンを含むHBSを充填する。皿の上に蓋を置き、パラフィルムで包み、摂氏4度で保管します。この後、35ミリメートルのペトリ皿に白い遊び場の砂を入れて、コオロギが卵を産むために卵を集めるコレクション皿を作ります。約18×18センチメートルにカットペーパータオルスクエアで卵皿をカバー。ペーパータオルを通して水道水で満たします。その後、ペトリ皿を傾け、余分な水を取り除くために上を軽く絞ります。皿の下にペーパータオルの正方形の角をタックし、ペーパータオルを所定の位置に保つのに役立つ逆蓋に卵皿を置きます。卵皿をクリケットビンに入れ、大人のメスに卵を1~2時間オビポジットさせます。一方、アガロース皿は、注射のために卵を保持する準備として室温まで暖かくすることができます。準備ができたら、産きたての卵を含む卵の回収皿をクリケットビンから取り出します。ペーパータオルを取り出し、毛穴の大きさのストレーナーをビーカーの上に0.5〜1ミリメートル置きます。やさしく水道水を流す下で、産卵皿の内容物をストレーナーに洗い流します。クリケットの卵がストレーナーバスケットに残っている間、砂粒はストレーナーメッシュを通って下のビーカーに落ちるでしょう。逆浸透水で容器を充填し、トレイに入れます。容器の上にストレーナーを反転し、水に卵を取り除くために皿に対してそれをタップします。卵は容器の底に沈みます。次に、はさみを使用してP1000ピペットチップの先端を切り落とし、直径約3ミリメートルの開口部を作ります。このチップをP1000ピペッタに置き、容器からアガロースエッグモールドディッシュに卵を移します。プラスチック製のピンセットを使用して、アガロースの井戸に卵を並べます。各卵は、個々の井戸の底に沈みます。ペトリ皿を注入する準備ができるまで蓋をして覆います。まず、卵の皿を解剖顕微鏡の下に置き、約10倍の低倍率を選択します。20マイクロリットルの負荷チップとP10ピペットを使用して1.5マイクロリットルの注入溶液を作成し、注入針の広い端にローディングチップを挿入します。注射液を注射針に出します。注射ハウジングに針を挿入し、締め付け、針がハウジングに適切かつしっかりと挿入されていることを確認します。その後、慎重にマイクロマニピュレーターに注入ハウジングを挿入し、針で壊れたり怪我をしないように注意してください。顕微鏡を通して卵と針の両方を見ながら、皿グリッドの左上隅にあるXの近くに針を動かします。先端がHBSとペニシリンストレプトマイシン緩衝液に入るまで針を下げます。針を視野に入れ、卵皿を動かし、針が卵よりも皿の端に数ミリメートル近づくようにします。この後、ローダミンに適したフィルターに顕微鏡をセットし、針の蛍光を観察し、その先端に焦点を当てます。マイクロインジェクタでは、注入液が針から懸濁液に漏れ始めるまで、バランスノブをゆっくりと時計回りに回します。その後、染料が針から漏れるのを止めるまで、ノブを反時計回りに少し回します。針がまだ視野の中央に入ったまま、最初に注入する卵を針が狙われるように卵皿を動かします。1つの卵が視野の大部分を埋めるように、倍率を約50倍に調整します。マイクロマニピュレーターと手の両方を卵皿に使用して、注射のために針を動かします。マイクロマニピュレーターを使用して針を進め、注入する最初の卵にチップを挿入し、卵の長い軸に垂直な卵の後端から卵の長さの20〜30%の針を挿入することを確認します。マイクロインジェクターに注入フットペダルか注入ボタンで溶液を注入する。卵の中の蛍光材料の小さなボーラスは、正常な注入を示します。この後、卵から針を引っ込みます。卵が針の引き込み時に意図せずによく持ち上げられた場合は、小さな鉗子を使って卵を井戸に押し戻し、針を引き込みます。本研究では、クリケット卵は、RNA干渉およびゲノム操作を含むがこれらに限定されない多くの実験において基礎的方法として機能する技術を用いて注入される。生存率は、この実験の成功を評価するための重要なパラメータの1つです。ほとんどの死んだ卵は、卵の中の黄身と胚組織が不均一に凝集し始め、最終的には黄身のほとんどが卵の片側に移行するので、目で見やすい。4~6日後に評価すると、未注入卵の85%を超えて生存し、実験試薬を注射した卵は一般的に生存率が低かった。注射自体のすべての側面を制御する,針穿刺,色素と緩衝液の導入,または車両または二本鎖RNAの存在を含む,実験操作の真の効果を理解するために必要である。注射のフェノールツの結果を評価する方法は、注入されたものに依存します。例えば、特定のmRNAノックダウンの結果としての表現性変化は、総解剖学レベルで明白であり得る。一方、G.ビマクラタスアクチンプロモーターの制御下にあるヒストン2B遺伝子をタグ付けしたEGFPをコードするcDNAがピギーバックトランスポザーゼを用いてクリケットゲノムに挿入された場合、蛍光を用いて卵の各核を可視化し、EGFPタグ付きヒスト2Bタンパク質を興奮させることが可能になる。針が詰まらないようにバランスを調整することは、この手順の重要なステップです。黄身が針先を詰まらせることもあるので、その後の注射後に、いくつかの調整が必要な場合があります。クリケットはショウジョウバエのようなホロメタボリック昆虫をよく研究するために基礎的に分岐するヘミメバボロス昆虫です。クリケットの開発を研究することは、動物界全体の進化と発展についてもっと理解するのに役立ちます。このテクニックには、いくつかの練習が必要です。注射後4、5日の生存率が80%以上になるまで、コントロール注射を練習することをお勧めします
