手動解剖および組織学的観察は、通常、筋肉や結合組織などのヒト組織を調べるために使用される。しかし、手動解剖は繊細な構造を容易に損傷する可能性があり、組織学的観察は平らな断面面1、2に関する限られた情報を提供する。したがって、組織をより正確かつ効率的に検査するために改善された方法が必要である。
従来のコンピュータ断層撮影(CT)は、一般的に臨床現場で使用されるが、それは小さな構造2、3を区別する能力を欠いている。マイクロX線CT(マイクロCT)は、小さな構造物の3次元(3D)情報を破壊することなく、標本から得るための効果的なツールです。しかし、microCTは、高密度の組織だけが明確に視覚化することができるので、限られたアプリケーションを持っています。軟部組織の区別には使用できません。この制限を克服するために、染色剤を使用することができる。コントラスト増強剤は、ホスホトンスチン酸(PTA)、ホスホモリブジン酸、およびルゴルのヨウ素のような、走査時の軟部組織コントラスト率を向上させる4,5である。これらのエージェントを比較するいくつかの研究は、PTAが良好なパフォーマンスを示し、6、7、8を扱いやすいことを示唆しています。

我々は、眼窩領域内の繊細な構造であるヒトオービキュラリス保持靭帯の非侵襲的な調査において軟部組織のコントラストを高めるために、ホスホトゥンジン酸を用いたmicroCTを実施する。このプロトコルは、人間の軟部組織の非侵襲的な調査の研究者を支援するために最適化されており、合理的な費用で実行するのは簡単です。サンプルを入手するには、まず、色鉛筆を使用して、サンプル採取用の皮膚の切断領域を示します。マークされた線が内側のキャンパスに内側に延び、横方向のキャンパスに延び、下まぶたの上側の境界に優れ、軌道縁から線の下1センチメートル下に劣っていることを確認します。次に、ブレードを使用してマークされたラインに沿って顔のティッシュを切断し、ブレード先端が骨に触れるのに十分な深さがあることを確認します。採取されたサンプルには、皮膚、皮下組織、筋肉、脂肪、骨膜が含まれなければならない。収集時に、室温で5〜7日間、すぐに10%ホルマリンでサンプルを固定します。固定期間の終わりに、サンプルを5~7ミリメートルの厚さ3個でスライスします。針と黒糸を使用して、サンプルの方向を後の時点で決定できるように、各サンプルの超側側を縫います。1濃度につき1日、上昇エタノール濃度系列でサンプルを脱水し、染色されるまで70%エタノール溶液中にサンプルを残します。マイコン断層撮影スキャンが予定される1週間前に、2.1グラムのPTAを210ミリリットルの70%エタノールに加え、1分間に55〜60回転でシェーカーに溶液を置きます。スライスした各部分に70ミリリットルのプラスチック容器を3個用意します。染料がアルコール溶液に溶解したら、容器に溶液を充填する。1%PTAの各容器に1つの標本を入れ、5〜7日間毎分55〜60回転でサンプルを振ります。染色が完了したら、サンプルを新鮮な70%エタノールで1週間保管し、スキャンの準備をします。マイクロCTスキャンの場合は、パラフィルムでサンプルをラップして乾燥を防ぎ、サンプルをスキャナートレイに置きます。スキャナのソース電圧を70キロボルト、ソース電流を114マイクロアンペア、アルミニウムフィルタを0.5ミリメートル、画像ピクセルサイズを20平方マイクロメートル、ピクセルを2,240 x 2,240、エクスポージャーを500ミリ秒、回転ステップを0.3度に設定します。次に、スキャンを開始します。スキャンが完了したら、再構築ソフトウェアを開き、[アクション] と [データセットを開く] を選択して、スキャンしたファイルを起動します。[設定]タブを選択し、リングアーティファクトの削減を7に設定し、ビーム硬化補正を行います。再構築を開始するには、[開始] を選択します。最終的なデータは、指定されたフォルダに格納されます。再構築の最後に、ファイルサイズ変更ソフトウェアを開き、[ソース データ セット] を選択して再構築されたファイルを起動します。[宛先データ セット] タブで jpg を選択し、[画質] オプションの [補間なし] の [1/2 サイズ変更] オプションを選択し、[イメージ圧縮] タブでスライド バーを 100 に調整して変換を開始します。3D 再構築の場合は、3D ボリューム レンダリング ソフトウェアを開き、[アクション] と [ボリューム データのロード] を選択します。明るさとコントラストレベルを調整するには、[関数の転送エディタ]タブのヒストグラムで形状転送機能を変更します。次に、[オプションと照明]を選択し、[影]アイコンと[サーフェス照明]アイコンを選択して、リアルなモデリングトーンを実現します。クリックしてドラッグし、右クリックしてドラッグして3D画像を移動および回転させて最適なビューを見つけ、必要に応じてズームインまたはズームアウトします。Shift キーを押しながらドラッグして、断面画像を表示する場所をスライドさせます。[ライト]アイコンをオンにして、表示に最適な照明を見つけるために照明表示バーを調整し、[ライト]アイコンをオフにして[照明]タブを閉じます。[オプション] タブで、[表示とクリッピング ボックス] を選択して、最終イメージのボックスを非表示にします。次に、画像を保存するアクションと画像の保存を選択します。このORLの詳細な再構築は、PTA調製物を用いたmicroCTによって実証され、真皮と骨膜の間に斜めに伸びる靭帯性線維筋構造の視覚化を可能にした。コロナの観点では、繊維の量と複雑さは横に増加します。水平ビューでは、樹状の形成を伴う精巧なメッシュワークが観察される。矢状の観点では、ORL繊維の厚みは劣って減少する。全体として、この多方向観測は、ORLが、その位置に応じて繊維の数と厚さのばらつきを持つ連続プレートの多層メッシュワークで構成されていることを証明しています。染色の持続時間が検体の体積に比べて不十分な場合、最終像は、試料の中央領域に空の穴を含み得る。PTA以外の他のエージェントを使用することができ、異なるエージェントは異なるメリットまたは染色機能の興味を持っています。したがって、実験に従って染色領域を最適化すると便利です。
