筋線維壊死は、複数の神経筋疾患の特徴であり、病因の中心的な役割を果たしています。筋肉変性を評価するための特定の信頼性の高い方法は、診断および翻訳研究にとって重要です。この非常に適応可能な技術は、そのミオファイバー内の同じセクション内で同時に複数の抗原の標識を可能にする。それは重要な染料で可能である。H&E染色は、信頼性が高く再現可能な活用には適応できない経験的手法です。IgGの取り込み染色は、しかし、壊死性筋細胞に特異的であり、ヒトの生検に適用可能である。この技術は、筋性脳神経症を特徴とする全ての神経筋疾患に関する洞察を提供することができる。興味のある脚から髪を取り除いた後、4週齢のオスのmdx動物をsupineの位置に置き、足をコルクボードに固定します。精密なはさみや鉗子を使用して脛脛知の前部の全長を露出させるために足から膝に足の皮膚を取り除く。はさみを使用して脛目と脛岩前部の間に切開を行います。鉗子を使用して、筋肉の表面から上層を慎重に除去します。足首では、鉗子を使用して脛表リス前腱を他の腱から分離し、腱をそっと引き上げて脛体および周囲の筋肉から分離する。脛表症前腹が完全に分離した場合、脛側前筋の近位部分のみが膝に付着するように腱を保持する。そして、膝蓋骨にできるだけ近い近位腱を穏やかに切断します。乾燥を避けるために、生理液で軽く湿らせたガーゼの上にチバイアリス前部を置きます。70~100ミリリットルのイソペンタンを含むビーカーを、ビーカーの底部のイポペンタンが白色固体になるまで液体窒素の容器に部分的に浸す。凍結後に生検を適切に識別できるようにコルクの反対側に慎重に事前ラベルを付け、約0.5ミリリットルのトラガカンスガムを20%11、8ミリメートルのコルクの円形部分に加えます。鉗子を使用して、歯茎遠位腱側に脛表を埋め込みます。コルク表面に垂直な軸で筋肉を保持します。脛表前部の腱側の少なくとも半分は、ガムによって発見されたままであるべきである。その後、すぐに完全な凍結を可能にするために、チルド、凍結されていないイポランタンに埋め込まれたコルクを約2分間浸します。凍結組織の切片を取得し、クライオスタット温度をマイナス25°Cに設定し、最適な切断温度化合物を使用して、組織をクライオスタット切断ブロックに固定します。筋肉の腹に達するまでサンプルをトリミングします。セクションがキャプチャされると、室温でスライドを配置ガラススライドごとに少なくとも2つのセクションを収集する7〜10マイクロメートルのセクションを得る。換気された環境で、組織を室温で少なくとも20分間乾燥させます。その後、セクションが染色されるまでマイナス80°Cで保管します。組織を免疫標識する。まず、換気環境で最低15分間室温でスライドを解凍する。次に、疎水性ペンでセクションを線分します。湿気の多いチャンバーで10分間2%パラホルムアルデヒドで組織を固定します。固定の終わりに、洗浄ごとに新鮮なPBSで2つの5分の洗浄でセクションをすすいだ。PBSで希釈した10%ヤギ血清で非特異的な結合をブロックします。室温で1時間標識した後、目的の一次抗体を用いて切片を、室温で2時間5%ヤギ血清に希釈した。インキュベーションの終了時に、PBSで3回の5分間の洗剤でスライドをすすいで、光から保護された室温で45分間適切なフルオロフォア共役二次抗体を標識します。インキュベーションの最後に、PBSで3回の10分間の洗浄でスライドを洗います。DAPIの1ミリリットル当たり100ナノグラムとカバースリップを含む蛍光実装媒体でセクションを取り付けます。その後、画像化する前に4°Cで一晩スライドを乾燥させます。4週齢のmdxマウスの脛眼筋は、しばしば同じ断面で一緒に退化および再生領域の影響を受けにくい領域を含む異種プロファイルを表示します。軽度の影響を受けない筋肉領域には、大きく均質な大きさのミオファイバーと核が含まれ、主に周囲またはミオファイバーの間に位置する低密度である。IgG陽性のミオファイバーは、一般的にこのような健康または軽度の影響を受けた領域には存在しないが、筋線維内のIgG免疫反応性は、ミオネクロシスを示す。新たに形成された筋繊維は、筋肉前駆物質が融合して同期性細胞に寄与するにつれて、再生の初期段階で徐々に拡大し、小さなサイズで存在する。このプロセスの間、ミオ核は、最近再生された筋繊維のクラスターを有する再生組織の中心にとどまり、人工的な人工繊維のリスクを最小限に抑えるために、組織を凍結するとき、正しい温度で筋肉をまっすぐに保つことが重要である。イセプタンとPFAは常に慎重に、煙のフードの下で処理する必要があります。
